摘要
本研究在采集黄渤海鱼类生物凭证标本及相应DNA条形码的基础上,结合GenBank序列,采用BLAST分析进行数据校正分析、构建系统树检验和Barcoding Gap校验防错等方法核验所采用条形码的可靠性,通过统一提交数据的规范格式,实现物种、凭证标本和DNA条形码的相互关联,构建了黄、渤海鱼类DNA条形码信息平台(http://www.fishdna.com.cn/)。该平台数据信息由物种文献数据库、标本数据库和DNA条形码数据库组成,涵盖300余种鱼类的基本生物学信息和DNA条形码资源。平台能提供快捷直观的互联网查询,通过DNA条形码实现未知鱼类样本的物种查询和鉴定。本研究首次构建针对黄、渤海地区的重要鱼类生物的DNA条形码信息平台,访问该网站平台能认知未知鱼类样本,促进国内各海域、各地区乃至国外数据共享和合作交流,为我国北方主要经济鱼类生物分类鉴定、种质资源的开发利用、濒危海洋鱼类的保护和经济鱼类物种甄别提供重要数据资源。
The DNA barcode information platform of fish in Yellow Sea and Bohai Sea was constructed(http://www.fishdna.com.cn/)by BLAST analysis for data correction and analysis,construction of system tree test,barcoding gap verification and error proofing to verify the reliability of the barcodes used,and to realizes the correlation between species,voucher samples and DNA barcoding by unifying the standard format of data submission based on the collection of biological voucher samples and corresponding DNA barcodes of fishes in Yellow Sea and Bohai Sea,combined with GenBank sequences.The data information of the platform was composed of species literature database,specimen database and DNA barcoding database,covering the basic biological information and DNA barcoding resources of more than 300 species of fish.The platform can provide fast and intuitive internet query,and realize species identification of unknown fish samples through DNA barcoding,which was the first to build a DNA barcoding information platform for important fish organisms in the Yellow Sea and Bohai Sea,realizing the cognition of unknown fish samples through the access to the website platform,promoting the data sharing and cooperation and exchange among domestic waters,regions and even foreign countries.The finding provided important data resources for the biological classification and identification of major economic fish in northern China,the development and utilization of germplasm resources,the protection of endangered marine fish and the identification of economically important fish species.
