摘要
目的探讨亲代同型半胱氨酸(Hcy)水平升高对子代脑组织及骨骼肌端粒长度的影响。方法将8只小鼠随机分为实验组和对照组,每组包括3只雌性和1只雄性,将雌雄小鼠分开饲养,实验组喂以含1%蛋氨酸饲料,对照组喂以普通饲料,4周后合笼直至均生下子代小鼠,统计子代数量,留取亲代血液0.5 mL,酶联免疫吸附测定法测量血浆Hcy物质的量浓度,从2组里每只雌鼠子代中各随机选取3只,构成新的实验组(子代)和对照组(子代),每组9只,同时留取亲代及子代脑组织和后肢骨骼肌组织各10 mg,实时荧光定量PCR法测端粒长度。结果与对照组比较,亲代血浆Hcy水平显著升高,子代数量显著降低(P<0.05),但脑组织和骨骼肌组织中端粒长度差异无统计学意义(P>0.05)。子代脑组织端粒长度与对照组比较显著缩短(P<0.05),骨骼肌组织与脑组织端粒长度呈正相关关系(r=0.727,P=0.041)。结论亲代血浆Hcy水平升高可通过影响子代端粒长度,进而对子代的生长发育造成潜在的不良影响。
Objective To explore the effects of elevated homocysteine levels levels in parental generation on the telomere length of offspring brain and skeletal muscles.Methods 8 mice were randomly divided into experimental group and control group,each group consisting of 3 females and 1 male.The experimental group was fed with 1%methionine and the control group with ordinary feed.4 weeks later,female and male mice were mated respectively until the birth of their progeny,whose number was recorded.0.5 mL blood of the parental generation was retained,and the plasma Hcy concentration was measured by the enzyme-linked immunosorbent assay method.From each of the two groups,3 mice were randomly selected from each female,forming a new experimental group(offspring)and a control group(offspring)with 9 rats in each.At the same time,10 mg tissue from the parent and offspring’s brain and the skeletal muscle of the hind limbs was taken and the real-time fluorescence quantitative polymerase chain reaction method was usedto determine the length of the telomere.Results Compared with the control group,the level of plasma Hcy in the parents increased significantly while the number of progenies decreased(P<0.05),but there was no significant difference in the telomere length of either the brain tissue or the skeletal muscle(P>0.05).The telomere length of the offspring brain tissue was significantly shorter than that of the control group(P<0.05),and the skeletal muscle tissue was positively correlated with the telomere length of the brain tissue(r=0.727,P=0.041).Conclusion The increase of Hcy level in parental plasma may have potential an adverse effect on the growth offspring through affecting the telomere length.
