Memory and self-regulation mechanisms of IA were used to avoid PSO plunging into local optima. Vaccination and immune selection mechanisms were used to prevent the undulate phenomenon during the evolutionary process. The algorithm was introduced through an application in the direct maintenance cost (DMC) estimation of aircraft components. Experiments results show that the algorithm can compute simply and run quickly. It resolves the combinatorial optimization problem of component DMC estimation with simple and available parameters. And it has higher accuracy than individual methods, such as PLS, BP and v-SVM, and also has better performance than other combined methods, such as basic PSO and BP neural network.展开更多
To promote the modeling standardization process of the integrated circuits, an improved electrical simulation model for a direct power injection (DPI) setup which was used to measure the conducted immunity of a 16-b...To promote the modeling standardization process of the integrated circuits, an improved electrical simulation model for a direct power injection (DPI) setup which was used to measure the conducted immunity of a 16-bit microcontroller to radio frequency aggression was investigated. Based on the existing model of the same microcontroller, the PDN module was modified by adding the core, PLL and MD network models, which could reflect the actual electric distribution situation within the microcontroller more accurately. By comparing the simulation results with the measurement results, the effectiveness of the modified model can be improved to 500 MHz, and its uncertainty is within +1.8 dB (+2 dB is acceptable). Then, to improve the simulation accuracy of the complete model in the high frequency range, the I/O model which contained the dynamic and nonlinear characteristics reflecting the variation of the internal impedance of the microcontroller with increasing the frequency of the external noise was introduced. By comparing the simulation results with the measurement results, the effectiveness of the second modified model can be improved up to 1.4 GHz with the uncertainty of ~1.8 dB. Thus, a conclusion can be reached that the proposed model can be applied to a much wider frequency range with a smaller uncertainty than the latest model of the similar type. Furthermore, associated with the electromagnetic emission testing platform model, the PDN module can also be used to predict the electromagnetic conducted and radiated emission characteristics. This modeling method can also be applied to other integrated circuits, which is very helpful to the standardization of the IC electromagnetic compatibility (EMC) modeling process.展开更多

Six diets...Exogenous alanyl-glutamine(Aln-Gln) was evaluated for its effects on growth performance, intestinal structure and function, antioxidant status and non-specific immunity of young carp(Cyprinus carpio L.). Six diets supplemented with 0, 2.5, 5.0, 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln were fed to fish for 12 weeks. Supplementation with 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln significantly increased weight gain rate(WGR), protein efficiency ratio(PER), but feed conservation rate(FCR) and survival were not affected(P〉0.05). The intestinal fold height and number, digestive enzyme, Na+, K+-ATPase activities was found to be significantly high(P〈0.05) with increasing dietary Aln-Gln supplementation up to 7.5 g · kg-1, but there were no significant differences for Aln-Gln supplementation from 7.5 to 15.0 g · kg-1. The glutathione peroxidase(GPX) activity, glutathione(GSH), superoxide dismutase(SOD) activity increased and malondialdehyde(MDA) levels decreased significantly(P〈0.05) in the intestine, hepatopancreas, plasma and muscles. The plasma complement-3(C3) and complement-4(C4) levels were significantly(P〈0.05) improved at 5.0 g · kg-1 level and decreased when over 7.5 g · kg-1. The plasma lysozyme(LSZ) activity increased significantly(P〈0.05) at 7.5, 10.0, or 15.0 g · kg-1 level. In summary, the results showed that Aln-Gln improved growth performance, development and function of the intestine, the activity of the antioxidant defense system and the plasma non-specific immunity of the carps. The optimal Aln-Gln level was 8.24 g · kg-1 diet for WGR based on broken-line regression model analysis.展开更多

Simulation programme and three functions were used to check the effect of the algorithm. The advanced algorithm were compared with clonal selection algorithm and particle swarm algorithm. The control group (Group I) was fed with corn-soybean meal as basal diet, and the trial groups (group Ⅱ, Ⅲ, Ⅳ, Ⅴ and Ⅵ) were fed with basal diet plus 0.1% manoligosaccharides (MOS), 0.3% soybean-oligosaccharides (SBOS), 0.3% fructooligosaccharides (FOS), 0.3% α-glucooligosaccharides(α-GOS) and 0.03% chlortetracycline (CTC), respectively. The results showed that oligosaccharides supplementation significantly (P< 0.05) increased bifidobacteria concentration of young broilers and did not affect on bifidobacteria or escherichia coli concentration of the birds ( P >0.05). Cecal weights of 21 d old broilers fed with FOS and α-GOS were significantly(P <0.05) increased, higher than those with MOS and SBOS, but addition of oligosaccharides had no effect on the birds. The effect of oligosaccharides on the cecal length was the same as on the cecal weights. Oligosaccharides supplement did not affect on indexes of immune organs.展开更多
Hepatocellular carcinoma(HCC),which is essentially primary liver cancer,is closely related to CD8^(+)T cell immune infiltration and immune suppression.We constructed a CD8^(+)T cells related risk score model to predic...Hepatocellular carcinoma(HCC),which is essentially primary liver cancer,is closely related to CD8^(+)T cell immune infiltration and immune suppression.We constructed a CD8^(+)T cells related risk score model to predict the prognosis of HCC patients and provided therapeutic guidance based on the risk score.Using integrated bulk RNA sequencing(RNA-seq)and single-cell RNA sequencing(scRNA-seq)datasets,we identified stable CD8^(+)T cell signatures.Based on these signatures,a 3-gene risk score model,comprised of KLRB1,RGS 2,and TNFRSF1B was constructed.The risk score model was well validated through an independent external validation cohort.We divided patients into high-risk and low-risk groups according to the risk score and compared the differences in immune microenvironment between these two groups.Compared with low-risk patients,high-risk patients have higher M2-type macrophage content(P<0.0001)and lower CD8^(+)T cells infiltration(P<0.0001).High-risk patients predict worse response to immunotherapy treatment than low-risk patients(P<0.01).Drug sensitivity analysis shows that PI3K-β inhibitor AZD6482 and TGFβRII inhibitor SB505124 may be suitable therapies for high-risk patients,while the IGF-1R inhibitor BMS-754807 or the novel pyrimidine-based anti-tumor metabolic drug Gemcitabine could be potential therapeutic choices for low-risk patients.Moreover,expression of these 3-gene model was verified by immunohistochemistry.In summary,the establishment and validation of a CD8^(+)T cell-derived risk model can more accurately predict the prognosis of HCC patients and guide the construction of personalized treatment plans.展开更多

AIM:Regulatory T cells(Tregs)are a specialized subset of CD4^(+)T cells primarily involved in im⁃munosuppressive functions.AMP-activated protein kinase(AMPK)serves as a metabolic sensor that governs the differen⁃tiati...AIM:Regulatory T cells(Tregs)are a specialized subset of CD4^(+)T cells primarily involved in im⁃munosuppressive functions.AMP-activated protein kinase(AMPK)serves as a metabolic sensor that governs the differen⁃tiation,maturation,and immune functions of Tregs through metabolic reprogramming.However,the impact of AMPKα1(the catalytic subunit of AMPK)knockout specifically in Tregs on the host's immune microenvironment remains largely un⁃explored.METHODS:Histological changes in immune organs were assessed using HE staining.The types of immune cells and their relative population percentages in immune organs and blood were quantified through flow cytometry in both AMPKα1flox/flox(AMPKα1^(fl/fl))mice and Treg-specific AMPKα1 knockout mice(AMPKα1^(fl/fl)Foxp3^(cre)mice).RESULTS:Compared to AMPKα1^(fl/fl)mice,the percentage of eosinophils in the bone marrow of AMPKα1^(fl/fl)Foxp3^(cre)mice was significant⁃ly reduced.Additionally,while the thymus of AMPKα1^(fl/fl)Foxp3^(cre)mice exhibited normal structure,both its size and the ratio of thymus weight to body weight were significantly decreased.The knockout of AMPKα1 in Tregs led to a notable reduction in the total percentage of immature double-negative(DN)cells.Consequently,the percentage of CD4^(+)T cells derived from these DN cells also decreased,even though the percentages of DN1 and DN4 cells were higher in the thymus of AMPKα1^(fl/fl)Foxp3^(cre)mice compared to AMPKα1^(fl/fl)mice.Importantly,the proportion of Siglec-F+CD11b^(+)eosinophils in the thymus was significantly lower in AMPKα1^(fl/fl)Foxp3^(cre)mice.Knockout of AMPKα1 in Tregs resulted in a marked increase in the percentage of CD4^(+)T cells in peripheral blood,alongside a decrease in the proportion of mature CD8^(+)T cells.Similarly,the proportion of CD4^(+)T cells in the spleen of AMPKα1^(fl/fl)Foxp3^(cre)mice was elevated compared to AMPKα1^(fl/fl)mice.In contrast,the proportion of neutrophils significantly decreased,while mononuclear cell proportions increased in the spleen of AMPKα1^(fl/fl)Foxp3^(cre)mice.In lymph nodes,the medullary boundaries in AMPKα1^(fl/fl)Foxp3^(cre)mice were blurred,and the lymphoid follicles were missing,a feature not observed in AMPKα1^(fl/fl)mice.Furthermore,the knockout of AMPKα1 in Tregs reduced the CD3^(+)T cell population,particularly the CD8^(+)T cell population,in lymph nodes.Although the mature Treg cell population was significantly lower in AMPKα1^(fl/fl)Foxp3^(cre)mice,the percentage of CD4^(+)T cells was markedly in⁃creased.In contrast,there was no statistically significant difference in granulocyte populations between AMPKα1^(fl/fl)Foxp3^(cre)and AMPKα1^(fl/fl)mice.CONCLUSION:The populations of mature Tregs,CD8^(+)T cells and eosinophils in various im⁃mune organs were significantly altered in mice with Treg-specific AMPKα1 knockout,suggesting a potential remodeling of the host immune microenvironment in response to inflammatory stimuli.展开更多
Objective:To evaluate the predictive value of the neutrophil⁃to⁃lymphocyte ratio(NLR)and the systemic immune⁃inflammation index(SII)in predicting patients with anti⁃melanoma differentiation⁃associated gene 5⁃positive(...Objective:To evaluate the predictive value of the neutrophil⁃to⁃lymphocyte ratio(NLR)and the systemic immune⁃inflammation index(SII)in predicting patients with anti⁃melanoma differentiation⁃associated gene 5⁃positive(anti⁃MDA5+)dermatomyositis(DM)develop into the rapidly progressive interstitial lung disease(RPILD).Methods:We retrospectively analyzed the clinical and laboratory data of 124 anti⁃MDA5+DM patients from the First Affiliated Hospital of Nanjing Medical University between March 2019 and September 2023.We identified independent risk factors associated with the development and mortality of RPILD with the Cox regression analysis,and determined the optimal cut⁃off values for predicting adverse outcomes with the receiver operating characteristic(ROC)curve analysis.Results:Among the 124 patients,36 patients(29.03%)developed RPILD,and 39 patients(31.45%)died during the follow⁃up period.The results of multivariate Cox regression analysis showed that the elevated NLR was an independent risk factor for RPILD development,while the elevated SII expression was independently associated with the increased mortality of RPILD.Based on the ROC curve analysis,NLR>6.12 was a predictor for RPILD,and SII>875.79 was associated with increased mortality risk of RPILD.Conclusion:Both NLR and SII are accessible,cost⁃effective,and reliable prognostic indicators for the prognosis of patients with anti⁃MDA5^(+)DM,providing a valuable guidance for clinical management and risk stratification of the disease.展开更多
Perilla essential oil(PEO)is reported as an aromatic yellowish oily substance with a volatile odor extracted from perilla leaves.It exhibits various biological activities except anti-tobacco mosaic virus(TMV)activity....Perilla essential oil(PEO)is reported as an aromatic yellowish oily substance with a volatile odor extracted from perilla leaves.It exhibits various biological activities except anti-tobacco mosaic virus(TMV)activity.In this study,we investigated the main components and anti-TMV activity of PEO,identified its primary active components,and examined its mode of action.The results indicated that PEO exhibited anti-TMV activity(65.58%)at 800μg/mL,with perillaldehyde identified as the main active component.The protective,curative,and inactivation activities of perillaldehyde at 800μg/mL were 80.41%,73.42%,and 34.93%,respectively.These values were significantly higher than those of the control drug(commercial chitosan oligosaccharide)and the protective and curative activities were superior to those of ningnanmycin.The results of the mode of action showed that perillaldehyde induced a hypersensitive response(HR)in tobacco.Transmission electron microscope(TEM)observation revealed that perillaldehyde had no direct effect on TMV particles.The treatment of Nicotiana glutinosa with perillaldehyde at 800μg/mL indicated that perillaldehyde had significant induction activity(58.46%).The expression of three pathogenesis-related tobacco genes(PR genes),including nonexpressor of pathogenesis-related genes 1(NPR1),pathogenesis-related protein 1 gene(PR1),and pathogenesis-related protein 5 gene(PR5),were induced and upregulated by perillaldehyde treatment.Perillaldehyde also induced the overexpression of the phenylalanine ammonia-lyase gene(PAL),respiratory burst oxidase homolog B gene(RBOHB),and protochlorophyllide oxidoreductase gene 1(POR1).Furthermore,perillaldehyde increased the salicylic acid(SA)and H2O2 contents in tobacco leaves,and enhanced the activities of four defense enzymes:superoxide dismutase(SOD),catalase(CAT),peroxidase(POD),and phenylalanine ammonia-lyase(PAL).N.glutinosa was treated with perillaldehyde at 800μg/mL for 24 h,and the results showed that the highest SA and H_(2)O_(2) contents(1032.08 pmol/L and 23.40μmol/g FW,respectively)were obtained in tobacco leaves.Defense enzyme activities also reached a maximum at 800μg/mL,and the activities of CAT,PAL,POD,and SOD increased by 1.76,1.95,2.17,and 3.78 times,respectively,compared to the control.The results of the study showed that perillaldehyde may enhance resistance to pathogen infection by inducing systemic acquired resistance(SAR),which may contribute to the activation of SA signal transduction pathway.Therefore,perillaldehyde has the potential for application in agriculture as a novel antiviral agent and immune inducer.展开更多
Objective:Osteosarcoma is a highly aggressive primary malignant bone tumor commonly seen in children and adolescents,with a poor prognosis.Anchorage-dependent cell death(anoikis)has been proven to be indispensable in ...Objective:Osteosarcoma is a highly aggressive primary malignant bone tumor commonly seen in children and adolescents,with a poor prognosis.Anchorage-dependent cell death(anoikis)has been proven to be indispensable in tumor metastasis,regulating the migration and adhesion of tumor cells at the primary site.However,as a type of programmed cell death,anoikis is rarely studied in osteosarcoma,especially in the tumor immune microenvironment.This study aims to clarify prognostic value of anoikis and tumor immune microenvironment-related gene in the treatment of osteosarcoma.Methods:Anoikis-related genes(ANRGs)were obtained from GeneCards.Clinical information and ANRGs expression profiles of osteosarcoma patients were sourced from the therapeutically applicable research to generate effective therapies and Gene Expression Omnibus(GEO)databases.ANRGs highly associated with tumor immune microenvironment were identified by the estimate package and the weighted gene coexpression network analysis(WGCNA)algorithm.Machine learning algorithms were performed to construct long-term survival predictive strategy,each sample was divided into high-risk and low-risk subgroups,which was further verified in the GEO cohort.Finally,based on single-cell RNA-seq from the GEO database,analysis was done on the function of signature genes in the osteosarcoma tumor microenvironment.Results:A total of 51 hub ANRGs closely associated with the tumor microenvironment were identified,from which 3 genes(MERTK,BNIP3,S100A8)were selected to construct the prognostic model.Significant differences in immune cell activation and immune-related signaling pathways were observed between the high-risk and low-risk groups based on tumor microenvironment analysis(all P<0.05).Additionally,characteristic genes within the osteosarcoma microenvironment were identified in regulation of intercellular crosstalk through the GAS6-MERTK signaling pathway.Conclusion:The prognostic model based on ANRGs and tumor microenvironment demonstrate good predictive power and provide more personalized treatment options for patients with osteosarcoma.展开更多

Experiments results show that the algorithm can compute simply and run quickly. It resolves the combinatorial optimization problem of component DMC estimation with simple and available parameters. And it has higher accuracy than individual methods, such as PLS, BP and v-SVM, and also has better performance than other combined methods, such as basic PSO and BP neural network.

文摘To promote the modeling standardization process of the integrated circuits, an improved electrical simulation model for a direct power injection (DPI) setup which was used to measure the conducted immunity of a 16-bit microcontroller to radio frequency aggression was investigated. Based on the existing model of the same microcontroller, the PDN module was modified by adding the core, PLL and MD network models, which could reflect the actual electric distribution situation within the microcontroller more accurately. By comparing the simulation results with the measurement results, the effectiveness of the modified model can be improved to 500 MHz, and its uncertainty is within +1.8 dB (+2 dB is acceptable). Then, to improve the simulation accuracy of the complete model in the high frequency range, the I/O model which contained the dynamic and nonlinear characteristics reflecting the variation of the internal impedance of the microcontroller with increasing the frequency of the external noise was introduced. By comparing the simulation results with the measurement results, the effectiveness of the second modified model can be improved up to 1.4 GHz with the uncertainty of ~1.8 dB. Thus, a conclusion can be reached that the proposed model can be applied to a much wider frequency range with a smaller uncertainty than the latest model of the similar type. Furthermore, associated with the electromagnetic emission testing platform model, the PDN module can also be used to predict the electromagnetic conducted and radiated emission characteristics. This modeling method can also be applied to other integrated circuits, which is very helpful to the standardization of the IC electromagnetic compatibility (EMC) modeling process.

文摘Exogenous alanyl-glutamine(Aln-Gln) was evaluated for its effects on growth performance, intestinal structure and function, antioxidant status and non-specific immunity of young carp(Cyprinus carpio L.). Six diets supplemented with 0, 2.5, 5.0, 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln were fed to fish for 12 weeks. Supplementation with 7.5, 10.0, or 15.0 g · kg-1 of Aln-Gln significantly increased weight gain rate(WGR), protein efficiency ratio(PER), but feed conservation rate(FCR) and survival were not affected(P〉0.05). The intestinal fold height and number, digestive enzyme, Na+, K+-ATPase activities was found to be significantly high(P〈0.05) with increasing dietary Aln-Gln supplementation up to 7.5 g · kg-1, but there were no significant differences for Aln-Gln supplementation from 7.5 to 15.0 g · kg-1. The glutathione peroxidase(GPX) activity, glutathione(GSH), superoxide dismutase(SOD) activity increased and malondialdehyde(MDA) levels decreased significantly(P〈0.05) in the intestine, hepatopancreas, plasma and muscles. The plasma complement-3(C3) and complement-4(C4) levels were significantly(P〈0.05) improved at 5.0 g · kg-1 level and decreased when over 7.5 g · kg-1. The plasma lysozyme(LSZ) activity increased significantly(P〈0.05) at 7.5, 10.0, or 15.0 g · kg-1 level. In summary, the results showed that Aln-Gln improved growth performance, development and function of the intestine, the activity of the antioxidant defense system and the plasma non-specific immunity of the carps. The optimal Aln-Gln level was 8.24 g · kg-1 diet for WGR based on broken-line regression model analysis.

文摘Combining the clonal selection mechanism of the immune system with the evolution equations of particle swarm optimization, an advanced algorithm was introduced for functions optimization. The advantages of this algorithm lies in two aspects. Via immunity operation, the diversity of the antibodies was maintained, and the speed of convergent was improved by using particle swarm evolution equations. Simulation programme and three functions were used to check the effect of the algorithm. The advanced algorithm were compared with clonal selection algorithm and particle swarm algorithm. The results show that this advanced algorithm can converge to the global optimum at a great rate in a given range, the performance of optimization is improved effectively.

文摘The number of birth per year was an important indicator of the sow's productivity and it was related to the duration of breasffeeding. Weaning had been shown to be a stressful period. The weaning age had declined over time in order to increase sow productivity and the fact that weaning under modern-day commercial conditions inflicted stress (environmental, nutritional and psychological/social) on pigs and associated with marked changes in gastrointestinal tract (GIT) physiology, microbiology and immunology. However, the early weaning resulted in post-weaning diarrhea, mainly related to intestinal dysbiosis during this rapid adaptation to the new diet. The environment and feed had influence on physiology, gut morphology, immune system and diseases of weaned piglets.
文摘One hundred and forty-four Avin broilers (1 d old) were randomly divided into six treatments, with three replicates of eight birds each to investigate the effect of different oligosaccharides on index of immune organ, cecal weight and microflora in broilers. The control group (Group I) was fed with corn-soybean meal as basal diet, and the trial groups (group Ⅱ, Ⅲ, Ⅳ, Ⅴ and Ⅵ) were fed with basal diet plus 0.1% manoligosaccharides (MOS), 0.3% soybean-oligosaccharides (SBOS), 0.3% fructooligosaccharides (FOS), 0.3% α-glucooligosaccharides(α-GOS) and 0.03% chlortetracycline (CTC), respectively. The results showed that oligosaccharides supplementation significantly (P< 0.05) increased bifidobacteria concentration of young broilers and did not affect on bifidobacteria or escherichia coli concentration of the birds ( P >0.05). Cecal weights of 21 d old broilers fed with FOS and α-GOS were significantly(P <0.05) increased, higher than those with MOS and SBOS, but addition of oligosaccharides had no effect on the birds. The effect of oligosaccharides on the cecal length was the same as on the cecal weights. Oligosaccharides supplement did not affect on indexes of immune organs.

文摘Hepatocellular carcinoma(HCC),which is essentially primary liver cancer,is closely related to CD8^(+)T cell immune infiltration and immune suppression.We constructed a CD8^(+)T cells related risk score model to predict the prognosis of HCC patients and provided therapeutic guidance based on the risk score.Using integrated bulk RNA sequencing(RNA-seq)and single-cell RNA sequencing(scRNA-seq)datasets,we identified stable CD8^(+)T cell signatures.Based on these signatures,a 3-gene risk score model,comprised of KLRB1,RGS 2,and TNFRSF1B was constructed.The risk score model was well validated through an independent external validation cohort.We divided patients into high-risk and low-risk groups according to the risk score and compared the differences in immune microenvironment between these two groups.Compared with low-risk patients,high-risk patients have higher M2-type macrophage content(P<0.0001)and lower CD8^(+)T cells infiltration(P<0.0001).High-risk patients predict worse response to immunotherapy treatment than low-risk patients(P<0.01).Drug sensitivity analysis shows that PI3K-β inhibitor AZD6482 and TGFβRII inhibitor SB505124 may be suitable therapies for high-risk patients,while the IGF-1R inhibitor BMS-754807 or the novel pyrimidine-based anti-tumor metabolic drug Gemcitabine could be potential therapeutic choices for low-risk patients.Moreover,expression of these 3-gene model was verified by immunohistochemistry.In summary,the establishment and validation of a CD8^(+)T cell-derived risk model can more accurately predict the prognosis of HCC patients and guide the construction of personalized treatment plans.
文摘Objective Ulcerative colitis is a prevalent immunoinflammatory disease.Th17/Treg cell imbalance and gut microbiota dysregulation are key factors in ulcerative colitis pathogenesis.The actin cytoskeleton contributes to regulating the proliferation,differentiation,and migration of Th17 and Treg cells.Wdr63,a gene containing the WD repeat domain,participates in the structure and functional modulation of actin cytoskeleton.Recent research indicates that WDR63 may serve as a regulator of cell migration and metastasis via actin polymerization inhibition.This article aims to explore the effect of Wdr63 deletion on Th17/Treg cells and ulcerative colitis.Methods We constructed Wdr63-/-mice,induced colitis in mice using dextran sulfate sodium salt,collected colon tissue for histopathological staining,collected mesenteric lymph nodes for flow cytometry analysis,and collected healthy mouse feces for microbial diversity detection.Results Compared with wild-type colitis mice,Wdr63-/-colitis mice had a more pronounced shortening of colonic tissue,higher scores on disease activity index and histological damage index,Treg cells decreased and Th17 cells increased in colonic tissue and mesenteric lymph nodes,a lower level of anti-inflammatory cytokine IL-10,and a higher level of pro-inflammatory cytokine IL-17A.In addition,WDR63 has shown positive effects on maintaining intestinal microbiota homeostasis.It maintains the balance of Bacteroidota and Firmicutes,promoting the formation of beneficial intestinal bacteria linked to immune inflammation.Conclusion Wdr63 deletion aggravates ulcerative colitis in mice,WDR63 inhibits colonic inflammation likely by regulating Th17/Treg balance and maintains intestinal microbiota homeostasis.

文摘AIM:Regulatory T cells(Tregs)are a specialized subset of CD4^(+)T cells primarily involved in im⁃munosuppressive functions.AMP-activated protein kinase(AMPK)serves as a metabolic sensor that governs the differen⁃tiation,maturation,and immune functions of Tregs through metabolic reprogramming.However,the impact of AMPKα1(the catalytic subunit of AMPK)knockout specifically in Tregs on the host's immune microenvironment remains largely un⁃explored.METHODS:Histological changes in immune organs were assessed using HE staining.The types of immune cells and their relative population percentages in immune organs and blood were quantified through flow cytometry in both AMPKα1flox/flox(AMPKα1^(fl/fl))mice and Treg-specific AMPKα1 knockout mice(AMPKα1^(fl/fl)Foxp3^(cre)mice).RESULTS:Compared to AMPKα1^(fl/fl)mice,the percentage of eosinophils in the bone marrow of AMPKα1^(fl/fl)Foxp3^(cre)mice was significant⁃ly reduced.Additionally,while the thymus of AMPKα1^(fl/fl)Foxp3^(cre)mice exhibited normal structure,both its size and the ratio of thymus weight to body weight were significantly decreased.The knockout of AMPKα1 in Tregs led to a notable reduction in the total percentage of immature double-negative(DN)cells.Consequently,the percentage of CD4^(+)T cells derived from these DN cells also decreased,even though the percentages of DN1 and DN4 cells were higher in the thymus of AMPKα1^(fl/fl)Foxp3^(cre)mice compared to AMPKα1^(fl/fl)mice.Importantly,the proportion of Siglec-F+CD11b^(+)eosinophils in the thymus was significantly lower in AMPKα1^(fl/fl)Foxp3^(cre)mice.Knockout of AMPKα1 in Tregs resulted in a marked increase in the percentage of CD4^(+)T cells in peripheral blood,alongside a decrease in the proportion of mature CD8^(+)T cells.Similarly,the proportion of CD4^(+)T cells in the spleen of AMPKα1^(fl/fl)Foxp3^(cre)mice was elevated compared to AMPKα1^(fl/fl)mice.In contrast,the proportion of neutrophils significantly decreased,while mononuclear cell proportions increased in the spleen of AMPKα1^(fl/fl)Foxp3^(cre)mice.In lymph nodes,the medullary boundaries in AMPKα1^(fl/fl)Foxp3^(cre)mice were blurred,and the lymphoid follicles were missing,a feature not observed in AMPKα1^(fl/fl)mice.Furthermore,the knockout of AMPKα1 in Tregs reduced the CD3^(+)T cell population,particularly the CD8^(+)T cell population,in lymph nodes.Although the mature Treg cell population was significantly lower in AMPKα1^(fl/fl)Foxp3^(cre)mice,the percentage of CD4^(+)T cells was markedly in⁃creased.In contrast,there was no statistically significant difference in granulocyte populations between AMPKα1^(fl/fl)Foxp3^(cre)and AMPKα1^(fl/fl)mice.CONCLUSION:The populations of mature Tregs,CD8^(+)T cells and eosinophils in various im⁃mune organs were significantly altered in mice with Treg-specific AMPKα1 knockout,suggesting a potential remodeling of the host immune microenvironment in response to inflammatory stimuli.
文摘Objective:To evaluate the predictive value of the neutrophil⁃to⁃lymphocyte ratio(NLR)and the systemic immune⁃inflammation index(SII)in predicting patients with anti⁃melanoma differentiation⁃associated gene 5⁃positive(anti⁃MDA5+)dermatomyositis(DM)develop into the rapidly progressive interstitial lung disease(RPILD).Methods:We retrospectively analyzed the clinical and laboratory data of 124 anti⁃MDA5+DM patients from the First Affiliated Hospital of Nanjing Medical University between March 2019 and September 2023.We identified independent risk factors associated with the development and mortality of RPILD with the Cox regression analysis,and determined the optimal cut⁃off values for predicting adverse outcomes with the receiver operating characteristic(ROC)curve analysis.Results:Among the 124 patients,36 patients(29.03%)developed RPILD,and 39 patients(31.45%)died during the follow⁃up period.The results of multivariate Cox regression analysis showed that the elevated NLR was an independent risk factor for RPILD development,while the elevated SII expression was independently associated with the increased mortality of RPILD.Based on the ROC curve analysis,NLR>6.12 was a predictor for RPILD,and SII>875.79 was associated with increased mortality risk of RPILD.Conclusion:Both NLR and SII are accessible,cost⁃effective,and reliable prognostic indicators for the prognosis of patients with anti⁃MDA5^(+)DM,providing a valuable guidance for clinical management and risk stratification of the disease.

文摘Perilla essential oil(PEO)is reported as an aromatic yellowish oily substance with a volatile odor extracted from perilla leaves.It exhibits various biological activities except anti-tobacco mosaic virus(TMV)activity.In this study,we investigated the main components and anti-TMV activity of PEO,identified its primary active components,and examined its mode of action.The results indicated that PEO exhibited anti-TMV activity(65.58%)at 800μg/mL,with perillaldehyde identified as the main active component.The protective,curative,and inactivation activities of perillaldehyde at 800μg/mL were 80.41%,73.42%,and 34.93%,respectively.These values were significantly higher than those of the control drug(commercial chitosan oligosaccharide)and the protective and curative activities were superior to those of ningnanmycin.The results of the mode of action showed that perillaldehyde induced a hypersensitive response(HR)in tobacco.Transmission electron microscope(TEM)observation revealed that perillaldehyde had no direct effect on TMV particles.The treatment of Nicotiana glutinosa with perillaldehyde at 800μg/mL indicated that perillaldehyde had significant induction activity(58.46%).The expression of three pathogenesis-related tobacco genes(PR genes),including nonexpressor of pathogenesis-related genes 1(NPR1),pathogenesis-related protein 1 gene(PR1),and pathogenesis-related protein 5 gene(PR5),were induced and upregulated by perillaldehyde treatment.Perillaldehyde also induced the overexpression of the phenylalanine ammonia-lyase gene(PAL),respiratory burst oxidase homolog B gene(RBOHB),and protochlorophyllide oxidoreductase gene 1(POR1).Furthermore,perillaldehyde increased the salicylic acid(SA)and H2O2 contents in tobacco leaves,and enhanced the activities of four defense enzymes:superoxide dismutase(SOD),catalase(CAT),peroxidase(POD),and phenylalanine ammonia-lyase(PAL).N.glutinosa was treated with perillaldehyde at 800μg/mL for 24 h,and the results showed that the highest SA and H_(2)O_(2) contents(1032.08 pmol/L and 23.40μmol/g FW,respectively)were obtained in tobacco leaves.Defense enzyme activities also reached a maximum at 800μg/mL,and the activities of CAT,PAL,POD,and SOD increased by 1.76,1.95,2.17,and 3.78 times,respectively,compared to the control.The results of the study showed that perillaldehyde may enhance resistance to pathogen infection by inducing systemic acquired resistance(SAR),which may contribute to the activation of SA signal transduction pathway.Therefore,perillaldehyde has the potential for application in agriculture as a novel antiviral agent and immune inducer.

文摘Objective:Osteosarcoma is a highly aggressive primary malignant bone tumor commonly seen in children and adolescents,with a poor prognosis.Anchorage-dependent cell death(anoikis)has been proven to be indispensable in tumor metastasis,regulating the migration and adhesion of tumor cells at the primary site.However,as a type of programmed cell death,anoikis is rarely studied in osteosarcoma,especially in the tumor immune microenvironment.This study aims to clarify prognostic value of anoikis and tumor immune microenvironment-related gene in the treatment of osteosarcoma.Methods:Anoikis-related genes(ANRGs)were obtained from GeneCards.Clinical information and ANRGs expression profiles of osteosarcoma patients were sourced from the therapeutically applicable research to generate effective therapies and Gene Expression Omnibus(GEO)databases.ANRGs highly associated with tumor immune microenvironment were identified by the estimate package and the weighted gene coexpression network analysis(WGCNA)algorithm.Machine learning algorithms were performed to construct long-term survival predictive strategy,each sample was divided into high-risk and low-risk subgroups,which was further verified in the GEO cohort.Finally,based on single-cell RNA-seq from the GEO database,analysis was done on the function of signature genes in the osteosarcoma tumor microenvironment.Results:A total of 51 hub ANRGs closely associated with the tumor microenvironment were identified,from which 3 genes(MERTK,BNIP3,S100A8)were selected to construct the prognostic model.Significant differences in immune cell activation and immune-related signaling pathways were observed between the high-risk and low-risk groups based on tumor microenvironment analysis(all P<0.05).Additionally,characteristic genes within the osteosarcoma microenvironment were identified in regulation of intercellular crosstalk through the GAS6-MERTK signaling pathway.Conclusion:The prognostic model based on ANRGs and tumor microenvironment demonstrate good predictive power and provide more personalized treatment options for patients with osteosarcoma.
