摘要
目的:采用四极杆-静电场轨道阱高分辨质谱联用(HPLC-Q-Exactive-MS)技术建立蒙药蓝盆花含药血清色谱图并分析其入血成分。方法:采用ACE EXCEL 3 C18-PFP(100 mm×3 mm,3μm)色谱柱;以乙腈-0.1%甲酸溶液为流动相,梯度洗脱;柱温为45℃;流速为0.5 mL/min;热电喷雾(HESI)质谱离子源,负离子全扫描模式下采集数据,建立15批蓝盆花含药血清和空白血清色谱图;采用SIMCA-P+14.0软件进行模式识别及多维统计分析。结果:建立了蒙药蓝盆花血清图谱,共确定17个成分,聚类分析结果表明15批蓝盆花样品分为3类,与主成分分析结果一致;正交偏最小二乘判别分析法筛选出绿原酸、新绿原酸及芹菜素3种差异较大药源性成分。结论:本研究首次采用HPLC-Q-Exactive-MS技术建立蒙药蓝盆花血清图谱,该方法可较全面地反映蓝盆花口服给药后吸收入血情况,为其体内药效物质研究及质量控制奠定基础。
Objective:To establish the serum chromatogram of Mongolian medicine Flos Scabiosae and to analyze its components in blood by using HPLC-Q-Exactive-MS.Methods:ACE EXCEL 3 C18-PFP(100 mm×3 mm,3 μm)chromatographic column was used,the mobile phase was acetonitrile-0.1% formic acid solution with gradient elution,the column temperature was 45 ℃,the flow rate was 0.5 mL/min.Thermal Spray(HESI)mass spectrometry ion source was used to collect data with negative ion full scan mode.15 batches of Flos Scabiosae drug-containing serum and blank chromatogram of serum were establish,SIMCA-P+14.0 software was used for pattern recognition and multidimensional statistical analysis.Results:The serum chromagraphy of Flos Scabiosae was established,17 components were ensured.The results of cluster analysis showed that 15 batches of Flos Scabiosae samples were divided into 3 categories,which were consistent with the results of principal component analysis.The orthogonal partial least squares discriminant analysis was used to screen three different pharmaceutical ingredients,including chlorogenic acid,neochlorogenic acid and apigenin.Conclusion:In this study,HPLC-Q-Exactive-MS technique is used for the first time to establish the serum chromatographic of Mongolian medicine Flos Scabiosae,which can reflect the absorption of Flos Scabiosae into the blood after oral administration comprehensively and lay a foundation for the study of its pharmacodynamic substances in vivo and quality control.
