摘要
目的 反相高效液相色谱法(RP—HPLC)测定人血浆中盐酸二甲双胍的浓度。方法 采用色谱柱为Hypersil C18柱,流动相为甲醇-水-三乙胺(75:25:1),流速为1.0ml/min^(-1),检测波长为233nm,内标为吗啉双胍。结果 盐酸二甲双胍的线性范围为0.05~4.00μg/ml,回归方程为Y=0.580C+0.003 5(r=0.999 3);日内精密度为2.28%~8.16%,日间精密度为0.62%~8.68%。结论 本方法操作简便、快速;测定结果准确,重现性好。
Objective To establish a RP--HPLC method for the determination of metformin in human plasma. Methods
The Hypersil C18 column was used. The mobile phase included methanol, water and triethylamine (75: 25: 1), and the
flowrate was 1. 0 ml/min^(-1). The detection wavelength was 233 nm. Internal standard was moroxydine. Results The linear
range of the calibration was 0. 05~4. 00 μg/ml (Y = 0. 580C+0. 003 5, r = 0. 999 3). Within--day RSD was 2. 28%~8. 16%
Between--day RSD was 0. 62%~8. 68%. Conclusion The method is simple and accurate.
