Insights on Peripheral Blood Biomarkers for Parkinson’s Disease

Parkinson’s disease(PD)is a common neurodegenerative disorder with profound impact on patients’quality of life and long-term health,and early detection and intervention are particularly critical.In recent years,the search for precise and reliable biomarkers has become one of the key strategies to effectively address the clinical challenges of PD.In this paper,we systematically evaluated potential biomarkers,including proteins,metabolites,epigenetic markers,and exosomes,in the peripheral blood of PD patients.Protein markers are one of the main directions of biomarker research in PD.In particular,α‑synuclein and its phosphorylated form play a key role in the pathological process of PD.It has been shown that aggregation ofα-synuclein may be associated with pathologic protein deposition in PD and may be a potential marker for early diagnosis of PD.In terms of metabolites,uric acid,as a metabolite,plays an important role in oxidative stress and neuroprotection in PD.It has been found that changes in uric acid levels may be associated with the onset and progression of PD,showing its potential as an early diagnostic marker.Epigenetic markers,such as DNA methylation modifications and miRNAs,have also attracted much attention in Parkinson’s disease research.Changes in these markers may affect the expression of PD-related genes and have an important impact on the onset and progression of the disease,providing new research perspectives for the early diagnosis of PD.In addition,exosomes,as a potential biomarker carrier for PD,are able to carry a variety of biomolecules involved in intercellular communication and pathological regulation.Studies have shown that exosomes may play an important role in the pathogenesis of PD,and their detection in blood may provide a new breakthrough for early diagnosis.It has been shown that exosomes may play an important role in the pathogenesis of PD,and their detection in blood may provide new breakthroughs in early diagnosis.In summary,through in-depth evaluation of biomarkers in the peripheral blood of PD patients,this paper demonstrates the important potential of these markers in the early diagnosis of PD and in the study of pathological mechanisms.Future studies will continue to explore the clinical application value of these biomarkers to promote the early detection of PD and individualized treatment strategies. 展开更多

OBJECTIVE Our previous studies demonstrated that various ingredients from the traditional Chinese medicine(TCM)for promoting blood circulation and removing blood stasis,as exemplified by cryptotanshinone and salvianolic acid B,exerted striking effects on modulating angiogenesis and vascular permeability,which suggests that they may be effective in treating vascular leak-driven diseases(e.g.tumor,cerebral cavernous malformation and diabetic retinopathy).However,the lack of reliable and advanced technologies and models sets up difficult hurdles for better understanding the role of TCM for promoting blood circulation and removing blood stasis.To this end,this study is to outline numerous cutting-edge platforms that can be utilized for exploring the function of TCM for promoting blood circulation and removing blood stasis in vascular leak-driven diseases.METHODS Two-photon laser scanning fluorescence microscopy was used to observe the interactions between neutrophils and blood vessels in a real-time manner.Dynamic flow system was employed to mimic the in vivo behaviors of neutrophils.RIP1-Tag5 spontaneous pancreatic cancer model was used to study the function of tumor blood vessels.CCM2ECKO(deletion of CCM2 in endothelial cells)mice were employed to establish the cerebral cavernous malformation(CCM)animal model.Micro-computed tomography(micro-CT)was utilized to assess the CCM lesion.Müller cell-knockout mouse model was used to study the progression of diabetic retinopathy.Vascular permeability in this model was assessed by fluorescein angiography.RESULTS The interactions between neutrophils and endothelial cells involve a series of complicated processes,including rolling,adhesion,intraluminal crawling and transmigration,which were all monitored in vivo by two-photon laser scanning fluorescence microscopy in a real-time manner.Dynamic flow system was capable of recapitulating the biological behaviors of neutrophils in vitro.Tumor vascular function in particular vascular perfusion could be assessed in the RIP1-Tag5 spontaneous pancreatic cancer model.In terms of CCM studies,specific deletion of CCM2 in endothelial cells resulted in the initiation of CCM lesion.The size and number of CCM lesions could be visualized and quantified by micro-CT.Furthermore,the Müller cell-knockout mouse model was able to precisely reflect the clinical symptoms of diabetic retinopathy.Vascular leak could be monitored at different time points using fluorescein angiography.CONCLUSION An array of high technologies and animal models can be used in investigating the occurrence and progression of multiple vascular leak-driven diseases.The pre-clinical and clinical studies of TCM for promoting blood circulation and removing blood stasis provide fundamental support for the application of the above-mentioned platforms,with the purpose of uncovering the scientific basis of TCM for promoting blood circulation and removing blood stasis. 展开更多

According to the characteristics of dynamic firing in pulse coupled neural network (PCNN) and regional configuration in retinal blood vessel network, a new method combined with simplified PCNN and fast 2D-Otsu algorithm was proposed for automated retinal blood vessels segmentation. Firstly, 2D Gaussian matched filter was used to enhance the retinal images and simplified PCNN was employed to segment the blood vessels by firing neighborhood neurons. Then, fast 2D-Otsu algorithm was introduced to search the best segmentation results and iteration times with less computation time. Finally, the whole vessel network was obtained via analyzing the regional connectivity. Experiments implemented on the public Hoover database indicate that this new method gets a 0.803 5 true positive rate and a 0.028 0 false positive rate on an average. According to the test results, compared with Hoover algorithm and method of PCNN and 1D-Otsu, the proposed method shows much better performance. 展开更多

OBJECTIVE To investigate the role of chemokine-like factor 1(CKLF1),a novel C-C chemokine,on brain-blood barrier(BBB)integrity in rat focal cerebral ischemia and reperfusion model.METHODS Antibodies against CKLF1 was applied to the rightcerebral ventricle immediately after transient middle cerebral artery occlusion.Brain water content,Evans blue leakage and the expression of aquaporin-4(AQP-4),matrix metalloproteinase-9(MMP-9),zonula occludens-1(ZO-1)and occludin were measured.RESULTS After treatment with antiCKLF1 antibody,brain water content and Evans blue leakage in ipsilateral hemisphere were decreased in a dose-dependent manner at 24 h after reperfusion,but not changed in contralateral hemisphere.Anti-CKLF1 antibody reduced the expression of AQP-4 and MMP-9,and upregulated the expression of ZO-1 and Occludin.These results suggest that CKLF1 is involved in BBB disruption after reperfusion.CONCLUSION Inhibition of CKLF1 protects against cerebral ischemia by maintaining BBB integrity,possibly via inhibiting the expression of AQP-4 and MMP-9,and increasing the expression of tight junction protein. 展开更多

OBJECTIVE To evaluate the effects of Tong-Qiao-Huo-Xue decoction(TQHXD)on the bloodbrain barrier(BBB)permeability and the expressions of related proteins on the rats;and to analyse the constituents in the cerebrospinal fluid on the rats with cerebral ischemic injury.METHODS Cerebral ischemia rats were induced by middle cerebral artery occlusion(MCAO).Adult male sprague-dawley(SD)rats were randomly divided into seven groups:sham-group;model group;nimodipine(NMP)-treated group and nao mai tai(NMT)-treated group were set as positive drug control groups;TQHXD-treated group(3,6 and 12g·kg-1body weight);The neurological function of rats was estimated by neurological defect scoring after the 1,7and 15 dafter administration.Histological structure of the brain in rats were observed by hematoxylin and eosin(H&E)staining.Ultramicrostructural features of hippocampus neurons and the opening of tight junction(TJ)of BBB in rats were observed by transmission electron microscope(TEM).Western blotting was performed to detect the expression of ZO-1,occludin,claudin-5,AQP-4 and MMP-9 in BBB after cerebral ischemia injury.Component analysis experiments:adult male SD rats were randomly divided into four groups:Distilled water was administered intragastrically sham-operated rats;Distilled water was administered intragastrically model rats by MCAO;TQHXD was administered intragatrically to rats in sham-operated group;TQHXD was administered intragestrically to rats in model group by MCAO.GC and HPLC was used to detect three compounds,namely,muscone,ligustilide and hydroxysafflor yellow A,in rats cerebrospinal fluid(CSF)after oral administration of TQHXD.Finally,samples of cerebrospinal fluid of rats in each group were compared with single medicine so as to explicit the three compounds come from which herb.RESULTS TQHXD significantly reduced the neurological defect scores.Histological examination indicated that dense neuropil and largely surviving neurons had been seen in TQHXD-treated rats.TEM observation revealed that TQHXD could significantly inhibit the damage of hippocampal neurons and reduce the opening of TJ.The decreased protein expression levels of claudin-5,occludin,ZO-1 and the increased protein expression levels of AQP-4 and MMP-9in cerebral ischemia tissue were significantly prevented by treatment of TQHXD.Analysis of experimental results showed that muscone,ligustilide and hydroxysafflor yellow A could penetrate the BBB into the CSF,and the content of the model group was lower than that of sham group after intragastric administration of TQHXD.CONCLUSION These results demonstrated that TQHXD may act as a potential neuroprotective agent against BBB damage for cerebral ischemia through protecting of hippocampus neurons,reducing the opening of TJ and decreasing the permeability of BBB by up-regulating ZO-1,occludin,claudin-5 expressions,down-regulating AQP-4 and MMP-9 expressions.The effect of TQHXD on the decrease of the opening of TJ also reduced the content of muscone,ligustilide and hydroxysafflor yellow A in cerebrospinal fluid. 展开更多

The effects of various toxic carbonyls such as malondialdehyde(MDA),a secondary product of lipid peroxidation,and other aldehydes on rheological parameters and their relationship with aging-associated alterations were studied.Both MDA and glutaraldehyde(Glu) in different concentrations significantly increase viscosity,plastic viscosity and yield stress of human plasma and erythrocyte suspensions.MDA(20 mmol/L) reduces sharply the typical fluorescence of proteins(excitation 280 nm/emission 350 nm),and produces age pigment-like fluorescence with a strong emission peak at 460 nm when excites at 395 nm by only being incubated for some hours.In contrast,Glu decreases merely the fluorescence of proteins without producing age pigment-like fluorescence.These data suggest interestingly that the MDA-induced gradual protein cross linking seems to form from different mechanisms compared to the fast rheological changes of blood materials which may take place either in acute and chronic diseases or during aging.On the other hand,MDA induces various deleterious alterations of erythrocytes whereas glutathione(GSH) inhibits the MDA-related carbonyl stress in a concentration-dependent manner.The results indicate that carbonyl-amino reaction exists in the blood widely and GSH has the ability to interrupt or reverse this reaction in a certain way.It implies that carbonyl stress may be one of the important factors in blood stasis and suggests a theoretical and practical approach in anti-stresses and anti-aging. 展开更多

The purpose of this study was to develop a quantitative structure–property relationship(QSPR) model based on the enhanced replacement method(ERM) and support vector machine(SVM) to predict the blood-to-brain barrier partitioning behavior(log BB) of various drugs and organic compounds. Different molecular descriptors were calculated using a dragon package to represent the molecular structures of the compounds studied. The enhanced replacement method(ERM) was used to select the variables and construct the SVM model. The correlation coefficient, R^2, between experimental results and predicted log BB was 0.878 and 0.986, respectively. The results obtained demonstrated that, for all compounds, the log BB values estimated by SVM agreed with the experimental data, demonstrating that SVM is an effective method for model development, and can be used as a powerful chemometric tool in QSPR studies. 展开更多

OBJECTIVE To explore the effect of ethanolic extract of Physalis minima on lowering blood pressure in the DOCA-salts hypertensive rats.METHODS To produce hypertension,male rats were induced DOCA at dose of 10mg·kg-1 ip.twice weekly and 0.9% NaCl in drinking water ad libitum for six weeks.The ethanolic extract of Physalis minima were given at dose of 500,1500 and 2500mg·kg-1,respectively for 4weeks.After completion of treatment schedule,systolic blood pressure was evaluated using animal blood pressure analyzer.The serum nitric oxide levels was measured using the ELISA methods.SOD activity and MDA level were determined spectrophotometrically.Endothelial dysfunction was determined by acetylcholine-induced vasorelaxation studies on isolated rat aortas.RESULTS The extract decreased systolic blood pressure significantly(P<0.001)in hypertensive rats.In addition,the extract also suppressed the production of MDA and restored SOD activity to normal level.There was a reduced relaxation response to acetylcholine in isolated aortic rings and serum NO level from hypertensive rats that were reversed by extract.CONCLUSION Physalis minima can lower blood pressure in hypertensive rats and its mechanism may be through the improvement of endothelial function due to the decrease of stress oxidative. 展开更多

Blood cells are mainly(~99%)comprised of red blood cells.The most remarkable properties of are their high deformability,which allow they flow through microcapillaries of diameter even smaller than their size.The RBC’s remarkable mechanical properties originate from the unique architecture of its membrane.To study the mechanism of RBC’s deformability,a commonly adopted approach is to localize the cytoskeleton protein by immunofluorescence,followed by exploring the changes of cytoskeleton protein during cell deformation.During this process,the fixed treatment of RBC using GA and PFA is of great importance.However,RBC’s deformability is reduced by the fixation process and skeletal protein of membrane is changed accordingly.The flow behavior of red RBCs through the microchannel also changed.Given the difficulty of observing RBC flow in vivo,in vitro simulation by virtue of microfluidic devices provides a feasible alternative.An important physiological phenomenon of the blood flow is the formation of cell free layer(CFL),with RBCs show a tendency to concentrate towards the central axis of the pipeline and move faster than the plasma layer.However,this phenomena is weaken if the stiffness of the membrane increase,which occurs in some disease,such as hereditary spherocytosis and hereditary elliptocytosis.To study the effects of GA and PFA fix treatment on RBC deformability,a microfluidic platform is employed to measuring the CFL and flow velocity of blood flow in this work.The PDMS micro flow channel used is 100 micrometers in width and 50 micrometers in deep.The RBC suspension is fed into the flow channel by the injection pump(NE-1000.USA),and the experiments are observed and recorded by the inverted microscope(IX70,Olympus,Japan)and high-speed camera(Memrecam GX-1,NAC,Japan)system.Three GA concentrations,i.e.,0.000 5,0.000 75,and 0.001 wt.%were used.Meanwhile,the effect of PFA at a concentration of 2wt.%work with GA was also investigated.Images of the flowing RBCs are processed mainly based on Memrecam GXLink.The results show that,the diameter of the RBC be treated is bigger and the shape of the RBC is became more flat after treated.Some of RBCs lost their biconcave structure.When the RBC suspension with 5%Hct flow in the microchannel,the CFL thickness decrease after being treated.And with concentrations of GA increase,the CFL thickness become thinner.The CFL thickness decrease significantly when GA and 2 wt.%PFA work combined.The velocity of RBCs decreases after treated with the GA or/and 2wt.%PFA.GA is known to relieve the dissolution of red blood cells during fluorescence labeling.On the other hand,the crosslinking of the aldehyde group(-cho)of GA with the amino group(-nh2)of RBC membrane protein will change the conformation of the membrane protein and its visco-elastic properties in turn.Then,the transparent fluidity orrheology characteristics of RBC is altered.Since GA and PFA are commonly used to immobilize red blood cells and keep the fluorescence constant,and PFA works similarly as GA,as a result,the variation of membrane protein conformation is intensified,and the membrane becomes stiffer. 展开更多

It was found that after 9-11 Gy of radiation was given, the 90-daysurvival rate of the rats receiving BT(72%) and BMT was significantly higher than thatof those receiving BMT only(42%)(P<0.01).In those rats surviving over 100 days,cells of donor type could be found. In the first 30 days of surviving, the number of Tcells was significantly higher in the BMT alone group than in the group of BMT plusBT, but no difference In restoration of B cell was revealed. The findings suggest that BTcould promote the recipient's tolerance to BMT. The effects of BT on BMT are similarto those on skin grafting. 展开更多

To study the effects of Bai-Hua-Qian-Hu on cardiovascular system,emphasis was put on the research of Bai-Hua-Qian-Hu extracted with water and alcohol on coronary flow and some cardiovascular indices in both isolated preparations and intact normal animals. The results showed that extract obtained with water and alcohol (Pd-wa) increased coronary flow in isolated rabbit heart and in the anaesthetized cats, Pd-wa antagonised the strip contraction either in canine coronary artery or in rabbit aorta, and increased myocardial nutrient blood flow in mice, The values of LD<sub>50</sub> by peritoneal and intravenous injection proved that Pd-wa was very safe. 展开更多

The author has observed the epithelium and basement membrane of uterus, oviduct and fimbriae in mice under the TEM by oral administration of AgNo<sub>3</sub> tracer. The results showed that there was obvious deposit of argentum granules outside the basement membrane of mucosal epithelium of the uterus, oviduct and fimbriae as well as outside the basement membrane of uterine glands. And there was dilatation fully filled with high electron-dense argentum granules at the intercellular space just beneath the tight junction between the mucosal epithelial cells of uterus, oviduet, fimbriae. All these finding suggest that the uterus, oviduct and fimbriae serve as barriers to the heavy metal argentum ions. 展开更多

Clematichinenoside protects blood brain barrier against ischemic stroke superimposed on systemic inflammatory challenges through up-regulating A20

Suppression of excessive inflammation can ameliorate blood brain barrier (BBB) injury, which shows therapeutic potential for clinical treatment of brain injury induced by stroke superimposed on systemic inflammatory diseases. In this study, we investigated whether and how clematichinenoside (AR), an anti-inflammatory triter- pene saponin, protects brain injury from stroke superimposed on systemic inflammation. Lipopolysaccharide (LPS) was intraperitoneally injected immediately after middle cerebral artery occlusion (MCAO) in rats. Rat microvessel endothelial cells (rBMECs) were exposed to hypoxia/reoxygenation (H/R) coexisting with LPS. The results re- vealed that AR suppressed the excessive inflammation, restored BBB dysfunction, alleviated brain edema, de- lessened neurological dysfunction, and decreased infarct rate. Further study demon- creased neutrophil infiltration, inducible nitric oxide synthase (iNOS) , strated that the expression of nucleus nuclear factor kappa B (NF-KB), intercellular adhesion molecule-1 ( ICAM-1 ), tumor necrosis factor-α (TNF-α) and interlukin-1β ( IL-1β) were suppressed by AR via zinc finger protein A20. Besides, AR increased in vitro BBB integrity through A20. In con- clusion, AR alleviated cerebral inflammatory injury through A20-NF-KB signal pathway, offering an alternative medication for stroke associated with systemic inflammatory diseases. 展开更多

The integrity and function dysfunction of the blood brain barrier(BBB)is considered to be an early event in the pathogenesis of a variety of neurological diseases in old patients and dysfunction of BBB could be induced bycommon stress that old people often face,such as sepsis during which lipopolysaccharide(LPS)is released into circulation and BBB is damaged.Since decreased melatonin level had been shown in the serum and brain of old people and mice,we aim to investigate whether supplement with melatonin could alleviate LPSinduced BBB damage in old mice.Mice(24-28 months old)received one week melatonin(10 mg·kg-1·d-1,intraperitoneally,ip)or saline before challenge with LPS(1 mg·kg-1,ip).Evan′s blue(EB)and immunoglobulin G(Ig G)leakage were used to assess the BBB permeability.Immunofluoresence and Western blotting were used to detect the protein expression and distribution.Our results showed that LPS significantly increased BBB permeability in old mice accompanied by tight junction protein occludin and claudin-5 degradation,inhibition of AMP-activated protein kinase(AMPK)activation and increase of gp91phox protein expression.Interestingly,one week of melatonin treatment significantly decreased LPS-induced BBB hyperpermeability,activated AMPK and inhibited gp91phox expression upregualtion.Moreover,activation of AMPK by metformin significantly inhibited LPS-induced gp91phox expression upregualtion in endothelial cells.Taken together,our findings demonstrate that melatonin alleviates LPS-impaired integrity of BBB by activation of AMPK and inhibition of gp91phox expression in old mice. 展开更多
