摘要
目的优化黄芩苷磷脂复合物制备工艺。方法采用溶剂法制备黄芩苷磷脂复合物,黄芩苷含量测定采用高效液相色谱法,DIKMA C18色谱柱(250mm×4.6mm,5μm),流动相为甲醇-0.2%磷酸水溶液(53:47),柱温30℃,流速0.8mL/min,检测波长280nm,进样量10μL;以复合率作为评价指标,采用单因素试验依次考察投料比、药物浓度、反应温度和反应时间;设计四因素三水平正交试验,以复合率与复合物中黄芩苷质量分数为指标进行综合评分,优选出黄芩苷磷脂复合物制备工艺。结果确定最优工艺条件为投料比1:3,黄芩苷浓度2.5mg/mL,反应温度50℃,反应时间为1h。结论该制备工艺稳定可行。
Objective To optimize the preparation process of baicalin phospholipid complex.Methods The content of baicalin was determined by high performance liquid chromatography(HPLC)with DIKMA C18 column(250 mm×4.6 mm,5 um).The mobile phase was methanol-0.2%phosphoric acid solution(5347).The column temperature was 30℃and the flow rate was 0.8 mL/min.The detection wavelength was 280 nm and the injection volume was lOuL.With compound rate as the evaluation index,single factor test was used to investigate the feed ratio,drug concentration,reaction temperature and reaction time.Four factors and three levels orthogonal test were designed to optimize the preparation process of baicalin phospholipid complex.Results The optimal process conditions were determined as follows;1:3 weight ratio,2.5 mg/mL baicalin concentration,50℃reaction temperature,1 h reaction time.Conclusion The preparation process is stable and feasible.
