摘要
目的研究苦荞黄酮(TBF)对Ⅱ型糖尿病小鼠血糖水平的调控及其可能的机制。方法采用高糖高脂饮食联合链脲佐菌素(STZ)处理的方法复制Ⅱ型糖尿病小鼠模型,连续4周以苦荞黄酮灌胃,检测模型小鼠空腹血糖(FBG)和肝脏过氧化物酶体增殖剂激活受体γ(PPARγ)、磷脂酰肌醇-3-激酶(PI3K)、蛋白激酶B(Akt)蛋白表达及Akt(Ser 473)磷酸化水平的变化。结果与对照组比较,TBF降低Ⅱ型糖尿病小鼠空腹血糖的效果差异有显著性(P<0.05),能显著提高PPARγ和PI3K的蛋白表达(P<0.01;P<0.05)以及Akt(Ser 473)的磷酸化水平(P<0.01)。结论TBF可能通过提高肝脏PPARγ,PI3K蛋白的表达及Akt磷酸化水平,改善Ⅱ型糖尿病。
Objective To study the effects of tatary buckwheat flavonoid(TBF)on expression and phosphorylation of PPARγ-PI3K/Akt pathway in T2DM mice.Methods The models of T2DM mice were produced by intraperitoneal injection of streptozotocin and feeding with high glucose and high fat diet.Mice were administered with TBF once a day for consecutive 4 weeks.The fasting blood-glucose was measured once a week.Western-blot was used to assess the expression of peroxisome proliferators-activated receptorγ(PPARγ),phosphatidylinositol 3-hydroxy kinase(PI3K),protein kinase B(Akt)and phosphorylated Akt in liver.Results Compared to model group,in TBF treated mice,the reduction of fasting blood-glucose was associated with remarkable increase of PPARγ,PI3K and phosphorylated Akt.Conclusion TBF alleviates the insulin resistance in T2DM mice by increasing the expression of PPARγ-PI3K/Akt related proteins.
