Three breeds of sows were observed to investigate the relationship between Single Nucleotide Polymorphisms(SNPs) in Mu Opioid Receptor(MOR)and stereotypic behaviour,such as,sham-chewing,bar biting and standing still in order to better understand the mechanism of stereotypic development of the animals in restrained conditions.MOR exon 2 partial sequences were amplified to analyze single nucleotide polymorphisms by PCR-SSCP.One SNP,a silence mutant was found.A significant difference (P〈0.01)was found in the frequency of genotypes in these 3 breeds where only the BB genotype,which was identical to that published in GenBank,was found in the Duroc breed,while no AA genotype was found in Landrace,3 genotypes AA,BB and AB were found in Yorkshire.The result also indicated that the individuals with AA and AB genotypes tended to be more active in sham-chewing than those with the BB genotype(P〈0.05).The overall results of this study suggested that sham-chewing of sows may be subjected to both genetic control and environmental conditions,but activity level was more likely to be affected by their environment.We can putatively draw the conclusion that MOR gene has effect on the sham-chewing behavioral traits of sow. 展开更多

Objective The detection of RNA single nucleotide polymorphism(SNP)is of great importance due to their association with protein expression related to various diseases and drug responses.At present,splintR ligase-assisted methods are important approaches for RNA direct detection,but its specificity will be limited when the fidelity of ligases is not ideal.The aim of this study was to create a method to improve the specificity of splintR ligase for RNA detection.Methods In this study,a dualcompetitive-padlock-probe(DCPLP)assay without the need for additional enzymes or reactions is proposed to improve specificity of splintR ligase ligation.To verify the method,we employed dual competitive padlock probe-mediated rolling circle amplification(DCPLP-RCA)to genotype the CYP2C9 gene.Results The specificity was well improved through the competition and strand displacement of dual padlock probe,with an 83.26%reduction in nonspecific signal.By detecting synthetic RNA samples,the method demonstrated a dynamic detection range of 10 pmol/L-1 nmol/L.Furthermore,clinical samples were applied to the method to evaluate its performance,and the genotyping results were consistent with those obtained using the qPCR method.Conclusion This study has successfully established a highly specific direct RNA SNP detection method,and provided a novel avenue for accurate identification of various types of RNAs. 展开更多

试验旨在研究饲料中不同水平的核苷酸(Nucleotide)对草鱼(Ctenopharyngodon idella)生长性能的影响,为核苷酸在草鱼养殖生产中的高效应用提供参考。试验选择560尾体重约为10g的健康的草鱼,按照单因素试验设计,将其均分4组,每组5个重复,... 试验旨在研究饲料中不同水平的核苷酸(Nucleotide)对草鱼(Ctenopharyngodon idella)生长性能的影响,为核苷酸在草鱼养殖生产中的高效应用提供参考。试验选择560尾体重约为10g的健康的草鱼,按照单因素试验设计,将其均分4组,每组5个重复,每组重复28尾草鱼。对照组草鱼投喂基础饲料,核苷酸组草鱼分别投喂含有0.2%、0.4%和0.6%的核苷酸的试验饲料。投喂60 d后,检测各组草鱼的生长性能。结果显示,与对照组比较,草鱼投喂0.4%的核苷酸可以使试验末重增加(P<0.05),增重率增加17.28%(P<0.05),可以使饲料系数显著降低(P<0.05)。而核苷酸对草鱼的特定生长率、成活率、脏体比和肝体比无显著影响(P>0.05)。研究表明,适量的核苷酸可以促进草鱼生长,且核苷酸添加量为0.4%时,生长性能效果最好。 展开更多

Selective killing p53wT lung cancer cells harboring Arg72 homozygote by a small molecular CypA inhibitor

TP53, encoding a well-known tumor suppressor p53, plays essential roles in tumor initiation and pro- gression, and is frequently mutated in lung cancer. However, pharmacological stabilization and reactivation of p53 have not been actively explored for targeted cancer therapies. Here, we identified a novel Cyclophilin A (CypA) small molecule inhibitor ( HL001 ) that induces non-small cell lung cancer (NSCLC) cell cycle arrest and apoptosis via restoring p53 expression, and further stabilizes p53 through inhibiting the MDM2-mediated p53 ubiqutination. The down-regulation of G3BP1 by HL001 also contributes to p53 stabilization by inhibiting p53 redistribution from nucleus to cytoplasm. Furthermore, HE001 selectively suppresses tumor growth in p53 wild-type NSCLC harboring Arg72 homozygous alleles (p53-72R) through disrupting interaction between MDM2 and p53-72R in a CypA-de- pendent manner. Finally, administration of HE001 alone or co-treatment with cisplatin promotes significant tumor suppression in orthotopic NSCLC mouse model. Collectively, our preclinical study demonstrated that HE001, a small molecule inhibitor of CypA, selectively activated p53WT 72R homozygote and thus inhibits growth of human lung cancer cells. The results presented here demonstrate that the utility of CypA inhibitors serve as an approach to the targeted therapy for individual lung cancer patient. 展开更多

The function of Rad23 protein involved in DNA damage repair from many environmental factors has been studied extensively, but it is not clear from ultraviolet irradiation. To further investigate the photo-protective function of Rad23 protein on HeLa cells damaged from UV light irradiation, firstly, HeLa cells were irradiated by UV light and incubated with the fusion protein of pCold-Rad23, then the cell viability and apoptosis rate were detected by MTT and Hoechst33342/Pl fluorescent staining, respectively. The results show that the recombinant Rad23 protein can protect the HeLa cells from UV irradiation, and inhibit the apoptosis of HeLa cell by UV irradiation. 展开更多

OBJECTIVE SNPs in lnc RNAs may alter the expression or secondary structure of lnc RNAs and then impact their functions.Whether lnc RNA SNPs affect the prognosis of acute myeloid leukemia(AML)remains unknown.To search the association between lnc RNA SNPs and AML outcomes,thirty tag SNPs in GAS5,H19,MALAT1,WT1-as and SRA were genotyped in313 AML patients.METHODS Survival analysis was performed in both AML patients recruited presently and GEO samples.The expression of GAS5 and TP63 was analyzed by real-time quantitative PCR.Dual-luciferase reporter gene assay was used to confirm the interactions between GAS5 rs55829688 and TP63.RESULTS Survival analysis indicated that rs55829688(T>C),located in GAS5 promoter,was significantly associated with the prognosis of AML.The average overall survival(OS)for patients with the rs55829688 CC genotype was significantly shorter than those carrying the rs55829688 T allele(P=0.018).Patients with rs55829688 CC genotype showed higher GAS5 expression in PBMCs than carriers of rs55829688T allele(P=0.025).Rs55829688 CC homozygotes also harbored a longer platelets recovery than those with rs55829688 T allele(P=0.040).In vitro study showed that GAS5 promoter harboring the rs55829688 C al ele showed marginal y increased reporter gene activity(P=0.054),and the promoter activity was increased by TP63 in a dose-dependent manner(P=0.001).Moreover,GAS5 expression was associated with AML OS in the GEO GSE12417 dataset,and GAS5 higher expression predict shorter OS(P=0.011).CONCLUSION Rs55829688 polymorphism could increase GAS5 expression by interacting with TP63 and was associated with worse OS in Chinese AML patients. 展开更多
