摘要 (目的)建立高效液相色谱法(HPLC)测定可溶性微针中草乌甲素含量的方法。(方法)采用HPLC法,选择Ecosil 120-5-C18 EPS(4.6 mm×250 mm,5μm)色谱柱,以0.2%三乙胺水溶液(pH=3.1±0.1)-乙腈(60∶40)为流动相,检测波长为260 nm,流速为1.0 mL·min-1,柱温为30℃,进样量为20μL。(结果)方法专属性好,草乌甲素在2.002~200.200μg/mL浓度范围内与峰面积呈良好的线性关系(Y=32.909X-34.539,R2=0.9997);平均加样回收率99.48%,RSD值为0.48%(n=9);日内精密度和日间精密度的平均RSD分别为0.11%和0.49%,重复性和供试品24 h稳定性均符合要求。(结论)HPLC法测定草乌甲素含量的方法简单、准确,可用于草乌甲素可溶性微针中的含量测定,为后续的质量评价奠定基础。 Objective:To establish a high performance liquid chromatography(HPLC)method to determine the content of aconitine in soluble microneedles.Method:Using HPLC method,selecting Ecosil120-5-C18 EPS(4.6 mm×250 mm,5μm)chromatographic column,using 0.2%triethylamine aqueous solution(pH=3.1±0.1)-acetonitrile(60∶40).Is the mobile phase,the detection wavelength is 260 nm,the flow rate is 1.0 mL·min-1,the column temperature is 30℃,and the injection volume is 20μL.Result:This method has good specificity,and the concentration range of aconitine A has a good linear relationship with the peak area in the concentration range of 2.00~200μg/mL(Y=32.909 X-34.539,R2=0.997)The average recovery rate of sample addition is 99.48%,and the RSD value is 0.48%(n=9);the average RSD of intra-day precision and inter-day precision are 0.11%and 0.49%,respectively,repeatability and The stability of the test products for 24 hours all met the requirements.Conclusion:The HPLC method is simple and accurate to determine the content of Aconitine A.It can be used to determine the content of Aconitine A in soluble microneedles,which lays the foundation for subsequent quality evaluation.
