Objective To explore the sequential effects of hypoxic exercising on miR-27/PPARγand lipid metabolism targetgene and protein expression levels in the obesity rats’liver.Methods 13-week-old male diet-induced obesity rats were randomlydivided into three groups(n=10):normal oxygen concentration quiet group(N),hypoxia quiet group(H),hypoxic exercise group(HE).Exercise training on the horizontal animal treadmill for 1 h/d,5 d/week for a total of 4 week,and the intensity of horizontaltreadmill training was 20 m/min(hypoxic concentration was 13.6%).Comparison of the weights of perirenal fat and epididymal fat in rats across different groups and calculation of Lee’s index based on body weight and body length of rats in each group were done.And the serum concentrations of total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high-densitylipoprotein cholesterol(HDL-C)levels were detected.RT-PCR and Western Blot were used to detect the levels of miR-27,PPARγ,CYP7A1 and CD36.Results Hypoxic exercise decreased the expression levels of miR-27 in the obese rats’liver,however,theexpression level of PPARγwas gradually increased.The expression levels of miR-27 in HE group were significantly lower than Ngroup(P<0.05).The expression levels of PPARγmRNA in N group were significantly lower than H group(P<0.05),especially lowerthan HE group(P<0.01).The protein expression of PPARγprotein in N group was significantly lower than that other groups(P<0.01).The expression of lipid metabolism-related genes and proteins increased in the obese rats’liver.The expression of CYP7A1mRNA in N group was significantly lower than H group(P<0.05),especially lower than HE group(P<0.01).The expression ofCYP7A1 protein in the obese rats’liver in N group was extremely lower than H group and HE group(P<0.01).The proteinexpression of CD36 in N group was significantly lower than that in HE group(P<0.05).Hypoxia exercise improved the relatedphysiological and biochemical indexes of lipid metabolism disorder.The perirenal fat weight of obese rats in HE group wasextremely lower than N group and H group(P<0.01),and the perirenal fat weight in N group was significantly higher than H group(P<0.05).The epididymal fat weight in N group was significantly higher than H group(P<0.05),and extremely higher than HEgroup(P<0.01).The Lee’s index in HE group was extremely lower than N group and H group(P<0.01).The serum concentration ofTC in obese rats in HE group was extremely lower than N group and H group(P<0.01).The serum concentration of TG in HE groupwas extremely lower than N group and H group(P<0.01).The serum concentration of LDL-C in N group was extremely higher thanHE group(P<0.01).The serum concentration of HDL-C in N group was extremely lower than H group(P<0.01).Conclusion Hypoxiaand hypoxia exercise may negatively regulate the levels of PPARγby inhibiting miR-27 in the obese rats’liver,thereby affecting theexpression of downstream target genes CYP7A1 and CD36,and promoting cholesterol,fatty acid oxidation and HDL-C transport inthe liver,and ultimately the lipid levels in obese rats were improved.The effect of hypoxia exercise on improving blood lipid isbetter than simple hypoxia intervention. 展开更多

Pulse electrical arc stimulator based on single-electrode for active exercise in tail-suspension rat

To make rat do active exercise to counteract bone loss in the rat tail-suspension model, a pulse electrical stimulator based on single-electrode with a low-current and a high-voltage was designed. The stimulator was controlled by SCM (single chip micyoco) that could accurately control the stimulation duration and the interval between stimulations, and cease the operation after the recorded number of stimulation had reached the value set by the program. With the help of posture estimation part, the device would operate intelligently by determining whether to stimulate or not, depending on the posture of rat's limb. Software was developed to make operator control the stimulator using computer, save the experiment data and print the report. In practical experiment, the voltaic arc is generated by the stimulator, and impacted on the rat's thenar. This induced pain to the rat and the rat would actively contract its hindlimb to evade the pain, so active exercise was carried out. The tail-suspension rats were trained twice every day for 14 d. At the 0 and 14th day, bone mineral density of rat femurs was determined by dual energy X-ray absorptiometry (DXA). The results show that the active exercise stimulated by the pulse electrical arc stimulator can attenuate weightlessness-induced bone loss, and this device is a convenient steady performance electrical stimulator that can surely induce rat's hindlimb to do active exercise. 展开更多

目的观察高强度间歇训练(HIIT)对肥胖女大学生体成分、血脂和空腹胰岛素(FIN)水平的影响。方法 2015年1—6月选取浙江旅游职业学院符合纳入与排除标准的在校肥胖女大学生36例为研究对象。按研究对象体质指数(BMI)从小到大进行排序,先以... 目的观察高强度间歇训练(HIIT)对肥胖女大学生体成分、血脂和空腹胰岛素(FIN)水平的影响。方法 2015年1—6月选取浙江旅游职业学院符合纳入与排除标准的在校肥胖女大学生36例为研究对象。按研究对象体质指数(BMI)从小到大进行排序,先以4例为1层(共分为9层),采用随机数字表法将每层研究对象分为运动组和对照组,最终每组纳入18例。两组均给予自我运动锻炼宣教,运动组在此基础上给予集中的HIIT。于试验开始前1周及试验结束后1周检测研究对象体成分指标〔体质量、体质指数(BMI)、体脂肪、体脂百分比、内脏脂肪、去脂体质量、静息代谢率、腹围、臀围〕、血脂〔三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)〕水平、FIN水平。结果运动组试验后体质量、BMI、体脂肪、体脂百分比、腹围小于对照组,去脂体质量、静息代谢率大于对照组(P<0.05)。运动组试验后体质量、BMI、体脂肪、体脂百分比、腹围小于试验前,去脂体质量、静息代谢率大于试验前(P<0.05)。运动组试验后HDL-C水平高于对照组,LDL-C、FIN水平低于对照组(P<0.05)。运动组试验后HDL-C水平高于试验前,LDL-C、FIN水平低于试验前(P<0.05)。结论HIIT可以降低肥胖女大学生体脂含量,增加静息代谢率,同时改善血脂和FIN水平,对改善肥胖女大学生的健康水平有积极影响。 展开更多

目的:分析不同强度跑台运动对幼龄大鼠海马组织脑源性神经营养因子(BDNF)和谷氨酸N-甲基-D-天冬氨酸(NMDA)受体mRNA水平的影响。方法:将43只5周龄雄性SD大鼠随机分为对照组、小强度、中等强度和大强度运动组。通过定量real-time PCR技术... 目的:分析不同强度跑台运动对幼龄大鼠海马组织脑源性神经营养因子(BDNF)和谷氨酸N-甲基-D-天冬氨酸(NMDA)受体mRNA水平的影响。方法:将43只5周龄雄性SD大鼠随机分为对照组、小强度、中等强度和大强度运动组。通过定量real-time PCR技术,分析1周跑台运动后各组大鼠海马组织内BDNF和NMDA R1 mRNA的水平。结果:各组大鼠海马内BDNF mRNA水平分别是:对照组为746±338,小强度运动组为1 308±688,中等强度运动组为1 036±649,大强度运动组为722±426;NMDA R1 mRNA水平分别是:对照组为34±19,小强度运动组为286±230,中等强度运动组为174±132;大强度运动组为20±14。结果表明小强度运动既可明显诱导海马BDNF基因表达(P<0.01),也可明显诱导海马内NM- DA R1基因的表达(P<0.01);中等强度运动可明显诱导NMDA R1受体基因的表达(P<0.01),但对BDNF的表达已没有明显作用;而大强度运动对2种因子的基因表达皆无明显影响。结论:不同强度的运动可调节海马内BDNF和谷氨酸NMDA R1受体基因的表达,但以小强度跑台运动促进作用最强,中等运动强度次之,而大强度运动基本没有影响。 展开更多

目的探讨12周及24周的有氧运动对急性心肌梗死患者心功能及氧代谢功能的影响。方法选取50例急性ST段抬高型心肌梗死并行急诊PCI的患者,随机分为有氧运动治疗(运动组)及非运动治疗(对照组)各25例,2组患者给予相同药物治疗,于入院后第8天... 目的探讨12周及24周的有氧运动对急性心肌梗死患者心功能及氧代谢功能的影响。方法选取50例急性ST段抬高型心肌梗死并行急诊PCI的患者,随机分为有氧运动治疗(运动组)及非运动治疗(对照组)各25例,2组患者给予相同药物治疗,于入院后第8天、第12周及24周检查无氧阈值、峰值氧耗量、以及LVEF、N末端B型钠尿肽前体(NT-proBNP)水平。结果与对照组比较,运动组第12周及24周后无氧阈值[(15.05±5.80)ml/(kg·min)vs(14.78±1.50)ml/(kg·min),(15.94±0.86)ml/(kg·min)vs(14.43±1.82)ml/(kg·min),峰值耗氧量[(22.31±2.50)ml/(kg·min)vs(19.50±2.52)ml/(kg·min),(23.06±2.13)ml/(kg·min)vs(19.79±2.89)ml/(kg·min)],LVEF[(55.8±2.8)%vs(53.3±5.1)%,(56.5±2.9)%vs(53.7±5.2)%]明显升高,而NT-proBNP[(2171.4±1014.0)ng/L vs(2922.8±1342.6)ng/L,(1628.2±740.1)ng/L vs(2476.7±964.8)ng/L]明显降低(P<0.05,P<0.01)。与同组第8天比较,2组12、24周后无氧阈值、峰值氧耗量、LVEF明显升高,而NT-proBNP明显降低(P<0.05,P<0.01),运动组24周较12周时NT-proBNP明显降低(P<0.05)结论有氧运动治疗可以明显改善急性心肌梗死患者的心功能及氧代谢指标。 展开更多

Establishment of an animal model of acute mountain sickness

Aim To simulate the condition when people travel to the plateau. To make a primary investigation about the mice models, which are respectively provoked by exhaustive exercise, cold and fatigue, and epinephrine. Methods Divide the Kunming mice into 8 groups in random, with 10 in a single group, control group in plain; exhaustive exercise group in plain; fatigue & cold group in plain; epinephrine stimulating group in plain; control group in plateau; exhaustive exercise group in plateau; fatigue & cold group in plateau; epinephrine stimulating group in plateau. To simulate the condition when people travel to Lhasa whose altitude reaches 3700m, those mice were carried there during a five-day period. Day 1: Weight the mice after fasting for 12hrs, then treated different with corresponding methods. Day 2: Collect the data of mortality in each group and sacrifice the alive. Weight the cardiac, pulmonary and cerebral tissues and calculate for the Viscera Index. Results After the journey to the plat- eau, the weight of mice decreases significantly, in exhaustive exercise group and fatigue & cold group also with the increase of pulmonary and cerebral index and decrease of cardiac index compared with groups in plain. As for those who are stimulated with epinephrine, the ones in plateau suffer more from pneumonedema but have a longer life span. The sensitivity to epinephrine can decrease in female mice in plateau, which can be reversed in plain. Con- clusions After the journey to plateau, acute plateau pneumonedema and cerebral edema can be provoked by ex- haustive exercise, fatigue and cold, and starvation; The severity of pneumonedema caused by epinephrine are relat- ed to the environment, strength and gender. 展开更多
