摘要
目的探讨四妙丸通过调控Interleukin(IL)-25-IL-17RA/IL-17RB轴对咪喹莫特诱导的银屑病样小鼠模型的保护作用及作用机制。方法将30只BALB/c雄性小鼠随机分为正常组、模型组、阳性药组(甲氨蝶呤3 mg·kg^(-1))、四妙丸高、低剂量组(4 g·kg^(-1)、2 g·kg^(-1)),每组6只。在等面积(2 cm×3 cm)小鼠背部皮肤上除去被毛,除正常组外其余各组小鼠涂抹咪喹莫特乳膏(62.5 mg/只);各组灌胃给予相应溶媒或受试药物,连续干预5天。期间观察并记录各组小鼠背部PASI评分。在第5天收集血清及小鼠皮肤样品,剖取脾脏计算脏器系数,HE染色观察表皮厚度,ELISA法检测各组小鼠皮损组织及血清中IL-17A、IL-25、IL-23和IL-36水平,IHC法检测各组皮肤中IL-25、IL-17RA、IL-17RB的表达水平。结果PASI评分及HE染色结果显示,给予四妙丸后,可显著改善小鼠背部皮肤发红、增厚、皮屑增多等银屑病特征性临床表现,小鼠背部皮肤结痂及表皮增厚现象明显减轻;在皮肤组织及血清样本中,模型组IL-17A、IL-25、IL-23及IL-36水平较对照组明显上升(P<0.001),各治疗组IL-17A、IL-25、IL-23及IL-36水平较模型组均明显降低,四妙丸高剂量组IL-25和IL-36水平均显著降低(P<0.001);模型组小鼠皮损组织中IL-25、IL-17RA及IL-17RB表达水平显著高于对照组,各治疗组IL-25、IL-17RA及IL-17RB表达水平较模型组均明显降低。结论四妙丸可调节IL-25-IL-17RA/IL-17RB,从而降低银屑病小鼠血清及皮肤组织中IL-17A、IL-25、IL-23和IL-36的水平。
Objective To investigate the protective effect and mechanism of Simiao Pill on imiquimod induced psoriasis like mouse model by regulating Interleukin(IL)-25-IL-17RA/IL-17RB.Methods Thirty BALB/C male mice were randomly divided into control group,model group,positive drug group(methotrexate 3 mg·kg^(-1))and Simiao pill high and low dose groups(4 and 2 g·kg^(-1)),with 6 mice in each group.In equal area(2 cm×3 cm)remove the hair from the back skin of mice,and apply imiquimod cream(62.5 mg/each)to other groups except the control group;Each group was given corresponding solvent or test drug by gavage for 5 days.The PASI scores on the back of mice in each group were observed and recorded.On the 5th day,the serum and mouse skin samples were collected.The spleen was dissected to calculate the organ coefficient.The epidermal thickness was observed by H&E staining.The levels of IL-17A,IL-25,IL-23 and IL-36 in the skin lesions and serum of mice in each group were detected by ELISA.The expression levels of IL-25,IL-17RA and IL-17RB in the skin of each group were detected by IHC method.Results PASI score and he staining showed that after administration of Simiao pill,it could significantly improve the characteristic clinical manifestations of psoriasis such as redness,thickening and increased dandruff of mouse back skin,and significantly reduce the phenomenon of scab and epidermal thickening of mouse back skin;In skin tissue and serum samples,IL-17A,IL-25,IL-23 and IL-36 in the model group.The level of IL-17A,IL-25,IL-23 and IL-36 in each treatment group was significantly higher than that in the control group(P<0.001).The levels of IL-25 and IL-36 in Simiao pill high-dose group were significantly lower than those in model group.The levels decreased significantly(P<0.001);The expression levels of IL-25,IL-17RA and IL-17RB in the skin lesions of mice in the model group were significantly higher than those in the control group.The expression levels of IL-25,IL-17RA and IL-17RB in each treatment group were significantly lower than those in the model group.Conclusion Simiao pill can regulate IL-25-IL-17RA/IL-17RB and reduce IL-17A,IL-25,IL-23 and IL-36 in serum and skin tissue of psoriasis mice.
