Von Willebrand Factor(VWF)is a concatameric glycoprotein that plays a key role in rapid hemostasis and thrombosis.VWF has different functional domains that can bind to various molecules such as collagen,hemostatic factorⅧ,integrin,and platelet glycoprotein lbα(GPlbα)to achieve multiple biological functions.During hemostasis,the A1 domain of VWF binds to GPIbαwhere platelets accumulate in the injured vascular endothelium.Due to forces generated by the hemodynamic gradient flow,the relations of bond-dissociation rates versus forces show that the lifetime of molecular bond has multiple states under the external force.We processed the experimental data of receptor-ligand in a single molecule obtained from optical tweezers by two different methods,including a Dudko-Hummer-Szabo equation,and another method combining force4ime history and force induced bond rupture.Then we used a recently developed physical equation regarding protein unfolding rate to fit our results.The lifetime of the bond between A1 and GPlbαobtained by the above mentioned two methods shows a'three-stage'change upon gradually increasing the external force.When the external force was below 8 pN,the lifetime of the bond deceased as the external force increased,which is a typical expression of a catch bond.The lifetime of the bond started to increase when the external force increased from 8 to 11 pN,and then decrease again when the external force increased to above 11 pN.Kim et al.used different processing methods and proposes a'flex-bond'model:the lifetime of the bond will decrease as the external force increases,then suddenly increase to a peak,and continue to decrease with the increase of force.A recently developed model based on the structural-elastic properties of molecules fits our data well,indicating that the bond formed by Al and GPlbαhas a catch-bond phenomenon in a certain interval of external forces,and a flex bond in other force intervals.In conclusion,A1-GPIbαbond will have a'slip-catch-slip'bond tendency.Our result provides a alternative understanding about the role of Al-GPlbαinteractions in the mechanism of hemostasis. 展开更多

A new approach is proposed to analyze the settlement behavior for single pile embedded in layered soils. Firstly, soil layers surrounding pile shaft are simulated by using distributed Voigt model, and finite soil layers under the pile end are assumed to be virtual soil-pile whose cross-section area is the same as that of the pile shaft. Then, by means of Laplace transform and impedance function transfer method to solve the static equilibrium equation of pile, the analytical solution of the displacement impedance fimction at the pile head is derived. Furthermore, the analytical solution of the settlement at the head of single pile is theoretically derived by virtue of convolution theorem. Based on these solutions, the influences of parameters of soil-pile system on the settlement behavior for single pile are analyzed. Also, comparison of the load-settlement response for two well-instrumented field tests in multilayered soils is given to demonstrate the effectiveness and accuracy of the proposed approach. It can be noted that the presented solution can be used to calculate the settlement of single pile for the preliminary design of pile foundation. 展开更多

Single grit grinding is the simplified model to abstract the macro scale grinding.Finite element analysis is a strong tool to study the physical fields during a single grit grinding process,compared to experimental research.Based on the dynamic mechanical behavior of 2Cr12Ni4Mo3VNbN steel and the mathematical statistics of abrasive grit,modeling of the single grit grinding process was conducted by using commercial software AdvantEdge.The validation experiment was designed to validate the correctness of the FEA model by contrast with grinding force.The validation result shows that the FEA model can well describe the single grit grinding process.Then the grinding force and multi-physics fields were studied by experimental and simulation results.It was found that both the normal and tangential grinding forces were linearly related to the cutting speed and cutting depth.The maximum temperature is located in the subsurface of the workpiece in front of the grit,while the maximum stress and strain are located under the grit tip.The strain rate can reach as high as about 106 s–1 during the single grit grinding,which is larger than other traditional machining operations. 展开更多

Decreasing in emissions of greenhouse gases to confront the global warming needs to replace fossil fuels as the main doer of the world climate changes by renewable and clean fuels produced from biomass like wood waste which is neutral on the amount of CO2. An analytical and engineering model for pyrolysis process of a single biomass particle has been presented. Using a two-stage semi global kinetic model which includes both primary and secondary reactions, the effects of parameters like shape and size of particle as well as porosity on the particle temperature profile and product yields have been investigated. Comparison of the obtained results with experimental data shows that our results are in a reasonable agreement with previous researchers' works. Finally, a sensitivity analysis is done to determine the importance of each parameter on pyrolysis of a single biomass particle which is affected by many constant parameters. 展开更多

A series of TL rectifiers are derived by using double TL and multi TL methods, and their characteristics are compared at the same time. A novel seven-switch double TL rectifier is constructed, and its operational principle and control algorithm are illustrated as well. Objective Triple-negative breast cancer(TNBC)is the breast cancer subtype with the worst prognosis,and lacks effective therapeutic targets.Colony stimulating factors(CSFs)are cytokines that can regulate the production of blood cells and stimulate the growth and development of immune cells,playing an important role in the malignant progression of TNBC.This article aims to construct a novel prognostic model based on the expression of colony stimulating factors-related genes(CRGs),and analyze the sensitivity of TNBC patients to immunotherapy and drug therapy.Methods We downloaded CRGs from public databases and screened for differentially expressed CRGs between normal and TNBC tissues in the TCGA-BRCA database.Through LASSO Cox regression analysis,we constructed a prognostic model and stratified TNBC patients into high-risk and low-risk groups based on the colony stimulating factors-related genes risk score(CRRS).We further analyzed the correlation between CRRS and patient prognosis,clinical features,tumor microenvironment(TME)in both high-risk and low-risk groups,and evaluated the relationship between CRRS and sensitivity to immunotherapy and drug therapy.Results We identified 842 differentially expressed CRGs in breast cancer tissues of TNBC patients and selected 13 CRGs for constructing the prognostic model.Kaplan-Meier survival curves,time-dependent receiver operating characteristic curves,and other analyses confirmed that TNBC patients with high CRRS had shorter overall survival,and the predictive ability of CRRS prognostic model was further validated using the GEO dataset.Nomogram combining clinical features confirmed that CRRS was an independent factor for the prognosis of TNBC patients.Moreover,patients in the high-risk group had lower levels of immune infiltration in the TME and were sensitive to chemotherapeutic drugs such as 5-fluorouracil,ipatasertib,and paclitaxel.Conclusion We have developed a CRRS-based prognostic model composed of 13 differentially expressed CRGs,which may serve as a useful tool for predicting the prognosis of TNBC patients and guiding clinical treatment.Moreover,the key genes within this model may represent potential molecular targets for future therapies of TNBC. 展开更多

Study on the phytoplankton community structure and its driving factors in Furong Creek during spring and autumn under the influence of cascade weirs

[Objective]The construction of weirs changes the hydraulic characteristics of rivers and affects the structure of phytoplankton communities and the health of aquatic ecosystems in the river.This study aims to explore the nonlinear response relationship between phytoplankton community structure and its driving factors in spring and autumn in Furong Creek under the construction of cascade weirs.[Methods]The structure of phytoplankton communities and related environmental factors were investigated in Furong Creek from 2023 to 2024.This study focused on the analysis of the changes of nutrient concentrations and biomass of phytoplankton in autumn and spring within the same dry season in Furong Creek.Redundancy analysis was used to identify the key factors influencing the structure of phytoplankton communities.The MIKE 11 model was employed to simulate the hydrodynamic changes in the river.Combined with total nitrogen and permanganate index,a GAM model of phytoplankton diversity index and hydrodynamic factors was developed,and the change of phytoplankton diversity after the optimized layout of the cascade weirs was fitted.[Results]The result showed that the annual average value of Shannon-Wiener diversity index of phytoplankton in Furong Creek was 2.79,which was in a state of mild pollution.A total of 239 species from 95 genera in 8 phyla were identified.Among the phytoplankton,Chlorophyta was the dominant group throughout the year in Furong Creek,followed by Bacillariophyta and Cyanophyta.The cell abundance of phytoplankton ranged from 3.11 to 20.64 mg/L and from 0.23 to 6.31 mg/L in spring and autumn,which indicated a clear seasonal succession of phytoplankton community structure.Compared with autumn,the relative abundance of Cyanophyta significantly decreased in spring across the whole river section,while Chrysophyta and Dinophyta showed significant increase at some monitoring sites,leading to water bloom phenomenon and a noticeable decline in the diversity of phytoplankton.The dominant species in the water bodies throughout the year were Cyclotella catenata,Chlorella vulgaris,Scenedesmus bijuga,Scenedesmus quadricauda,Chroomonas acuta,Cryptomonas ovata,and Cryptomonas erosa.Redundancy analysis(RDA)showed that hydrodynamic factors(v,h)and water environmental factors(TN,COD_(Mn))were the main influencing factors of phytoplankton community structure.[Conclusion]The result show that the nutrient concentration,phytoplankton biomass,and density in Furong Creek in spring are significantly higher than in autumn.The GAM model,constructed by combining hydrodynamic and environmental factors,can effectively reflect the nonlinear relationship between phytoplankton diversity index and its driving factors.In spring,with an increase in nutrient concentration,the habitat conditions of low flow speed and high water depths formed by overflow weirs will lead to a decrease in the Shannon-Wiener index of phytoplankton and an intensified risk of eutrophication.However,a reasonable layout scheme of cascade weirs will improve the diversity of phytoplankton and reduce the risk of eutrophication in the river.The findings of this study can help deepen the understanding of the ecological and environmental effects of cascade weir construction in the river. 展开更多

The traditional mathematical modelling is not available for complex structure converters since the differential equation order increases very high. We have to search other way to establish mathematical modelling for power DC/DC converters.We have theoretically defined a new concept-Energy Factor (EF) in this paper and researched the relations between EF and the mathematical modelling for power DC/DC converters. EF is a new concept in power DC/DC conversion technology, which thoroughly differs from the traditional concepts such as power factor (PF), power transfer efficiency (η), total harmonic distortion (THD) and ripple factor (RF). EF and the subsequential EFV (and EFVD) can illustrate the system stability, reference response and interference recovery. This investigation is very helpful for system design and DC/DC converters characteristics foreseeing. Two DC/DC converters: Buck converter and Super-Lift Luo-Converter as the samples are analysed in this paper to demonstrate the applications of EF, EFV (and EFVD), PE, SE, VE (and VED), time constant τ and damping time constant τd. 展开更多

为了使HFACS这种事故分析方法能够在我国各种类型事故中得到应用,研究了HFACS的不安全监管因素中各指标的定义和分类,并将其与事故致因24 Model中的不安全动作因素进行对比,得出两者的对应关系和各自特点。结果表明:HFACS中多数不安全... 为了使HFACS这种事故分析方法能够在我国各种类型事故中得到应用,研究了HFACS的不安全监管因素中各指标的定义和分类,并将其与事故致因24 Model中的不安全动作因素进行对比,得出两者的对应关系和各自特点。结果表明:HFACS中多数不安全监管因素属于24 Model中的不安全动作,这些不安全动作的发出者都是监管者;24 Model中不安全动作的发出者既可以是事故的直接引发者,也可以是监管者。从监管的范畴来看,HFACS中不安全监管因素的范围小于24 Model中的监管范围,只包括组织内部的监管。 展开更多
