Objective To investigate whether 2,3,5,4'-tetrahydroxystilbene-2-O-β-glucoside(TSG)ameliorated polycystic ovary syndrome(PCOS)-like characteristics by inhibiting inflammation.Methods PCOS models were established by injecting subcutaneously with dehydroepiandrosterone into female Sprague-Dawley rats,followed by receiving intraperitoneal injection of TSG.The granular cells(GCs)KGN were transfected with small interfering RNAs(si-NC and si-CYP19A1).The cells were preincubated with lipopolysaccharide(LPS)and then treated with or without TSG.The estrous cycle was monitored using vaginal exfoliated cells.The morphology of ovarian follicles was analyzed by H&E staining.ELISA was used to analyze estradiol(E2),testosterone(T),follicle stimulating hormone(FSH),luteinizing hormone(LH),IL-6,TNF-α,AGEs,CRP and Omentin-1 levels in serum.Immunohistochemistry was performed to analyze PCNA and CYP19A1 expressions in the GCs of ovaries.Tunel staining was executed to detect the apoptosis of GCs.Quantitative polymerase chain reaction(qPCR)and Western blot were implemented to measure the expression of CYP19A1 in the ovaries and transfected cells.qPCR was used to analyze the expression of IL-6 and TNF-αin the transfected cells treated with LPS and TSG.Results The estrous cycles were restored in TSG group.Compared with model group,the sinus follicles were reduced and corpus luteums were increased in TSG group.TSG group showed increased E2,and decreased T and LH,compared with model group.Pro-inflammatory factors(IL-6,TNF-α,CRP and AGEs)were decreased,and anti-inflammatory factor(Omentin-1)was increased in TSG group compared with those in model group.TSG could partially inhibit decrease of PNCA-positive GCs and increase of Tunel-positive GCs caused by PCOS.The CYP19A1 expression of GCs in TSG group was upregulated compared with model group.The expressions of IL-6 and TNFαin si-CYP19A1 cells were increased compared with si-NC cells.Compared with cells(si-NC and si-CYP19A1)treated without LPS,the expressions of IL-6 and TNF-αcells were increased,and the expression of CYP19A1 was downregulated in LPS-preincubated cells.Compared with cells treated with LPS,the expression of IL-6 and TNF-αwere decreased,and the expression of CYP19A1 was increased in cells treated with LPS and TSG.Compared with si-NC cells treated with LPS and TSG,the expressions of IL-6 and TNF-αcells were increased in the si-CYP19A1 cells treated with LPS and TSG.Conclusion TSG could alleviate PCOS-like characteristics by increasing the expression of CYP19A1 in GCs to inhibit inflammatory response. 展开更多

Anti-inflammatory effects of ethanol fraction from Disporum cantoniense (Lour.) Merr. in LPS-stimulated RAW264. 7 cells

Merr. 70% ethanol extract with a cellular model of LPS-stimulated RAW264.7 cell. Methods RAW264.7 cells were treated with different concentrations of ethanol fraction (25,50 and 100 g · L^-1 ) and stimu- the conditioned media was collected and analyzed. The concentrations of inflammatory medi- 9 ators were calculated according to the standard curves generated by each of the recombinant cytokines provided with LPS can induce RAW264.7 cells to promote the pro- the ELISA kits. Results Compared with the control group, TNF-α, IL-1β and IL-6. Compared with the duction of inflammatory mediators (P 〈 0.01 ) , including NO, PGE2, model group, ethanol fraction significantly suppressed LPS induced release of inflammatory mediators such as nitric NO, PGE2, TNF-α, IL-1β and IL-6 in a good dose dependent manner (P 〈 0.05, P 〈 0.01 ). Conclusions Eth- anol fraction could significantly inhibit the production of LPS-induced inflammatory response in RAW264.7 cells, and its anti-inflammatory effect may be related to reduce the production of inflammatory mediators NO, PGE2, TNF-α, IL-1 β and IL-6. These results demonstrate that the ethanol fraction is the bioactive component of Disporum can- toniense (Lour.) Merr. , and the ethanol fraction will be further developed as a herbal remedy for preventive and/ or curative purposes in various inflammatory diseases. 展开更多

OBJECTIVE To study the therapeutic effects of TGP on SS both in C57BL/6J mice immunized by immu⁃nological induction(SS mice)and NOD/ShiltJNju(NOD)mice.METHODS TGP(180,360,720 mg·kg^-1)was intragastri⁃cally administered for 6 or 16 weeks for SS mice and NOD mice,respectively.Weekly food and water intake,saliva flow,submandibular gland(SMG)and spleen index,and SMG pathology were measured.ELISA was used to evaluate serum interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)and autoantigens(SSA/Ro,SSB/La,andα-fodrin).Real-time PCR and Luminex liquid suspension chip assay were applied to analyze SMG inflammatory cytokines mRNA TNF-α,IL-17A,CXCL9,CXCL13,and B-cell activating factor(BAFF)and protein(IL-1β,IL-6,TNF-α,and IFN-γ)expres⁃sion.RESULTS Compared with SS mice,TGP(720 mg·kg^-1)treatment increased saliva flow,reduced organ indexes,and decreased serum IL-6 and IFN-γ concentration.TGP(360 mg·kg^-1)treatment decreased serum IFN-γ concentra⁃tion.TGP(180,360,720 mg·kg^-1)treatment improved SMG pathological damage.Compared with NOD mice,the saliva flowincreased from 9 to 15 weeks of administration.After 2 weeks of administration,TGP(720 mg·kg^-1)treatment decreased serum SSA/Ro,SSB/La and a-fodrin concentration,increased SMG index,inhibited SMG IFN-γ concentra⁃tion,and down-regulated SMG TNF-α,IL-17A,CXCL9,CXCL13 and BAFF mRNA expression.TGP(360 mg·kg^-1)treat⁃ment decreased serum SSB/La and a-fodrin,and SMG TNF-α and IFN-γ concentration,and down-regulated SMG TNF-α,IL-17A,CXCL9 and BAFF mRNA expression.TGP(180 mg·kg^-1)treatment decreased serum SSB/La,a-fodrin,and SMG IL-1β concentration,and down-regulated SMG TNF-α,IL-17A and BAFF mRNA expression.After 8 weeks of administration,TGP(180,360,720 mg·kg^-1)treatment increased SMG index,and decreased serum a-fodrin concentra⁃tion.TGP(720 mg·kg^-1)treatment down-regulated mRNA expression of SMG TNF-α,IL-17A,CXCL9,CXCL13,and BAFF.TGP(360 mg·kg^-1)treatment reduced mRNA expression of TNF-α,CXCL9,CXCL13 and BAFF,and concentra⁃tion of IL-6 and TNF-α.TGP(180 mg·kg^-1)treatment down-regulated mRNA expression of TNF-α,CXCL9,and CXCL13,and decreased IL-6 and TNF-αconcentration in SMG.After 16 weeks of administration,TGP(180,360,720 mg·kg^-1)treatment reduced serum SSA/Ro and a-fodrin concentration,increased SMG index,and decreased SMG CXCL13 and BAFF mRNA expression.TGP(360,720 mg·kg^-1)treatment decreased serum SSB/Laconcentration and SMG TNF-α,IL-17A and CXCL9 mRNA expression.Besides,TGP(180,360,720 mg·kg^-1)treatment alleviated the pathological damage of SMG after 2 and 16 weeks of administration.CONCLUSION TGP has a certain therapeutic effect onmice through inhibiting inflammatory responses. 展开更多

OBJECTIVE Psoriasis is an immune system meditated disease,especially T cells.It disturbed many people around the world and hard to therapy.Paeonia lactiflora Pall has been used as a medicine in china for thousands of years.Recent studies has found that the main component of Paeonia lactiflora Pall can alleviates the immune response in many diseases.In this study,we researched the effects and possible mechanisms of total glucosides of paeony(TGP)on animal psoriasis in order to study the therapeutic effects and mechanisms of TGP in 5%propranolol creaminduced psoriasis in guinea pigs and Imiquimod(IMQ)cream-induced psoriasis in mice.METHODS The effect of TGP was evaluated using a psoriasis-like model of guinea pigs and mice.Ear thickness was accessed,and pathology injury was observed by HE staining.The levels of serum IL-1β,IL-6,IL-12,IL-17,IL-23,TNF-α,and IFN-γ,skin IL-17A,IL-22 and orphan nuclear receptor(RORγt)mRNA expression,proliferating cell nuclear antigen(PCNA),total or phosphorylated signal transducers and activators of transcription(STAT1 and STAT3)were determined by ELISA,real time PCR,immu⁃nohistochemical staining,and Western blotting,respectively.RESULTS Compared with model group,TGP treatment decreased the ear thickness,improved pathology of psoriasis,alleviated IMQ-induced keratinocyte proliferation,reduced the inflammatory cytokine,and downregulated IL-17A,IL-22,and RORγt mRNA in mice.Further study indicated that TGP inhibited STAT1 and STAT3 phosphorylation in lesion skins of psoriasis-like mice.CONCLUSION TGP alleviates the symptoms of psoriasis-like guinea pigs and mice,and the possible mechanism may relate to inhibit T helper 17(TH17)cell differentiation and keratinocytes proliferation by inhibiting STAT1 and STAT3 phosphorylation. 展开更多

Aim To study the anti-hypertensive effect of total flavonoids of Cydonia oblonga Mill. in spontaneously hypertensive rats, then preliminarily investigate the mechanism of action based on anti-inflammatory function. Methods Total flavonoids of COM (COMF) were separated and purified by the optimal process, SHR were divid- ed into 6 groups as SHR control group (SHR), captopril group (CAP, 25mg· kg^-1 · d^-1), Eucommia ulmoides Oliver group (EUO, 30 mg · kg^-1· d^-1), low (COMF-L, 40 mg · kg^-1· d^-1), middle (COMF-M, 80 mg · kg^-1 · d^-1) and high dose (COMF-H, 160 mg · kg^-1· d ^-1) of COMF groups. 8 Wistar-Kyoto (WKY) rats were given distilled water instead as control. The drugs were given by intragastric administration for 16 weeks, then the levels of IL-1β, IL-6, IL-10, TNF-α and CRP in serum were tested. Results Compared with WKY group, the levels of IL-I~, IL-6, TNF-α and CRP were increased and IL-10 was decreased; Compared with SHR group, the levels of IL-1β, IL-6, TNF-α and CRP were decreased in COMF groups (P 〈0.01 or P 〈0.05) , while IL-10 was increased a little but without any significant difference (P 〈 0.05 ). Conclusion Total flavonoids of Cydonia ob- longa Mill. showed the anti-hypertensive activity in spontaneously hypertensive rats and the mechanism was related to the function of anti-inflammatory. 展开更多

Expression of Inflammatory Factors in Bovine Reproductive Tract During Follicular and Luteal Phases

In order to improve reproduvtive efficieny and understand reproduvtive defense mechanism, the oviduct, uterine horn and uterine body of bovine were used to detect the changes of inflammatory factors and the relationship between estrogen and progesterone receptor protein during estrous cycle by real-time PCR and Elisa method. The results showed that interleukin-4(IL-4), interleukin-6(IL-6), interleukin-10(IL-10), interleukin-1α(IL-1α) and interleukin-1β(IL-1β) were expressed in cow oviduct, uterine horn and uterine body. In the follicular phase and the luteal phase, m RNA expression of five inflammatory factors in the uterine horn and uterine body was higher than that in the oviduct. In the follicular phase, IL-10 was highly expressed in the uterine horn and uterine body, IL-4 was highly expressed in the uterine horn, uterine body and oviduct. Additionally, in the luteal phase, IL-6 and IL-1β were highly expressed in the uterine horn, uterine body and oviduct, and the highest expression of IL-1β was observed in the uterine horn. The levels of Estrogen Receptor(ERα) protein in the oviduct, uterine horn and uterine body significantly increased in the follicular phase. The levels of Progesterone Receptor(PR) protein in the same portions of the reproductive tract in the luteal phase were significantly higher than those in the follicular phase. IL-4 and IL-10 in the cow reproductive tract might play a major role in the follicular phase, while IL-6 and IL-1β might play a major role in the luteal phase. The expression of five inflammatory factors was not directly regulated by ERα and PR. 展开更多

Currently,research on N^(6)-methyladenine(m^(6)A)is extensive in the field of oncology,while studies involving m^(6)A and skin diseases remain relatively limited.Based on existing reports,we searched PubMed and Web of Science for literature related to m^(6)A and dermatological conditions.Analysis of citation counts and journal impact factors revealed a significant upward trend in the volume of m^(6)A-related research.Term frequency analysis of titles and abstracts indicated that studies mainly focus on skin tumors and inflammatory or immune-related skin diseases,particularly melanoma,psoriasis,and skin development.Transcriptomic data from the Gene Expression Omnibus(GEO)were analyzed,revealing differential expression of m^(6)A-related genes in 4 types of skin tumors(including squamous cell carcinoma and basal cell carcinoma)as well as in inflammatory skin diseases such as psoriasis and atopic dermatitis,and potential mechanisms of action were also explored.Findings suggest that m^(6)A modifications exhibit heterogeneity between neoplastic and nonneoplastic skin diseases.However,the regulatory mechanisms of m^(6)A dynamic modifications on key genes involved in dermatological disorders remain unclear and warrant further investigation. 展开更多

目的研究酪酸梭菌干预对高尿酸血症大鼠血清尿酸、脂多糖、白介素6、肿瘤坏死因子α的影响。方法将40只SD大鼠随机分为正常对照组、高尿酸模型组、苯溴马隆干预组(3 mg/kg·d)和酪酸杆菌干预组(活菌1.5×107CFU/d),每组10只。... 目的研究酪酸梭菌干预对高尿酸血症大鼠血清尿酸、脂多糖、白介素6、肿瘤坏死因子α的影响。方法将40只SD大鼠随机分为正常对照组、高尿酸模型组、苯溴马隆干预组(3 mg/kg·d)和酪酸杆菌干预组(活菌1.5×107CFU/d),每组10只。除正常对照组外各组给予酵母膏联合氧嗪酸钾灌胃制备高尿酸血症模型,各组药物及活菌每天灌胃1次,连续12周。观察各组大鼠血尿酸、脂多糖、白介素6、肿瘤坏死因子α水平变化。结果实验期间高嘌呤饮食大鼠血尿酸水平较正常对照组明显升高(P<0.01),高尿酸血症模型建立成功且稳定。高尿酸模型组大鼠血脂多糖、白介素6、肿瘤坏死因子α水平与血尿酸水平呈正相关。苯溴马隆及酪酸梭菌干预处理后大鼠血尿酸、脂多糖、白介素6、肿瘤坏死因子α水平有明显下降(P<0.01),干预时间越长,效果越佳,其中苯溴马隆降尿酸效应更为明显,酪酸梭菌下调脂多糖、白介素6、肿瘤坏死因子α等炎性因子作用更为明显(P<0.01)。结论高尿酸血症大鼠可能存在慢性炎症反应,酪酸梭菌可有效降低血尿酸水平,同时抑制炎症细胞因子的生成,维持肠道免疫稳态。 展开更多
