摘要
目的:优化银丹活血通瘀胶囊的提取工艺。方法:以毛蕊异黄酮葡萄糖苷、丹酚酸B、迷迭香酸、人参皂苷Rg1、人参皂苷Rb1、三七皂苷R1的含量及出膏率为评价指标,采用层次分析-熵权法-独立性权法确定各指标的权重并计算综合评分。以加水量、提取时间、提取次数为考察因素,通过正交试验设计结合反向传播(BP)神经网络优选银丹活血通瘀胶囊的提取工艺并进行验证。结果:毛蕊异黄酮葡萄糖苷、迷迭香酸、三七皂苷R1、人参皂苷Rg1、丹酚酸B、人参皂苷Rb1的含量及出膏率的权重分别为0.435 4、0.091 9、0.044 1、0.069 0、0.029 9、0.294 7、0.035 1。BP神经网络预测的最佳工艺综合评分为97.71(RSD为0.47%),正交试验所优选工艺的综合评分为94.24(RSD为0.72%)。结论:最终优选出银丹活血通瘀胶囊的提取工艺为加6倍量水,提取2次,每次0.5 h,且该工艺稳定可行。
Objective:To optimize the extraction process of Yindan Huoxue Tongyu Capsules.Methods:With the content of calycosin-7-glucoside,rosmarinic acid,notoginsenoside R1,ginsenoside Rg1,salvianolic acid B,and ginsenoside Rb1 and the extraction yield of Yindan Huoxue Tongyu Capsules as indicators,analytic hierarchy process-entropy weight method-independent weight method was adopted to determine the weight coefficient of each indicator and calculate the comprehensive score.The orthogonal design and BP neural network were employed to optimize the extraction process(extraction time,water amount,and extraction times)of Yindan Huoxue Tongyu Capsules.Results:The weight coefficients for the content of calycosin-7-glucoside,rosmarinic acid,notoginsenoside R1,ginsenoside Rg1,salvianolic acid B,and ginsenoside Rb1 and the extraction yield were 0.4354,0.0919,0.0441,0.0690,0.0299,0.2947,and 0.0351,respectively.The comprehensive score of the optimal process predicted by BP neural network was 97.71(RSD=0.47%),and that of the optimal process obtained by the orthogonal design was 94.24(RSD=0.72%).Conclusion:The optimal extraction process of Yindan Huoxue Tongyu Capsules was extraction with 6 volumes of water twice(each time of 0.5 h).The optimized extraction process was stable and feasible.
