摘要
目的:采用气相色谱指纹图谱比较不同基原化橘红(毛橘红和光橘红)的挥发性成分差异,并用化学计量学法对不同基原的化橘红进行区分。方法:化橘红样品经乙酸乙酯超声提取后,采用DB-WAXETR(30 m×0.32 mm×0.25μm)毛细管柱程序升温的模式进行分离,以FID检测器检测,采用相似度评价、主成分分析、聚类分析和正交偏最小二乘判别分析等化学计量学方法对毛橘红和光橘红进行质量评价。结果:建立了不同基原化橘红的气相色谱指纹图谱,毛橘红和光橘红分别含有34、14个共有峰,共指认出9个成分。主成分分析、聚类分析和正交偏最小二乘判别分析可将28批样品分成毛橘红和光橘红2类。结论:该方法能很好地体现毛橘红和光橘红挥发性成分的整体性和差异性,可为岭南道地中药材化橘红的质量控制及等级标准的制订提供依据。
Objective:To compare the volatile components of Citri Grandis Exocarpium from different origins,including Citrus grandis′Tomentosa′and Citrus grandis(L.)Osbeck,by gas chromatographic fingerprint,and to distinguish Citri Grandis Exocarpium from different origins by stoichiometry.Methods:The Citri Grandis Exocarpium samples were ultrasonically extracted by ethyl acetate,separated on DB-WAXETR(30 m×0.32 mm×0.25μm)capillary column by temperature programmed mode,and detected by FID detector.Similarity evaluation,principal component analysis,cluster analysis and orthogonal partial least square discriminant analysis were used to evaluate the quality of pericarp of Citrus grandis′Tomentosa′and Citrus grandis(L.)Osbeck.Results:The GC fingerprints of Citri Grandis Exocarpium from different origins were established.There were 34 and 14 common peaks in the chromatograms of pericarp of Citrus grandis′Tomentosa′and Citrus grandis(L.)Osbeck,respectively.Nine components were identified.Principal component analysis,cluster analysis and orthogonal partial least squares discriminant analysis could divide 28 batches of samples into two categories,the pericarp of Citrus grandis′Tomentosa′and Citrus grandis(L.)Osbeck.Conclusion:This method can well reflect the integrity and difference of volatile components of the pericarp of Citrus grandis′Tomentosa′and Citrus grandis(L.)Osbeck,and can provide a basis for the quality control and grade standard formulation of the protected Lingnan authentic Chinese herbal medicine Citri Grandis Exocarpium.
