网织红细胞胞质因子调控肿瘤细胞基因表达的核酸杂交分析 Ⅰ.β-珠蛋白基因的表达 被引量:3

摘要 本实验采用核酸杂交技术,以小鼠和人β-珠蛋白基因为探针,分析了同种和异种细胞的胞质体杂交细胞中β-珠蛋白基因的表达状态。结果发现小鼠β-珠蛋白基因转录物不但在同种杂交(小鼠网织红细胞RM×小鼠骨髓瘤BW)的胞质体杂交细胞(BW-RM)中出现,而且在兔网织红细胞与小鼠骨髓瘤的异种杂交细胞(Bw-RR)中亦有表达。同样在小鼠网织红细胞与人浆细胞瘤(HMy)的异种胞质杂交细胞(HMy-RM)中出现人β-珠蛋白基因产物。说明网织红细胞胞质因子可激活原来处于不活动状态的肿瘤细胞β-珠蛋白基因并使之表达.兔和小鼠的胞质因子有同样的调节基因表达的作用而似无种属特异性。对网织红细胞胞质因子调控基因表达和肿瘤细胞恶性逆转的机理和分子生物学基础进行了讨论。 The present study was designed to analyse β-globin gene expression in homocellular and heterocellular cybrids by nucleic acid hybridization technique, with cloned mouse and human β-globin gene as probes. Results indicated that the mouse β-globin gene transcripts were synthsized in both homocellular cybrid subcultures of mouse myeloma BW × mouse reticulo-cyte (BW-RM6) and heterocellular cybrid subcultures of rabbit reticulo-cyte × mouse myeloma (BW-RR6) as well as human β-glodin gene transcripts expressed in heterocellular cybrid subcultures of mouse reticulocyte × human plasmocyte (HMy-RM3) . These results demonstrated that the mouse and rabbit reticulocyte cytoplasmic factors could activate expression of originally inactive β-globin gene of the tumor cells, and their effects seem ed to be without species specificity. Mechamism of regulatory effect of reticulocyte cytoplaismic factors and the molecular basis for gene expression and malignancy reversion of tumor cells were discussed.
