复合免疫应激对肉鸡免疫功能和肝脏脂代谢的影响及其调控 被引量:5

Effects of compound immune stress on immune function and liver lipid metabolism in broilers

摘要 旨在评估免疫应激状态下AA肉鸡新城疫疫苗免疫效果,观察其对免疫功能和肝脏脂代谢的影响以及抗应激剂的调控效果。选取160只AA肉仔鸡随机分为A、B、C、D、E等5个组,每组32只。A组为对照组,B、C组为250μg/kg细菌脂多糖(LPS)处理组,D、E组为500μg/kg LPS处理组;其中,在C、E组饲料中添加1%的抗应激剂。于12日龄起每隔3 d连续腹腔注射LPS共4次,于15,28 d各组分别接种鸡新城疫(ND)Ⅳ系苗,并于22,29,36,42 d将肉鸡称重、宰杀,检测抗体水平、免疫器官指数、肝脏胆固醇(TC)和甘油三酯(TG)含量及其脂代谢相关基因mRNA表达。结果显示,与A组相比,D组能够显著增加法氏囊、胸腺和脾脏等免疫器官指数(P<0.05),显著提高22 d ND疫苗抗体水平(P<0.05)以及29 d肝脏TG含量(P<0.05);B组能够显著增加脾脏指数(P<0.05),显著提高22 d肝脏TC含量(P<0.05)和29,42 d TG含量(P<0.05);而2组免疫应激组均能显著上调22 d(P<0.05)、下调22~42 d HMGR mRNA的表达量(P<0.05),上调29,42 d ACC mRNA的表达量(P<0.05),下调CPT-1和PPAR-αmRNA的表达量(P<0.05)。添加1%的抗应激剂,与空白组相比,脾脏指数显著提高(P<0.05);与免疫应激组相比,除了能显著提高36 d的脾脏指数外,法氏囊、胸腺指数差异不显著(P>0.05),能够显著提高抗体水平(P<0.05),降低42 d肝脏TC的含量(P<0.05)和29,42 d TG的含量(P<0.05),下调ACC mRNA表达量(P<0.05)和42 d HMGR mRNA表达量(P<0.05),上调CPT-1和PPAR-αmRNA表达量(P<0.05)。结果表明,适度的免疫应激能够提高免疫器官指数和新城疫疫苗抗体水平;免疫应激有增加肝脏TC和TG的含量和脂肪肝的风险,其机理可能是通过调控HMGR、ACC、CPT-1和PPAR-αmRNA的表达量,促进胆固醇和脂肪酸的生成,抑制脂肪酸的分解;日粮中添加1%的抗应激剂能增强肉鸡体液免疫,改善肝脏脂代谢。 The effect of immune stress on immune function,hepatic lipid metabolism,and the regulation of anti-stress agents under vaccination with Newcastle disease vaccine in broilers,was assessed.A total of 160 broilers were randomly assigned to 5 groups:group A served as the control,group B and C received 250μg/kg LPS,group D and E received 500μg/kg LPS,1%anti-stress agent was fed in group C and E.All the groups were vaccinated with live LaSota ND vaccine on 15 and 28 d.The LPS was intraperitoneal injected on 12,15,18 and 21 d,and the chickens were weighed and slaughtered on 22,29,36 and 42 d.The immune organ index was calculated,the antibody level of Newcastle disease vaccine was detected,and the levels of hepatic cholesterol(TC)and triglyceride(TG)and mRNA expression of lipid metabolism related genes were detected.Compared with group A,the index of immune organs,the antibody level against ND vaccine on22 dand the content of hepatic TG on 29 dwere significantly increased in group D(P<0.05).In group B,the spleen index was increased(P<0.05),and the hepatic TC content on 22 dand TG content on 29 and 42 dwere increased(P<0.05).The mRNA expression level of HMGR on 22-42 d,ACC mRNA expression level on 29 and 42 d,and the mRNA expression levels of CPT-1 and PPAR-α,were significantly up-regulated in group B and D(P<0.05).The spleen index was increased when 1%anti-stress agent was added in the diet(P<0.05).Compared with the immune stress group,no effect was observed in thymus and bursa index in anti-stress agent group(P>0.05),except that the spleen index was significantly improved on 36 d.It was significantly increased in the antibody level as compared with the control(P<0.05).It was remarkably reduced in the content of hepatic TC on 42 dand TG on 29 and 42 d(P<0.05),significantly down-regulated in the expression level of ACC,HMGR mRNA on 42 d(P<0.05),and increased markedly in the expression level of CPT-1 and PPAR-αmRNA in the anti-stress groups(P<0.05).In conclusion,appropriate immune stress could improve immune organ index and antibody titer against ND vaccine.Immune stress could increase the contents of hepatic TC and TG,and the risk of develop fatty liver disease of broilers,and its mechanism might be to promote the generation of cholesterol and fatty acids and inhibit the decomposition of fatty acids by regulating the expression of HMGR,ACC,CPT-1 and PPAR-αmRNA.The addition of 1%anti-stress agent in the diet could enhance the humoral immunity of broilers and improve hepatic lipid metabolism.
