摘要
目的:建立一种简单、准确、快速地鉴定马钱子药材及其易混品的方法,为马钱子基原真实性鉴定和质量控制提供实验依据。方法:利用性状和微性状鉴定方法分析比较马钱子及其易混品的主要区别特征。采用高效液相色谱法(HPLC)检测25批马钱子类样品中士的宁和马钱子碱的含量。建立HPLC指纹图谱及相似度评价,同时以共有峰面积作为原变量进行系统聚类分析。结果:马钱子及其易混品在性状和微性状方面主要表现在种子形态特征及子叶叶脉的数量。HPLC指纹图谱标定了10个共有峰并指认其中2个峰,样品间相似度在0.446~0.995之间,聚类分析可将样品分为3类。结论:马钱子及其易混品之间存在较大差异。山马钱子、牛眼马钱子中士的宁和马钱子碱含量均不符合2020年版中国药典的相关规定,但云南马钱子中士的宁和马钱子碱含量可满足该标准,可作为马钱子潜在药用资源进一步研究。
Objective:To establish a simple,accurate and rapid method for identification of Strychnos nux-vomica and confusion varieties,so as to provide experimental basis for authenticity identification and quality control of Strychnos nux-vomica.Methods:The main distinguishing characteristics of Strychnos nux-vomica and confusion varieties were analyzed and compared by the methods of character identification and micro character identification.HPLC method was used to detect the contents of strychnine and brucine in 25 batches of samples.HPLC fingerprints and similarity evaluation were established,and the common peak area was used as the original variable for systematic cluster analysis.Results:The morphological characteristics of seeds and the number of veins of cotyledon of Strychnos nuxvomica and confusion varieties were the main characters and micro characters.Ten common peaks were demarcated by HPLC fingerprint and two of them were identified.The similarity of samples was between 0.446 and 0.995.The samples could be divided into three categories by cluster analysis.Conclusion:There are great differences between Strychnos nux-vomica and confusion varieties.The contents of strychnine and brucine in Strychnos nux-blanda and Strychnos angustiflora do not conform to the relevant provisions of the Chinese pharmacopoeia in 2020 edition,but the contents in Strychnos pierriana can meet the standard.It can be further studied as potential medicinal resources of Strychnos nux-vomica.
