摘要
用灰木相思茎段腋芽为外植体,采用在木本植物茎叶组培材料上经常采用的3种消毒方法及前人已在相思类植物营养器官组培上取得成功的3种萌芽培养基、芽增殖培养基和根诱导培养基进行对比试验,结果表明,以1/800灭菌净10min+75%酒精50s+0.1%HgCl25min的消毒效果最好,以改良MS+6-BA1.0+NAA0.2的萌芽效果最好,以MS+6-BA2.0+IBA0.2的增殖效果最好,以改良MS+IBA1.5+NAA0.4生根效果最好。
In a comparative study, stem segments with axillary buds of Acacia implexa were used as explants in tissue culture. The best disinfection effect was achieved by 1/800 germicide for 10 min + 75% alcohol for 50 s + 0.1% HgCl_2 for 5 min. The best medium germination was a modified MS medium supplemented with 6-BA 1.0 mg/L and NAA 0.2 mg/L; the best medium for proliferation was MS + 6-BA 2.0 mg/L + IBA 0.2 mg/L and the best medium for rooting was a modified MS supplemented with IBA 1.5 mg/L and NAA (0.4) mg/L
