摘要
目的:比较角质形成细胞无血清培养基(KC-SFM)和含血清的黑素细胞培养基培养的人类黑素细胞的生物学特征的差异,寻找适合黑素细胞移植治疗的黑素细胞培养方法。方法:以KC-SFM和含血清的黑素细胞培养基培养黑素细胞,在培养7d内倒置显微镜观察黑素细胞形态,MTT法观察生长曲线,并测定酪氨酸酶活性和黑素含量。结果:KC-SFM培养的黑素细胞具有较多树突且树突明显延长,增殖活性低于含血清的黑素细胞培养基培养的黑素细胞,而酪氨酸酶活性、黑素含量测定吸光值分别为0.385±0.036、0.276±0.022,和含血清的黑素细胞培养基培养黑素细胞比较均有显著差异。结论:KC-SFM可促进黑素细胞树突增多和树突延长,提高酪氨酸酶活性和黑素含量,适合用于移植治疗的黑素细胞培养。
Objective To compare the different biolog ic characteris tics of human melanocytes cultured with ker-atinocyte s erum-free medium(KC-SFM)and s erum-contained melanocyte medium and to s earch for a kind of better method to culture melanocytes for cell trans plantation.Metho ds Melanocytes is olated with routine method were cultured both in KC-SFM and s erum-contained medium,res pectively.Morpholog y were obs erved by invert micros cope,viability and g rowth were detected by MTT reduction as s ay,activity of tyros inas e and s ynthes es of melanin were evaluated during7d of culture.Res ults Compared to cultured in s erum-contained medium,the melanocyte cultured in SK-FSM were lower proliferated activity and had more long er dendrite,while there were s ig nificant differences of the values of activities of tyros inas e and content of melanin,were0.385±0.036and0.276±0.022res pectively.Co nclus io n The KC-SFM can increas e the numbers and leng th of dendrite of melanocyte as well as ris e the activity of activities of tyros inas e and content of melanin.It may be applied in the culture of melanocyte for cell trans plantation.
