摘要
目的 观察层粘连蛋白和表皮生长因子单独及联合应用对兔角膜内皮细胞周期的影响。方法 体外培养角膜内皮细胞分为对照组、层粘连蛋白组、表皮生长因子 (EGF)组和联合应用组 ,倒置显微镜观察各组细胞形态及数目变化 ;计数 0、12、2 4、36h的各组细胞数目并绘制细胞生长曲线 ;流式细胞技术检测各组细胞周期。结果 倒置显微镜观察及细胞生长曲线显示 ,与对照组相比 ,层粘连蛋白组、EGF组和联合应用组细胞数目明显增多 ,其中联合应用组最为显著 ;细胞周期检测结果显示 ,层粘连蛋白组、EGF组和联合应用组G0~G1期细胞比例显著下降 (P <0 .0 5 ) ,G2~M期细胞比例显著提高 (P <0 .0 5 ) ;EGF组和联合应用组S期细胞比例显著提高 (P <0 .0 5 ) ;联合应用组与层粘连蛋白组相比 ,G0~G1期细胞比例显著下降 (P <0 .0 5 )。结论 层粘连蛋白和表皮生长因子单独及联合应用都可明显促进角膜内皮细胞分裂增生 ,且联合应用有协同作用。
Objective To investigate the relative effect of single and multiple applications of laminin and recombinant human epidermal growth factor (rhEGF) on cell cycle of cultured rabbit corneal endothelial cells.Methods The cultured endothelial cells were devided into four groups:control group,laminin(LN) group,rhEGF group and multiple applicated group.The changes of morphology and cell number was surveyed with the inverted microscope; Counting cell numbers on cultured 0 hour,12,24 and 36 hours obtained cell growth curve;The changes of cell cycle were examined by flow cytometry.Results As shown by the inverted microscope and the cell growth curve, comparing with the control group, cells of each test group increased evidently, especially the multiple applicated group. From the results of the cell cycle examination, the portion of cells in G0~G1 phase of LN group,rhEGF group and multiple applicated group decreased ( P <0.05) and increased evidently in G2~M phase ( P <0.05);The change of the portion of the cells in phase S was significant in rhEGF group and multiple applicated group ( P < 0.05 ).Furthermore,compared with multiple applicated group and LN group, the portion of cells in G0~G1 phase decreased markedly ( P <0.05).Conclusion Single and multiple applications of LN and rhEGF could prompt the corneal endothelial cells proliferation and division marketedly.
