摘要
目的:探究益气补血中药复方对原发性卵巢功能不全(POI)小鼠卵巢TGF-β超家族信号通路的影响。方法:取30只SPF级雌性BABL/c小鼠采用腹腔注射VCD溶液建立POI小鼠模型,并随机分为模型组及益气补血中药复方低、高剂量组,另取10只不予任何处理作为正常对照组。益气补血中药复方低、高剂量组分别按1.0、2.0 g/kg灌胃益气补血中药复方,正常对照组和模型组按10 mL/kg灌胃蒸馏水,造模结束后采集小鼠阴道脱落细胞,光镜下观察小鼠动情周期进行模型评价。给药结束后当天,观察动情周期变化;检测小鼠血清E;、LH、FSH、AMH水平;比较小鼠体质量、卵巢质量和子宫质量;观察小鼠卵巢组织病理学变化;检测小鼠颗粒细胞凋亡情况;Western blotting检测TGF-β超家族通路蛋白表达情况。结果:与正常对照组比较,模型组小鼠的动情周期紊乱率和血清LH、FSH水平及卵巢颗粒细胞凋亡率显著升高,血清E;、AMH水平及子宫质量、卵巢质量和卵巢组织中TGF-β、GDF-9、BMP15蛋白表达显著降低(P<0.05)。与模型组比较,益气补血中药复方低、高剂量组小鼠的动情周期紊乱率及血清LH、FSH水平和卵巢颗粒细胞凋亡率显著降低,血清E;、AMH水平及子宫质量、卵巢质量和卵巢组织中TGF-β、GDF-9、BMP15蛋白表达显著升高(P<0.05)。结论:益气补血中药复方能改善POI小鼠动情周期和血清激素水平,降低卵巢颗粒细胞凋亡率,其可能通过TGF-β超家族信号通路发挥对POI小鼠的治疗作用。
Objective To explore the effect of the Chinese herbal compound prescription of Yiqi buxue on the ovarian TGF-βsuperfamily signaling pathway of primary ovarian insufficiency(POI)mice.Methods:The POI mice model was established by intraperitoneal injection of VCD solution in 30 SPF BABL/c female mice,the mice were randomly divided into model group,low-dose(1.0 g/kg)and high-dose(2.0 g/kg)Chinese herbal compound prescription of Yiqi buxue groups,and 10 mice without any treatment were taken as normal control group.Normal control group and model group were given distilled water by intragastric administration at 10 mL/kg,exfoliative cells of mice vagina were collected after modeling,and the estrous cycle of mice was observed under light microscope for model evaluation.The estrus cycle was observed on the day after the end of administration;The levels of E;,LH,FSH and AMH in serum were detected;The body mass,ovary mass and uterine mass of mice were compared;The morphological changes of ovarian tissue were observed;The apoptosis of mice granulosa cells was detected and the protein expression of TGF-βsuperfamily pathway was detected by Western blotting.Results:Compared with the normal control group,the estrus cycle disturbance rate,serum LH,FSH levels and ovarian granulosa cell apoptosis rate of mice in model group were significantly increased,serum E;and AMH levels,uterine mass,ovarian mass and protein expressions of TGF-β,GDF-9 and BMP15 in ovarian tissue were significantly decreased(P<0.05).Compared with model group,the estrus cycle disturbance rate,serum LH and FSH levels,ovarian granulosa cell apoptosis rate of mice in low-dose and high-dose Chinese herbal compound prescription of Yiqi buxue groups were significantly decreased,serum E;and AMH levels,uterine mass,ovarian mass and protein expressions of TGF-β,GDF-9 and BMP15 in ovarian tissue were significantly increased(P<0.05).Conclusion:The Chinese medicine compound of Yiqi buxue can improve the estrous cycle and serum hormone levels in POI mice,reduce the apoptosis rate of ovarian granulosa cells.It may be through the TGF-βsuperfamily signaling pathway to exert a therapeutic effect on POI mice.
