分光光度法测定总氮含量——改进微量凯氏定氮法

摘要 各种天然有机物总氮含量通常采用经典的微量凯氏定氮法测定。本法对凯氏定氮法中水蒸汽蒸馏滴定步骤进行了改进,利用茚三酮与氨的显色反应,在570毫微米处进行比色测定,工作曲线在0.4288~3.814微克范围内,具有良好的线性关系。与经典微量凯氏定氮法相比,本法具有操作简单、干挠因素少、快速等特点。 The micro-Kjeldahl method is usually used for determination of the total Nitrogen content of various nature organic substances.This paper presents a method to improve the steam-generation and titration of Kje-Idahl method.It used the ninhydrin reaction between ninhydrin and am-monia carrying the spectrographic determination of total Nitrogen content at 570 nm.The working curve had a good linear relationship between 0.4288-3.814μg.Comparing with classical micro Kjeldahl method this method had characteristics of operating simply less interference factor and fast-analysis.
