摘要
目的:基于UPLC-MS/MS法同时测定栀子中14个萜类成分的含量。方法:以70%乙醇为提取溶剂,采用ACQUITY UPLC^(TM)BEH C_(18)(100 mm×2.1 mm,1.8μm)色谱柱;以含0.1%甲酸的乙腈-0.1%甲酸溶液为流动相,梯度洗脱;流速为0.6 mL/min;柱温为40℃;进样量为1.0μL。14个萜类成分中去乙酰车叶草苷酸甲酯、山栀苷、京尼平、京尼平-1-O-β-D-龙胆双糖苷、山栀子苷B、京尼平苷酸、栀子苷、鸡矢藤次苷甲酯、栀子新苷、西红花苷Ⅰ、西红花苷Ⅲ、藏红花酸采用负离子检测模式,山栀苷甲酯和西红花苷Ⅱ采用正离子检测模式。结果:栀子中上述14个萜类成分的方法学考察结果良好。其中,栀子苷、去乙酰车叶草苷酸甲酯和京尼平-1-O-β-D-龙胆双糖苷在栀子中含量较高,不同产地的栀子中14个萜类成分含量表现出一定差异。结论:该方法准确,重复性好,检测限低,能够客观反映不同产地栀子药材的主要萜类成分含量差异。
Objective:To establish an UPLC-MS/MS method for the simultaneous determination of the contents of 14 terpenoids in Gardeniae Fructus.Methods:The 70%ethanol was used as extraction solvent.The ACQUITY UPLC^(TM)BEH C_(18)(100 mm×2.1 mm,1.8μm)column was used,mobile phase was acetonitrile containing 0.1%formic acid-0.1%formic acid with gradient elution,flow rate was 0.6 mL/min,column temperature was 40℃,injection volume was 1.0μL.Among the 14 terpenoids,6-α-hydroxygeniposide,shanziside,genipin,genipin-1-O-β-D-gentiobioside,shanziside B,geniposidic acid,geniposide,scandioside methyl ester,gardoside,crocinⅠ,crocinⅢ,crocetin were detected under negative ion detection mode,and shanziside methyl ester and crocinⅡwere detected under positive ion detection mode.Results:The methodological investigation results of 14 terpenoids were good.Geniposide,6-α-hydroxygeniposide and genipin-1-O-β-D-gentiobioside had the higher contents in Gardeniae Fructus.The contents of 14 terpenoids in Gardeniae Fructus from different habitats showed certain differences.Conclusion:The method is accurate,reproducible and has low detection limit,which can objectively reflect the differences of main terpenoids in Gardeniae Fructus from different habitats.
