摘要
为制定盐爪爪全基因组测序以及染色体水平基因组组装策略方案,以二倍体盐生植物盐爪爪为研究对象,采用流式细胞术和基因组Survey对盐爪爪基因组大小以及杂合率等相关特征进行评估。流式细胞术结果表明,盐爪爪的基因组大小在0.93~1.01 Gb之间,通过基因组Survey测定盐爪爪基因组大小为987.17 Mb,杂合率为1.01%,重复序列比例为63.72%,表明盐爪爪基因组属于一般复杂基因组。采用K-mer=41进行初步组装,得到Contig N50为765 bp,总长为814715464 bp,Scaffold N50为1170 bp,总长为848776486 bp。
To formulate the whole genome sequencing and chromosome-level genome assembly strategy plan for Kalidium foliatum,taking the diploid halophyte Kalidium foliatum as the research object,flow cytometry and genome Survey were used to evaluate the genomic size and heterozygous rate and other related characteristics of Kalidium foliatum.The results of flow cytometry indicated that the genomic size of Kalidium foliatum was between 0.93 Gb and 1.01 Gb.Through genome Survey,the genomic size of Kalidium foliatum was determined to be 987.17 Mb,with a heterozygous rate of 1.01%and a proportion of repetitive sequences of 63.72%,suggesting that the genome of Kalidium foliatum belonged to a generally complex genome.Preliminary assembly was carried out using K-mer=41,resulting in Contig N50 of 765 bp with a total length of 814715464 bp,and Scaffold N50 of 1170 bp with a total length of 848776486 bp.
