摘要
目的探讨接绪二天汤对桥本甲状腺炎小鼠卵巢储备功能及生育能力的影响。方法将60只雌鼠随机等分为对照组、模型组、中药干预组,各组再随机等分为2个亚组(取材组、合笼组)。除对照组外,其余2组予以完全弗氏佐剂抗原2周+不完全弗氏佐剂抗原9周皮下注射配合高碘水喂养造模,造模第9周开始中药干预组给予接绪二天汤灌胃,对照组与模型组给予等量生理盐水灌胃。造模15周时取材组于动情前期处死小鼠并取材,合笼组以雌雄2∶1合笼观察产仔数及子代存活数。结果模型组小鼠甲状腺及卵巢病理切片符合桥本甲状腺炎及其卵巢损害。经中药干预后病理结果好转,血清TgAb、TPOAb、E2、P、AMH、各级卵泡数(除成熟卵泡数)、产仔总数及子代存活数均有统计学意义(P<0.05或P<0.01)。结论接绪二天汤可以增加小鼠各级卵泡数量(除闭锁卵泡数),提高卵巢储备功能及生育能力。
Objective To investigate the effects of Jie Xu Er Tian Tang on ovarian reserve function and fertility in mice with Hashimoto’s thyroiditis.Methods 60 female rats were randomly divided into control group,model group and Chinese medicine intervention group and then randomly divided into 2 subgroups(sampling group and cage group).Except for the control group,the other two groups were given complete Fredrin’s adjuvant antigen for 2 weeks+incomplete Fredrin’s adjuvant antigen for 9weeks combined with high-iodine water feeding for modeling.From the 9th week of modeling,the Chinese medicine intervention group was given Jie Xu Er Tian Tang for insertion and the control group and the model group were given the same amount of normal saline for intragastric administration.At 15 weeks of modeling,the mice in the control group were sacrificed in the preestrous period and the litter size and offspring survival were observed in the co-cage group with a ratio of male to female.Results The model group showed the pathological sections of thyroid and ovary were consistent with Hashimoto’s thyroiditis and ovarian damage.Serum Tg Ab,TPOAb,E2,P,AMH,the number of follicles at all levels(except the number of mature follicles)and the total number of offspring born and the number of surviving offspring were statistically significant(P<0.05 or P<0.01)after TCM intervention.Conclusion Jie Xu Er Tian Tang increase the number of follicles at all levels in mice(except the number of atresia follicles)and improve the ovarian reserve function and fertility.
