In order to solve the problems which are widespread in potato seeding planter, such as lower operating efficiency, unideal performance index, higher replay rate and leakage rate and uneven spacing sowing in seeding operating, a trailed potato fertilization seeder was designed. The key components of the special structure had been got through the description of the structure and working principle of the whole machine. A crossing seed-taken technology along with the vibration component was adopted to achieve precision seeding. The results showed in the feld test of the machine: all the performance indexes of the machine met the agronomic requirement of potato planting; the multiple and missing index were low; it completed the ditching, fertilizing, seeding as well as ridging in one planting process; its operation effciency was high and the stability was good. This design of the trailed potato fertilization seeder provided a reference for the development of large traction-type potato seeding machine. 展开更多

Based on the sweet potato starch,cationic starch,acetic starch and cationic-acetic compoundedly modified starch were made through chemical denaturalization.The above three kinds of static rheological parameter and dynamic rheological parameter were measured,respectively.The experimental result reveals that the thermal stability of starchy viscosity increases after chemical denaturalization.Under the condition of identical shearing rate,the shear stress of cationic-acetic ester compoundedly modified sweet potato starch paste is the largest among these kinds of sweet potato starch.This attributes to a phenomenon of shearing thinning.Furthermore,raw sweet potato starch has a larger gel intensity than that of modified starch. 展开更多

DNA fingerprints of 37 PVY resistant potato varieties (clones) were generated using PCR-based RAPD analysis. All 37 varieties were differentiated by banding patterns obtained from 17 primers that generated 164 polymorphisms. Similarity matrices were calculated with the simple matching coefficient. The genetic distance value among them was between 0.27 and 0.50, with average value being 0.35. The average link clustering algorithm was used to construct a dendrogram with Statistica. They were classified into three groups according to clustering results, and the results presented some geographic relationship. Dendrogram showed close genetic relationships between a number of varieties (clones) of similar pedigree. This study has shown that RAPD marker is a useful tool for potato variety identification, differentiation, and estimation of genetic relationships. 展开更多

Concentration and gradient experiments for four components (enzyme, MgCl2, DNA template and dNTPs) in PCR system were used in the research with the concentration of the other component remained the same; the orthogonal design L9 (34) was applied in the optimization of four sets of primers (STM0014, Pat, SSI, and UGP) in the reaction system at three levels; the temperature gradient selection was used to find out the optimum annealing temperature for the primer. The optimized multiplex PCR system of potato SSR marker with a total volume of 20 μL : 2.5 μL 25 mmol.L-1 MgCl2, 0.6 μL 10 mmol·L-1 dNTPs, 0.8 U Taq, 80 ng DNA template was ultimately established through the comparison and analysis of test results; the ratio of four pairs of 4 mmol. L1 primers was 2 : 1 : 2 : 3, and the annealing temperature was 54.7℃. The optimized reaction system could be repeated stably; and the stable and reliable amplification results were able to clearly distinguish different potato varieties. This research built the solid foundation for the further study of genetic diversity of potato germplasms and construction of DNA fingerprinting.. 展开更多

Potato is one of the important food crops in Shangnan County,Shaanxi Province.Potato planting strategy in this region has great significance for potato planting,according to local conditions and for optimizing regional layout.The ecological suitability of potato cultivation in Shangnan County was evaluated by using geographic information system(GIS)technology,county cultivated land resource management information system and analytic hierarchy process(AHP).The evaluation index system of ecological suitability of potatoes in Shangnan County was established by screening the evaluation index of each ecological factor with expert experience method.The AHP and fuzzy evaluation method were applied to the quantitative calculation of the weight of each index.The ecological suitability assessment map of potato planting in Shangnan County was obtained by using the overlay analysis function of ArcGIS 10.2.The results showed that highly suitable area for potato cultivation was 6974.04 hm^2,accounting for 28.07%of the cultivated land.The suitable area was 11205.75 hm^2,accounting for 45.10%of the cultivated land area.These two types of arable land mainly distributed in the shallow hilly area,the middle mountain landform and the river valley area.The remaining arable land in this region mainly distributed in the shallow hills and the mid-mountain landscapes,including the arable land which was reluctantly suitable for potato cultivation and unsuitable for potato cultivation,with 5646.15 and 1022.33 hm^2,respectively,accounting for only 22.72%and 4.11%of the total arable land areas.Therefore,based on the statistics and analyses of potato planting environment in Shangnan County,the pattern of local ecological adaptability distribution was summarized and the spatial distribution map was drawn,thus providing scientific basis for rational distribution and planning of potato in Shangnan County. 展开更多

Potato spindle tuber viroid(PSTVd)disease is one of the major diseases that threatens potato production.Therefore,an advanced,rapid and sensitive detection technology is needed to detect the disease for better control.In order to establish an easier nucleic acid spot hybridization(NASH)method,some studies were tried as the followings:(1)the pre-hybridization step of nucleic acid spot hybridization(NASH)was omitted compared with ordinary way;(2)RNA extraction(phenol extraction and Ames buffer extraction)methods were compared;(3)fixed RNA by UV lamp and oven compared with UV cross-linker;(4)hybridized the RNA in shaking incubator and so on.The results showed that RNA extracted by Ames buffer was more effective than by the phenol extraction method.Besides,the result of hybridization without pre-hybridization step was better than that with 1.5 h of pre-hybridization.The more important discovery was that the shaking incubator could replace the hybridization oven and the ordinary UV lamp could replace the UV cross-linker.After a long term repeated research and testing,a new hybridization system that could rapidly detect the PSTVd by improved NASH technique merely using common instruments and equipment was established. 展开更多

Diploid hybrid derived from dihaploid Neo-tuberosum × Solanum phurejaand progeny of the diploid hybrid backcrossed to dihaploid Neo-tuberosum,which could produce 2n pollen, were crossed as male to S.tuberosumssp. tubersumc.v. NEA303. The performance of the 4x hybrids obtained from 4x × 2x cross was investigated for yield and yield components in the first year clonal generation. The 4x hybrids shown a drastic heterosis in plant height. Marketable yield/plant for the 4x hybrid was no better than that for the 4x cultivar, but the best hybrid individual of the two hybrid populations exceeded the best individual of the cultivar by 260 g/plant and 60 g/plant, respectively. Total yield/plant for the 4x hybrids performed drastic heterosis, however tuber number/plant was high and mean tuber weight low. These limit the use of 4x × 2x hybrids in potato breeding programs and need to be improved in near future. 展开更多

The major flow type of 6 w/v% potato starch was shear-thinning fluid even the shear rate up to 800·s-1, and the gel formed by 6 w/v% potato starch fell to weak gel for its little difference between G' and G'', high dependence on frequency and low value of G'(Pa). The hardness and cohesiveness of potato starch gel were 31.3 g and 131.9 g·s, respectively. The thermal properties of potato starch were also determined by DSC at the starch:water=3:1. The To, Tp, and ΔH of potato starch were 62.23℃,67.31℃, and 2.22 J·g-1. 展开更多

The range of variation for all the characters were found to be wide and pronounced which indicated that characters were quantitative in nature and were under polygenic control. For all the characters, greater portion of the total δ2p was mostly contributed by the δ2g. Among the seven characters, tuber weight per plant showed the highest δ2g, PCV, GCV, H2b, GA and expected GA%. On the other hand, dry matter contents (%) showed the lowest H2b. Phenotypic and genotypic correlation coefficients of plant height with no. of tubers per plant and tuber weight per plant; no. of branches per plant with no. of tubers per plant and tuber weight per plant were found to be significant and positive. 展开更多

A cryV gene,specifically toxic to Lepidoptera and Coleoptera,was incorporated into binary vectors with different promoters and the presence or absence of the β-glucuronidase(gus) reporter gene.These constructs were integrated into potato cv.Spunta by Agrobacterium tumefaciens-mediated transformation.Highest expression of cryV gene,determined by mRNA levels and insect mortality,was obtained using the CaMV 35S promoter without the gus gene configuration.Detached leaf and tuber bioassays showed a mortality rate of up to 83% and 100%,respectively,for potato tuber moth(Phthorimaea operculella Zeller) in the transgenic lines.Our results demonstrated that the presence of the gus gene negatively affects the expression level of the cryV gene.Bt expression was also facilitated by using the(ocs)3 mas super promoter,whereas the Bt expression regulated by the patatin promoter(tuber-specific) was too low to have any effect upon the mortality of potato tuber moth.These results represent significant improvement in the level of host plant resistance for the control of potato tuber moth via Bt transgenes. 展开更多

本研究以马铃薯品种辽薯6号为试验材料,设置正常水分对照(CK)及20%聚乙二醇-6000(PEG)干旱胁迫(T1)、30μmol/L褪黑素+20%PEG干旱胁迫(T2)、60μmol/L褪黑素+20%PEG干旱胁迫(T3)、90μmol/L褪黑素+20%PEG干旱胁迫(T4)和120μmol/L褪黑素... 本研究以马铃薯品种辽薯6号为试验材料,设置正常水分对照(CK)及20%聚乙二醇-6000(PEG)干旱胁迫(T1)、30μmol/L褪黑素+20%PEG干旱胁迫(T2)、60μmol/L褪黑素+20%PEG干旱胁迫(T3)、90μmol/L褪黑素+20%PEG干旱胁迫(T4)和120μmol/L褪黑素+20%PEG干旱胁迫(T5)5个处理,通过叶面喷施褪黑素,研究干旱胁迫条件下马铃薯幼苗形态、光合特性及产量对外源褪黑素的响应机理。结果表明:与CK相比,20%聚乙二醇-6000干旱胁迫处理显著降低了马铃薯幼苗的株高、茎粗、光合特性参数和单株结薯重。干旱胁迫条件下,喷施适宜浓度的褪黑素可有效改善马铃薯幼苗的生长状况及生理生化特性。60μmol/L褪黑素+20%聚乙二醇-6000干旱胁迫处理下,马铃薯幼苗株高和茎粗分别比T1处理提高了15%和29%,净光合速率、气孔导度和蒸腾速率分别比T1处理提高了21%、28%和16%,最大光化学量子效率、表观电子传递速率、光化学猝灭系数分别比T1处理提高了8%、6%、14%,超氧化物歧化酶活性、过氧化氢酶活性和抗坏血酸过氧化物酶活性分别比T1处理提高了8%、39%和9%,单株结薯数、平均单薯重和单株结薯重分别比T1处理提高了12.5%、7.1%和22.8%。综合分析认为,在20%聚乙二醇-6000干旱胁迫下,叶片喷施60μmol/L褪黑素处理有利于促进马铃薯幼苗生长,增强光合能力和抗氧化性,提高产量。 展开更多

基于改进DeepLabv3+的马铃薯幼苗与杂草识别方法 被引量:2

针对自然环境下农作物与杂草相互交织,杂草种类繁多,难以准确识别等问题,该研究以马铃薯幼苗及其伴生杂草为研究对象,提出了一种改进DeepLabv3+模型的杂草识别方法。首先以DeepLabv3+语义分割模型为基准,将其主干网络替换为MobileNetV2... 针对自然环境下农作物与杂草相互交织,杂草种类繁多,难以准确识别等问题,该研究以马铃薯幼苗及其伴生杂草为研究对象,提出了一种改进DeepLabv3+模型的杂草识别方法。首先以DeepLabv3+语义分割模型为基准,将其主干网络替换为MobileNetV2,构成轻量化DeepLabv3+模型,随后为了提升模型的非线性能力,提出了一种基于注意力机制的激活函数(attention activate function,AAF),并将其融入到AAF-Conv卷积里,取代轻量化DeepLabv3+语义分割模型中主干网络MobileNetV2的第一个3×3Conv,建立AAF-DeepLabv3+模型。使用AAF-DeepLabv3+模型获取马铃薯幼苗的形态边界,采用图像学的方法识别图像中杂草区域。在轻量化DeepLabv3+模型基础上,AAF激活函数与常见激活函数进行对比试验,平均交并比(mean intersection over Union,mIoU)分别比ReLU6、SiLU、CeLU提升了1.58、1.31、1.99个百分点,平均像素识别准确率(mean pixel accuracy,mPA)分别提升了1.47、0.6、1.26个百分点,表现出良好的性能。AAF-DeepLabv3+模型在消融试验和与其他常见语义分割模型对比中,表现出了显著的性能优势,mIoU和m PA分别为90.82%和95.56%,比原始DeepLabv3+模型提升了1.07和1.15个百分点,帧率为69.21帧/s,比原始模型提高了30.77帧/s,模型大小为22.56 MB,比原始模型降低了185.96 MB。结果表明在同一试验环境下,该模型整体性能优于UNet、PSPNet、HrNet、DeepLabv3、FCN等主流的语义分割网络模型。该杂草识别方法不仅降低了前期图片标注工作量,还有效地解决了杂草与农作物目标交叠且杂草种类繁多带来的识别难题,为移动端设备进行农田杂草识别及研制智能化除草装置提供了技术参考。 展开更多

为探索适宜的马铃薯抗性淀粉制备方法,以天薯14号马铃薯为原料,分别采用超声波辅助耐高温α-淀粉酶法、超声波辅助普鲁兰酶法、超声波辅助双酶法(耐高温α-淀粉酶法、普鲁兰酶法)及马铃薯熟化后采用超声波辅助耐高温α-淀粉酶法4种方法... 为探索适宜的马铃薯抗性淀粉制备方法,以天薯14号马铃薯为原料,分别采用超声波辅助耐高温α-淀粉酶法、超声波辅助普鲁兰酶法、超声波辅助双酶法(耐高温α-淀粉酶法、普鲁兰酶法)及马铃薯熟化后采用超声波辅助耐高温α-淀粉酶法4种方法制备抗性淀粉,比较其抗性淀粉收率、含量、溶解度、膨胀度等理化性质,并采用扫描电镜、傅里叶红外光谱仪对不同方法制备的抗性淀粉颗粒形态及分子链构象进行比较。结果表明,马铃薯熟化后采用超声波辅助耐高温α-淀粉酶法制备的抗性淀粉收率(14.25%)、含量(19.31%)及溶解度和膨胀度均高于其他方法制备的抗性淀粉,并且未产生新的官能团,但其强度比略有增加,双螺旋结构程度减小。综上,将熟化后的马铃薯采用超声波辅助耐高温α-淀粉酶法制备的抗性淀粉性质稳定,形态结构与理化性质良好,推荐将其作为马铃薯抗性淀粉优质高效的生产方法。 展开更多

为明确水杨酸甲酯(MeSA)对鲜切马铃薯品质的调控作用及其机制,以‘冀张十二号’马铃薯为试材,对比了不同浓度(0.1、0.5、1.0、1.5、2.0 mmol/L)MeSA溶液浸泡处理对4℃贮藏条件下0~12 d内鲜切马铃薯L*值、硬度、菌落总数、淀粉和抗坏血... 为明确水杨酸甲酯(MeSA)对鲜切马铃薯品质的调控作用及其机制,以‘冀张十二号’马铃薯为试材,对比了不同浓度(0.1、0.5、1.0、1.5、2.0 mmol/L)MeSA溶液浸泡处理对4℃贮藏条件下0~12 d内鲜切马铃薯L*值、硬度、菌落总数、淀粉和抗坏血酸含量的影响,在此基础上分析了最适浓度处理组的促褐变酶活性、抗氧化酶活性、细胞氧化损伤和衰老程度以及抗病代谢相关指标。结果表明:MeSA溶液处理能够维持鲜切马铃薯的色泽、硬度、淀粉含量和抗坏血酸含量,并延缓微生物的生长,其中0.5 mmol/L处理组效果最显著,进一步分析发现贮藏过程中该组鲜切马铃薯的多酚氧化酶活性、过氧化物酶活性、离子渗透率和丙二醛含量显著下降,过氧化氢酶活性、超氧化物歧化酶活性、总酚含量、苯丙氨酸解氨酶活性和β-1,3-葡聚糖酶活性显著增加,0~6 h内StNPR1相对表达量显著提高。因此,MeSA在鲜切马铃薯品质控制方面具有一定的应用潜力。 展开更多

为研究不同比例明胶/马铃薯淀粉对大豆分离蛋白(soybean protein isolate,SPI)凝胶性能及3D打印性能的影响,该文以SPI、明胶和马铃薯淀粉为原料,通过热诱导的方式制备SPI复合凝胶,并对复合凝胶的水分分布、质构、流变特性、结构、微观... 为研究不同比例明胶/马铃薯淀粉对大豆分离蛋白(soybean protein isolate,SPI)凝胶性能及3D打印性能的影响,该文以SPI、明胶和马铃薯淀粉为原料,通过热诱导的方式制备SPI复合凝胶,并对复合凝胶的水分分布、质构、流变特性、结构、微观结构以及3D打印特性等进行了表征。结果表明,明胶与马铃薯淀粉比例在1∶8~6∶8(质量比,下同)范围内,随着比例增加,自由水比例降低,不易流动水比例升高,明胶与马铃薯淀粉比例为6∶8时,不易流动水比例最高,占比为2.48%。复配凝胶的硬度、内聚性和咀嚼性随着明胶与马铃薯淀粉比例的增加逐渐降低,6∶8时达到最低,质地最为柔软。随着剪切速率升高,凝胶的黏度均下降,样品的G′和G″随着明胶与马铃薯淀粉比例的增加也均呈升高趋势。傅里叶红外光谱结果显示,添加了明胶和马铃薯淀粉的样品在波长3295.44 cm-1处的—OH振动发生红移,氢键作用增强。随着明胶与马铃薯淀粉比例的增加,复配凝胶孔径逐渐减小,明胶与马铃薯淀粉比例为6∶8时,凝胶孔隙分布呈现出最高的均匀性,打印精度最好,高度打印精确度99.60%,直径打印精确度99.92%,且在1 h内未出现明显塌陷。综上所述,明胶/马铃薯淀粉比例为6∶8时,对大豆分离蛋白凝胶3D打印性能的改善最为显著,这为开发蛋白基3D打印油墨提供理论依据。 展开更多
