摘要
中药因其多成分多靶点的协同作用使其药理机制研究复杂而又困难,当前的单一靶点、单一成分的药物研发思路不能满足中药的研发需求。随着生物信息学和系统生物学的不断进展,网络药理学以其整体性的思维方式和强大的预测能力在中药研究中被广泛应用。网络药理学通过利用相关数据库和算法进行数据挖掘,使用可视化软件建立"药物-靶标-疾病"网络,通过分析,可以验证中药活性成分,对各组分间的相互作用进行分析,预测药物作用靶点,发现新的适应证并减少毒副反应。综述网络药理学的研究思路及当前的应用现状,以期为中药现代化和新药研发提供理论依据。
Due to multi-component and multi-target synergy effect,it is complicated and difficult to study pharmacological mechanisms of TCM.Current"single-target,single-component"drug development methods cannot meet the needs of TCM research.Based on bioinformatics and systems biology,network pharmacology is widely used in TCM research due to its holistic thinking mode and strong prediction ability.Using related databases and algorithms for data mining,and visualization software to establish a"drug-target-disease"network,network pharmacology can be used to verify the active ingredients of TCM,analyze the interactions between the components,predict the drug targets and new indications,and reduce of toxic or side effects,which provides a theoretical basis for the modernization and drug discovery of TCM.
