摘要
利用inter-重复序列引物扩增DNA多态性(ISSR-PCR)方法分析了6个产棉区棉铃虫间基因流动情况。筛选后的15个引物共扩增出138条DNA条带,其中多态性的条带为125条,占90.6%。6个种群间的平均遗传距离为0.522 1 0.093 6 (SD),平均遗传相似系数0.595 6 0.054 5 (SD)。结果表明,今年不同地理种群间有较弱的基因流。聚类结果显示,河南安阳、江西九江、湖北武汉、安徽阜阳四种群间曾有过较强的基因流,而江苏南京种群表现出较大的差异性,新疆库尔勒种群次之。
Population gene flow of Helicoverpa armigera Hubner over six major cotton growing regions were studied by ISSR-PCR. 15 primers were screened, and total 138 DNA bands were amplified, 125 of which (90.6%) were polymorphic. Among Anyang, Kuerle, Wuhan, Nanjing, Jiujiang and Fuyang populations, their mean genetic distance was 0.522 1 0.093 6 (SD) and mean genetic similarity coefficient was 0.595 6 0.054 5 (SD). It showed that there didnt exit high gene flow among these investigated populations this year. Cluster analysis indicated that there was once strong gene flow among Anyang, Jiujiang, Wuhan,and Fuyang populations, but Nanjing population had better diversity than Kuerle population with others.
