Winter bamboo shoots are widely used in Asian cuisine. We surveyed growth depth (tail depth and root depth), harvest speed, and harvest processes to provide information for developing new harvest technology. The tail depth of most winter bamboo shoots was 100 350 mm below the soil surface, and the root depth was 200 500 mm below the soil surface. Most winter bamboo shoots were difficult to locate with only the naked eye. The digging depth was 200 500 mm to cut winter bamboo shoots from the root. The highest harvest rate was 7.75 times faster than the slowest one and the average harvest speed ratio of men: women was about 1.71:1. The harvest process of winter bamboo shoots was divisible into three steps: searching, digging a hole (around the shoot) and cutting the root. The ratio of searching time to the sum of digging and cutting time was about 8:1, showing that searching required more time than digging and cutting together. 展开更多

Purpose:This study investigates key factors contributing to research impact and their interactions with the Research Impact Quintuple Helix Model by Arsalan et al.(2024).Design/methodology/approach:Using data from a global survey of 630 scientists across diverse disciplines,genders,regions,and experience levels,Structural Equation Modelling(SEM)was employed to assess the influence of 29 factors related to researcher characteristics,research attributes,publication strategies,institutional support,and national roles.Findings:The study validated the Quintuple Helix Model,uncovering complex interdependencies.Institutional support significantly affects research impact by covering leadership,resources,recognition,and funding.Researcher attributes,including academic experience and domain knowledge,also play a crucial role.National socioeconomic conditions indirectly influence research impact by supporting institutions,underscoring the importance of conducive national frameworks.Research limitations:While the study offers valuable insights,it has limitations.Although statistically sufficient,the response rate was below 10%,suggesting that the findings may not fully represent the entire global research community.The reliance on self-reported data may also introduce bias,as perceptions of impact can be subjective.Practical implications:The findings have a significant impact on researchers aiming to enhance their work’s societal,economic,and cultural significance,institutions seeking supportive environments,and policymakers interested in creating favourable national conditions for impactful research.The study advocates for a strategic alignment among national policies,institutional practices,and individual researcher efforts to maximise research impact and effectively address global challenges.Originality/value:By empirically validating the Research Impact Quintuple Helix Model,this study offers a holistic framework for understanding the synergy of factors that drive impactful research. 展开更多

鳓鱼全基因组survey分析及微卫星位点分布

为研究鳓鱼(Ilisha elongata)的遗传演化概况,对鳓鱼基因组进行了高通量测序并开展了survey分析和基因组范围内的遗传标记开发。K-mer分析结果显示,鳓鱼基因组大小约为692.8 Mb,杂合率为0.18%,重复序列比例为39.6%。基因组初步组装Scaff... 为研究鳓鱼(Ilisha elongata)的遗传演化概况,对鳓鱼基因组进行了高通量测序并开展了survey分析和基因组范围内的遗传标记开发。K-mer分析结果显示,鳓鱼基因组大小约为692.8 Mb,杂合率为0.18%,重复序列比例为39.6%。基因组初步组装Scaffold N50为27694 bp,Contig N50为6306 bp。利用MISA软件对鳓鱼基因组的微卫星标记(simple sequence repeat,SSR)进行检索和分析,总共检测到786123个SSR位点,相对丰度为1204个/Mb。在所有类型中,重复最多的是二碱基,占SSR总量的75.94%,其SSR重复频率主要集中在6~28次;其次为单碱基和四碱基,分别占SSR总量的15.31%和4.25%。对二碱基类型而言,AC型具有最多的重复数量,为131732个,单碱基重复数量最多的则是A型,有48775个。研究结果表明,鳓鱼基因组经组装后可得到高质量的全基因组序列,经过筛选的SSR位点可为后续的遗传分子标记开发提供有力支持,研究结果可为鳓鱼种质资源管理和保护、生物进化和群体遗传等研究工作提供基础资料。 展开更多

In order to survey the infectious situation of canine coronavirus (CCV) in giant panda population, a virus neutralization test detecting specific antibodies against CCV in giant panda抯 sera was established by using two-fold dilutions of serum and 100 TCID50 of the virus. The 62 sera samples of giant pandas, which were gathered from zoos and reserve region of Sichuan Province, China were detected. The neutralization antibody titer of 1:4 was recognized as the positive criterion, 8 sera samples were detected to be positive, and the positive rate was 12.9%. The titers of neutralizing antibody ranged from 1:8 to 1:32. It was the first comprehensive investigation on neutralization antibodies against CCV in giant panda population in China. The results of study showed that the infection of CCV in giant panda population was universal, which has posed a threat to the health of giant panda. Therefore, it is incumbent on us to study safe and effective vaccines to protect giant panda against CCV infection. 展开更多

Four future missions for deep space exploration and future space-based exoplanet surveys on habitable planets by 2030 are scheduled to be launched.Two Mars exploration missions are designed to investigate geological structure,the material on Martian surface,and retrieve returned samples.The asteroids and main belt comet exploration is expected to explore two objects within 10 years.The small-body mission will aim to land on the asteroid and get samples return to Earth.The basic physical characteristics of the two objects will be obtained through the mission.The exploration of Jupiter system will characterize the environment of Jupiter and the four largest Moons and understand the atmosphere of Jupiter.In addition,we further introduce two space-based exoplanet survey by 2030,Miyin Program and Closeby Habitable Exoplanet Survey(CHES Mission).Miyin program aims to detect habitable exoplanets using interferometry,while CHES mission expects to discover habitable exoplanets orbiting FGK stars within 10 pc through astrometry.The above-mentioned missions are positively to achieve breakthroughs in the field of planetary science. 展开更多

LAMOST-MRS-B is one of the sub-surveys of LAMOST medium-resolution(R~7500)spectroscopic survey.It aims at studying the statistical properties(e.g.,binary fraction,orbital period distribution,mass ratio distribution)of... LAMOST-MRS-B is one of the sub-surveys of LAMOST medium-resolution(R~7500)spectroscopic survey.It aims at studying the statistical properties(e.g.,binary fraction,orbital period distribution,mass ratio distribution)of binary stars and exotic stars.We intend to observe about 30000 stars(10 mag≤G<14.5 mag)with at least 10 visits in five years.We first planned to observe 25 plates around the galactic plane in 2018.Then the plates were reduced to 12 in 2019 because of the limitation of observation.At the same time,two new plates located at the high galactic latitude were added to explore binary properties influenced by the different environments.In this survey project,we set the identified exotic and low-metallicity stars with the highest observation priorities.For the rest of the selected stars,we gave the higher priority to the relatively brighter stars in order to obtain high quality spectra as many as possible.Spectra of49129 stars have been obtained in LAMOST-MRS-B field and released in DR8,of which 28828 and 3375 stars have been visited more than twice and ten times with SNR≥10,respectively.Most of the sources are B-,A-,and F-type stars with-0.6<[Fe/H]<0.4 dex.We also obtain 347 identified variable and exotic stars and about 250 stars with[Fe/H]<-1 dex.We measure radial velocities(RVs)by using 892233 spectra of the stars.The uncertainties of RV achieve about 1 km·s^(-1)and 10 km·s^(-1)for 95%of late-and early-type stars,respectively.The datasets presented in this paper are available at http://www.doi.org/10.57760/sciencedb.j00113.00035. 展开更多

禾本科沙鞭属仅包含沙鞭一个种。沙鞭具有根茎繁殖、克隆整合等典型沙生植物特征,是我国内蒙古高原非固定沙地的建群种。本研究采用流式细胞术和K-mer分析方法测定沙鞭基因组大小,建立和优化以芨芨草为内参物种、青海固沙草和方穗山羊... 禾本科沙鞭属仅包含沙鞭一个种。沙鞭具有根茎繁殖、克隆整合等典型沙生植物特征,是我国内蒙古高原非固定沙地的建群种。本研究采用流式细胞术和K-mer分析方法测定沙鞭基因组大小,建立和优化以芨芨草为内参物种、青海固沙草和方穗山羊草为外参物种的二倍体植物DNA含量(DNA C值)测定体系。研究结果表明:1)沙鞭流式细胞峰值荧光是青海固沙草的2。54倍,是芨芨草的1。35倍,峰值信号远小于方穗山羊草,推测沙鞭基因组大小约为1580。10±5。02 Mb;2)K-mer分析结果表明,沙鞭基因组大小为1563。54 Mb,杂合率1。15%,重复序列比例为66。27%,基因组GC含量为43。4%,属于高杂合、高重复的复杂基因组;3)沙鞭基因组可使用PacBio平台CLR或HiFi模式进行三代测序,测序深度应不低于20×。沙鞭基因组大小的准确测定不仅补充了禾本科针茅族植物的DNA含量数据,同时也为沙鞭基因组测序、进化基因组研究、种质资源开发和利用以及遗传资源保护提供了数据参考,可为针茅族近缘物种基因组大小测定提供借鉴。 展开更多

为了解疣吻沙蚕(Tylorrhynchus heterochaetus)基因组信息并高效地开发微卫星标记,指导其种质资源保护与新品种的遗传改良研究,采用低深度高通量测序开展全基因组survey,k-mer分析估计疣吻沙蚕基因组大小为759.53 Mb,杂合率1.41%,重复... 为了解疣吻沙蚕(Tylorrhynchus heterochaetus)基因组信息并高效地开发微卫星标记,指导其种质资源保护与新品种的遗传改良研究,采用低深度高通量测序开展全基因组survey,k-mer分析估计疣吻沙蚕基因组大小为759.53 Mb,杂合率1.41%,重复序列比例45.92%;初步组装获得2181621条scaffold,全长为840375821 bp。在基因组序列中检测到130216个微卫星位点,丰度为154.9个·Mb^(−1)。微卫星重复次数集中在4~18拷贝;单碱基重复比例最高(35.00%),二碱基(32.48%)、三碱基(14.42%)次之;二碱基、三碱基优势基序分别是AT/AT、AAT/ATT,表现出A/T碱基优势。从随机挑选的50对引物中筛选到15对多态标记,在30尾样本中共检测到87个等位基因,等位基因数(N_(a))为2.000~12.000(平均5.800),有效等位基因数(Ne)为1.164~6.713(平均3.328),期望杂合度(H_(e))为0.141~0.789(平均0.561),多态信息含量(PIC)为0.136~0.776(平均0.511);其中13个为高度或中度多态性位点,在遗传分析中有较高的实用价值。结果表明,疣吻沙蚕基因组为复杂基因组,其微卫星位点类型丰富且具备良好的多态性潜能,可为种质资源评价、群体遗传学及分子育种研究提供有效的标记资源。 展开更多

中国李是重要的落叶果树之一,具有较高的经济价值与生态价值,但其全基因组信息尚未被解析,限制了相关研究的深入开展。通过二代基因组Survey测序分析,对中国李品种"秋姬"全基因组信息进行初步解析,质控后共获得22.9 Gb高质量... 中国李是重要的落叶果树之一,具有较高的经济价值与生态价值,但其全基因组信息尚未被解析,限制了相关研究的深入开展。通过二代基因组Survey测序分析,对中国李品种"秋姬"全基因组信息进行初步解析,质控后共获得22.9 Gb高质量数据,经17-mer分析,预估其基因组大小为305.6 Mb,基因组杂合度为0.92%,属于高杂合中型基因组;采用SOAPdenovo软件进行组装,所得Contig N50为440 bp,Scaffold总长约为267 Mb。后续拟用第三代测序技术进行精细组装。 展开更多
