In order to evaluate effects of Chinese medicinal herb (CMH) on growth and immunization of laying male chicks,eleven CMHs were used in this study,which are Astragalus membranacens (AM),Schisandra chinensis (SC),Ligustrum Lucidum(LL),Codonopsis,Scutellaria baicalensis (SB),Atractylodes macrocephala(AMA),Haw,Ginger,Acanthopanax scenticosns (AS),Angelica and Lycium,added to basal diet as 1%,respectively,with the basal diet supplemented with 50 mg·kg^(-1) bacitracin zinc(BZ) as the control.The body weight (BW) of birds were recorded at 1,21 and 42 days after birth,individually.The birds were vaccinated 0.50 mL against Newcastle disease (ND) with La sota vaccinel (containing mineral oil as adjuvant) by i.m.inoculation at 21 d of age.All of birds was vaccinated with F48E9 NDV by i.m.inoculation at 49 day of age.Blood samples were taken via wing vein from each bird on the day receiving the vaccination (Day-1) and on 14,21,28,30 and 34 d after vaccination (Days 14,21,28,30 and 34). Results showed that bodyweight (BW) and bodyweight gain(BWG) of the trial groups are similar to the control group.No significant differences of relative weight (RW) of bursa and spleen were observed among trial groups except for AS (P<0.05).Serum antibody titers of SC,LL,Codonopsis,SB,AMA and Lycium groups were significantly increased (P<0.05) in contrast to these control group on day-21 or 28 after immunized ND La sota vaccinel,which suggested that SC,LL,Codonopsis,SB,AMA and Lycium could augment antibody formulation.Furthermore,Compared with the control,antibody titers in SC,Codonopsis,AS,Lycium,SB and AMA group were higher (P<0.05) after vaccinated with F48E9 NDV,which suggested that SC,Codonopsis,AS,Lycium,SB and AMA have activity of antivirus.In conclusion,CMH used in the present study have similar effect on BW and BWG of chicks compared with the control.Moreover,these eleven CMH have little influence on RW of immunized organ except AS.However,SC,Codonopsis,LL,Lycium,SB and AMA could augment antibody formulation.Furthermore,SC,Codonopsis,AS,Lycium,SB and AMA have activity of antivirus compared with the control. 展开更多

Objective Ulcerative colitis is a prevalent immunoinflammatory disease.Th17/Treg cell imbalance and gut microbiota dysregulation are key factors in ulcerative colitis pathogenesis.The actin cytoskeleton contributes to regulating the proliferation,differentiation,and migration of Th17 and Treg cells.Wdr63,a gene containing the WD repeat domain,participates in the structure and functional modulation of actin cytoskeleton.Recent research indicates that WDR63 may serve as a regulator of cell migration and metastasis via actin polymerization inhibition.This article aims to explore the effect of Wdr63 deletion on Th17/Treg cells and ulcerative colitis.Methods We constructed Wdr63-/-mice,induced colitis in mice using dextran sulfate sodium salt,collected colon tissue for histopathological staining,collected mesenteric lymph nodes for flow cytometry analysis,and collected healthy mouse feces for microbial diversity detection.Results Compared with wild-type colitis mice,Wdr63-/-colitis mice had a more pronounced shortening of colonic tissue,higher scores on disease activity index and histological damage index,Treg cells decreased and Th17 cells increased in colonic tissue and mesenteric lymph nodes,a lower level of anti-inflammatory cytokine IL-10,and a higher level of pro-inflammatory cytokine IL-17A.In addition,WDR63 has shown positive effects on maintaining intestinal microbiota homeostasis.It maintains the balance of Bacteroidota and Firmicutes,promoting the formation of beneficial intestinal bacteria linked to immune inflammation.Conclusion Wdr63 deletion aggravates ulcerative colitis in mice,WDR63 inhibits colonic inflammation likely by regulating Th17/Treg balance and maintains intestinal microbiota homeostasis. 展开更多

AIM:Regulatory T cells(Tregs)are a specialized subset of CD4^(+)T cells primarily involved in im⁃munosuppressive functions.AMP-activated protein kinase(AMPK)serves as a metabolic sensor that governs the differen⁃tiation,maturation,and immune functions of Tregs through metabolic reprogramming.However,the impact of AMPKα1(the catalytic subunit of AMPK)knockout specifically in Tregs on the host's immune microenvironment remains largely un⁃explored.METHODS:Histological changes in immune organs were assessed using HE staining.The types of immune cells and their relative population percentages in immune organs and blood were quantified through flow cytometry in both AMPKα1flox/flox(AMPKα1^(fl/fl))mice and Treg-specific AMPKα1 knockout mice(AMPKα1^(fl/fl)Foxp3^(cre)mice).RESULTS:Compared to AMPKα1^(fl/fl)mice,the percentage of eosinophils in the bone marrow of AMPKα1^(fl/fl)Foxp3^(cre)mice was significant⁃ly reduced.Additionally,while the thymus of AMPKα1^(fl/fl)Foxp3^(cre)mice exhibited normal structure,both its size and the ratio of thymus weight to body weight were significantly decreased.The knockout of AMPKα1 in Tregs led to a notable reduction in the total percentage of immature double-negative(DN)cells.Consequently,the percentage of CD4^(+)T cells derived from these DN cells also decreased,even though the percentages of DN1 and DN4 cells were higher in the thymus of AMPKα1^(fl/fl)Foxp3^(cre)mice compared to AMPKα1^(fl/fl)mice.Importantly,the proportion of Siglec-F+CD11b^(+)eosinophils in the thymus was significantly lower in AMPKα1^(fl/fl)Foxp3^(cre)mice.Knockout of AMPKα1 in Tregs resulted in a marked increase in the percentage of CD4^(+)T cells in peripheral blood,alongside a decrease in the proportion of mature CD8^(+)T cells.Similarly,the proportion of CD4^(+)T cells in the spleen of AMPKα1^(fl/fl)Foxp3^(cre)mice was elevated compared to AMPKα1^(fl/fl)mice.In contrast,the proportion of neutrophils significantly decreased,while mononuclear cell proportions increased in the spleen of AMPKα1^(fl/fl)Foxp3^(cre)mice.In lymph nodes,the medullary boundaries in AMPKα1^(fl/fl)Foxp3^(cre)mice were blurred,and the lymphoid follicles were missing,a feature not observed in AMPKα1^(fl/fl)mice.Furthermore,the knockout of AMPKα1 in Tregs reduced the CD3^(+)T cell population,particularly the CD8^(+)T cell population,in lymph nodes.Although the mature Treg cell population was significantly lower in AMPKα1^(fl/fl)Foxp3^(cre)mice,the percentage of CD4^(+)T cells was markedly in⁃creased.In contrast,there was no statistically significant difference in granulocyte populations between AMPKα1^(fl/fl)Foxp3^(cre)and AMPKα1^(fl/fl)mice.CONCLUSION:The populations of mature Tregs,CD8^(+)T cells and eosinophils in various im⁃mune organs were significantly altered in mice with Treg-specific AMPKα1 knockout,suggesting a potential remodeling of the host immune microenvironment in response to inflammatory stimuli. 展开更多

Construction of CD8^(+)T cell-associated Risk Model in Hepatocellular Carcinoma Based on Bulk and Single-cell RNA-seq Data

Hepatocellular carcinoma(HCC),which is essentially primary liver cancer,is closely related to CD8^(+)T cell immune infiltration and immune suppression.We constructed a CD8^(+)T cells related risk score model to predict the prognosis of HCC patients and provided therapeutic guidance based on the risk score.Using integrated bulk RNA sequencing(RNA-seq)and single-cell RNA sequencing(scRNA-seq)datasets,we identified stable CD8^(+)T cell signatures.Based on these signatures,a 3-gene risk score model,comprised of KLRB1,RGS 2,and TNFRSF1B was constructed.The risk score model was well validated through an independent external validation cohort.We divided patients into high-risk and low-risk groups according to the risk score and compared the differences in immune microenvironment between these two groups.Compared with low-risk patients,high-risk patients have higher M2-type macrophage content(P<0.0001)and lower CD8^(+)T cells infiltration(P<0.0001).High-risk patients predict worse response to immunotherapy treatment than low-risk patients(P<0.01).Drug sensitivity analysis shows that PI3K-β inhibitor AZD6482 and TGFβRII inhibitor SB505124 may be suitable therapies for high-risk patients,while the IGF-1R inhibitor BMS-754807 or the novel pyrimidine-based anti-tumor metabolic drug Gemcitabine could be potential therapeutic choices for low-risk patients.Moreover,expression of these 3-gene model was verified by immunohistochemistry.In summary,the establishment and validation of a CD8^(+)T cell-derived risk model can more accurately predict the prognosis of HCC patients and guide the construction of personalized treatment plans. 展开更多

Objective:To evaluate the predictive value of the neutrophil⁃to⁃lymphocyte ratio(NLR)and the systemic immune⁃inflammation index(SII)in predicting patients with anti⁃melanoma differentiation⁃associated gene 5⁃positive(anti⁃MDA5+)dermatomyositis(DM)develop into the rapidly progressive interstitial lung disease(RPILD).Methods:We retrospectively analyzed the clinical and laboratory data of 124 anti⁃MDA5+DM patients from the First Affiliated Hospital of Nanjing Medical University between March 2019 and September 2023.We identified independent risk factors associated with the development and mortality of RPILD with the Cox regression analysis,and determined the optimal cut⁃off values for predicting adverse outcomes with the receiver operating characteristic(ROC)curve analysis.Results:Among the 124 patients,36 patients(29.03%)developed RPILD,and 39 patients(31.45%)died during the follow⁃up period.The results of multivariate Cox regression analysis showed that the elevated NLR was an independent risk factor for RPILD development,while the elevated SII expression was independently associated with the increased mortality of RPILD.Based on the ROC curve analysis,NLR>6.12 was a predictor for RPILD,and SII>875.79 was associated with increased mortality risk of RPILD.Conclusion:Both NLR and SII are accessible,cost⁃effective,and reliable prognostic indicators for the prognosis of patients with anti⁃MDA5^(+)DM,providing a valuable guidance for clinical management and risk stratification of the disease. 展开更多

Objective:Osteosarcoma is a highly aggressive primary malignant bone tumor commonly seen in children and adolescents,with a poor prognosis.Anchorage-dependent cell death(anoikis)has been proven to be indispensable in tumor metastasis,regulating the migration and adhesion of tumor cells at the primary site.However,as a type of programmed cell death,anoikis is rarely studied in osteosarcoma,especially in the tumor immune microenvironment.This study aims to clarify prognostic value of anoikis and tumor immune microenvironment-related gene in the treatment of osteosarcoma.Methods:Anoikis-related genes(ANRGs)were obtained from GeneCards.Clinical information and ANRGs expression profiles of osteosarcoma patients were sourced from the therapeutically applicable research to generate effective therapies and Gene Expression Omnibus(GEO)databases.ANRGs highly associated with tumor immune microenvironment were identified by the estimate package and the weighted gene coexpression network analysis(WGCNA)algorithm.Machine learning algorithms were performed to construct long-term survival predictive strategy,each sample was divided into high-risk and low-risk subgroups,which was further verified in the GEO cohort.Finally,based on single-cell RNA-seq from the GEO database,analysis was done on the function of signature genes in the osteosarcoma tumor microenvironment.Results:A total of 51 hub ANRGs closely associated with the tumor microenvironment were identified,from which 3 genes(MERTK,BNIP3,S100A8)were selected to construct the prognostic model.Significant differences in immune cell activation and immune-related signaling pathways were observed between the high-risk and low-risk groups based on tumor microenvironment analysis(all P<0.05).Additionally,characteristic genes within the osteosarcoma microenvironment were identified in regulation of intercellular crosstalk through the GAS6-MERTK signaling pathway.Conclusion:The prognostic model based on ANRGs and tumor microenvironment demonstrate good predictive power and provide more personalized treatment options for patients with osteosarcoma. 展开更多

Perilla essential oil(PEO)is reported as an aromatic yellowish oily substance with a volatile odor extracted from perilla leaves.It exhibits various biological activities except anti-tobacco mosaic virus(TMV)activity.In this study,we investigated the main components and anti-TMV activity of PEO,identified its primary active components,and examined its mode of action.The results indicated that PEO exhibited anti-TMV activity(65.58%)at 800μg/mL,with perillaldehyde identified as the main active component.The protective,curative,and inactivation activities of perillaldehyde at 800μg/mL were 80.41%,73.42%,and 34.93%,respectively.These values were significantly higher than those of the control drug(commercial chitosan oligosaccharide)and the protective and curative activities were superior to those of ningnanmycin.The results of the mode of action showed that perillaldehyde induced a hypersensitive response(HR)in tobacco.Transmission electron microscope(TEM)observation revealed that perillaldehyde had no direct effect on TMV particles.The treatment of Nicotiana glutinosa with perillaldehyde at 800μg/mL indicated that perillaldehyde had significant induction activity(58.46%).The expression of three pathogenesis-related tobacco genes(PR genes),including nonexpressor of pathogenesis-related genes 1(NPR1),pathogenesis-related protein 1 gene(PR1),and pathogenesis-related protein 5 gene(PR5),were induced and upregulated by perillaldehyde treatment.Perillaldehyde also induced the overexpression of the phenylalanine ammonia-lyase gene(PAL),respiratory burst oxidase homolog B gene(RBOHB),and protochlorophyllide oxidoreductase gene 1(POR1).Furthermore,perillaldehyde increased the salicylic acid(SA)and H2O2 contents in tobacco leaves,and enhanced the activities of four defense enzymes:superoxide dismutase(SOD),catalase(CAT),peroxidase(POD),and phenylalanine ammonia-lyase(PAL).N.glutinosa was treated with perillaldehyde at 800μg/mL for 24 h,and the results showed that the highest SA and H_(2)O_(2) contents(1032.08 pmol/L and 23.40μmol/g FW,respectively)were obtained in tobacco leaves.Defense enzyme activities also reached a maximum at 800μg/mL,and the activities of CAT,PAL,POD,and SOD increased by 1.76,1.95,2.17,and 3.78 times,respectively,compared to the control.The results of the study showed that perillaldehyde may enhance resistance to pathogen infection by inducing systemic acquired resistance(SAR),which may contribute to the activation of SA signal transduction pathway.Therefore,perillaldehyde has the potential for application in agriculture as a novel antiviral agent and immune inducer. 展开更多

In order to ensure overall optimization of the underground metal mine production scale, multidisciplinary design optimization model of production scale which covers the subsystem objective function of income of production, safety and environmental impact in the underground metal mine was established by using multidisciplinary design optimization method. The coupling effects from various disciplines were fully considered, and adaptive mutative scale chaos immunization optimization algorithm was adopted to solve multidisciplinary design optimization model of underground metal mine production scale. Practical results show that multidisciplinary design optimization on production scale of an underground lead and zinc mine reflect the actual operating conditions more realistically, the production scale is about 1.25 Mt/a (Lead and zinc metal content of 160 000 t/a), the economic life is approximately 14 a, corresponding coefficient of production profits can be increased to 15.13%, safety factor can be increased to 5.4% and environmental impact coefficient can be reduced by 9.52%. 展开更多

In order to achieve the computerization and digitization of the daily system of stud-farm, the immunization system technology, racecourse health monitoring technologies and the stud-farm prevention technologies were combined to built the studfarm daily management system. By using Visual Studio 2008 and Microsoft Office Access 2003, an immunization information system was developed based on Client/Server structure and combined with the key immunization link of horses. The system included studfarm immune management and racecourse health monitoring system. The immune management contained immunization information input, the inquiry of immunization information, immunization statistics, immunization-reminding and system management. The racecourse health monitoring system included stud-farm monitoring, horses monitoring and inquiry and statistics of health monitoring. The software realized the analysis and tips of the daily information in stud-farm. The software could record the appropriate data and carry on the statistics. The stud-farm daily management system greatly improved the efficiency of the expanded program about immunization and horses' healthy rearing technique. 展开更多

In order to control the locomotive wheel(axle) load distribution, a shimming process to adjust the locomotive secondary spring loads was heretofore developed. An immune dominance clonal selection multi-objective algorithm based on the artificial immune system was presented to further improve the performance of the optimization algorithm for locomotive secondary spring load adjustment, especially to solve the lack of control on the output shim quantity. The algorithm was designed into a two-level optimization structure according to the preferences of the problem, and the priori knowledge of the problem was used as the immune dominance. Experiments on various types of locomotives show that owing to the novel algorithm, the shim quantity is cut down by 30% 60% and the calculation time is about 90% less while the secondary spring load distribution is controlled on the same level as before. The application of this optimization algorithm can significantly improve the availability and efficiency of the secondary spring adjustment process. 展开更多

To fight against malicious codes of P2P networks, it is necessary to study the malicious code propagation model of P2P networks in depth. The epidemic of malicious code threatening P2P systems can be divided into the active and passive propagation models and a new passive propagation model of malicious code is proposed, which differentiates peers into 4 kinds of state and fits better for actual P2P networks. From the propagation model of malicious code, it is easy to find that quickly making peers get their patched and upgraded anti-virus system is the key way of immunization and damage control. To distribute patches and immune modules efficiently, a new exponential tree plus (ET+) and vaccine distribution algorithm based on ET+ are also proposed. The performance analysis and test results show that the vaccine distribution algorithm based on ET+ is robust, efficient and much more suitable for P2P networks. 展开更多

The weapon transportation support scheduling problem on aircraft carrier deck is the key to restricting the sortie rate and combat capability of carrier-based aircraft.This paper studies the problem and presents a novel solution architecture.Taking the interference of the carrier-based aircraft deck layout on the weapon transportation route and precedence constraint into consideration,a mixed integer formulation is established to minimize the total objective,which is constituted of makespan,load variance and accumulative transfer time of support unit.Solution approach is developed for the model.Firstly,based on modeling the carrier aircraft parked on deck as convex obstacles,the path library of weapon transportation is constructed through visibility graph and Warshall-Floyd methods.We then propose a bi-population immune algorithm in which a population-based forward/backward scheduling technique,local search schemes and a chaotic catastrophe operator are embedded.Besides,the randomkey solution representation and serial scheduling generation scheme are adopted to conveniently obtain a better solution.The Taguchi method is additionally employed to determine key parameters of the algorithm.Finally,on a set of generated realistic instances,we demonstrate that the proposed algorithm outperforms all compared algorithms designed for similar optimization problems and can significantly improve the efficiency,and that the established model and the bi-population immune algorithm can effectively respond to the weapon support requirements of carrier-based aircraft under different sortie missions. 展开更多

Aiming at the problem on cooperative air-defense of surface warship formation, this paper maps the cooperative airdefense system of systems (SoS) for surface warship formation (CASoSSWF) to the biological immune system (BIS) according to the similarity of the defense mechanism and characteristics between the CASoSSWF and the BIS, and then designs the models of components and the architecture for a monitoring agent, a regulating agent, a killer agent, a pre-warning agent and a communicating agent by making use of the theories and methods of the artificial immune system, the multi-agent system (MAS), the vaccine and the danger theory (DT). Moreover a new immune multi-agent model using vaccine based on DT (IMMUVBDT) for the cooperative air-defense SoS is advanced. The immune response and immune mechanism of the CASoSSWF are analyzed. The model has a capability of memory, evolution, commendable dynamic environment adaptability and self-learning, and embodies adequately the cooperative air-defense mechanism for the CASoSSWF. This pa- per proposes an immune algorithm based method (BN-IA) for the learning of the BN structure with the idea of vaccination. Further- more, the methods on how to extract the effective vaccines from local optimal structure and root nodes are also described in details. Finally, the simulation studies are implemented with the helicopter convertor BN model and the car start BN model. Memory and self-regulation mechanisms of IA were used to avoid PSO plunging into local optima. Vaccination and immune selection mechanisms were used to prevent the undulate phenomenon during the evolutionary process. The algorithm was introduced through an application in the direct maintenance cost (DMC) estimation of aircraft components. Experiments results show that the algorithm can compute simply and run quickly. It resolves the combinatorial optimization problem of component DMC estimation with simple and available parameters. And it has higher accuracy than individual methods, such as PLS, BP and v-SVM, and also has better performance than other combined methods, such as basic PSO and BP neural network. 展开更多
