中波紫外线诱导的提前衰老成纤维细胞培养液对正常人真皮成纤维细胞的氧化损害 被引量:4

Oxidative Damage Effects of Conditioned Medium of Premature Senescent Fibroblasts Induced by UVB on Normal Human Dermal Fibroblasts

摘要 目的观察细胞中波紫外线(Ultraviolet b,UVB)诱导的提前衰老成纤维细胞培养液对正常人真皮成纤维细胞的影响。方法将UVB诱导的提前衰老成纤维细胞培养液孵育正常人真皮成纤维细胞,随后使用CCK-8法检测细胞增殖活性,并利用荧光显微镜和流式细胞仪检测细胞中活性氧及线粒体膜电位的变化。结果经过UVB诱导的提前衰老成纤维细胞培养液孵育的正常人真皮成纤维细胞活性显著低于对照组,并且细胞中的活性氧及线粒体膜电位荧光强度也均高于对照组。上述现象可被抗氧化剂所逆转。结论 UVB诱导的提前衰老成纤维细胞可通过释放可溶性活性物质,对周围正常成纤维细胞造成氧化损害。 Objective To observe the effects of conditioned medium of premature senescent fibroblasts induced by UVB rays on normal human dermal fibroblasts. Methods The normal human dermal fibroblasts were incubated by the conditioned medium of premature senescent fibroblasts which induced by UVB, and then CCK-8 was used to observe the ability of proliferation of human dermal fibroblasts. Fluorescence microscope and flow cytometry were used to observe the change of reactive oxygen species and mitochondrial membrane of human dermal fibroblasts. Results The ability of proliferation of fibroblasts which incubated by the conditioned medium of photoaging fibroblasts was significantly lower than the fibroblasts which incubated by the conditioned medium of normal fibroblasts. The reactive oxygen and mitochondrial membrane of fibroblasts which incubated by the conditioned medium of photoaging fibroblasts significantly higher than the control group.And phenomenons were observed can be reversed by antioxidants. Conclusion The premature senescent fibroblasts induced by UVB rays can secrete soluble active substances, and they could cause oxidative damage to the surrounding normal fibroblasts.
