摘要
目的探讨醌茜素对肺腺癌细胞株A549迁移和侵袭能力的影响。方法体外培养人肺癌A549细胞,分别用0、1、3、10、30μmol/L浓度的醌茜素培养液预处理,空白对照组为0μmol/L浓度的醌茜素培养液组。使用MTT法检测肺腺癌细胞A549细胞的增长抑制情况,使用Transwell检测肺腺癌细胞A549侵袭能力,并染色观察肺癌A549细胞数目变化;划痕实验法检测检测肺腺癌细胞A549迁移能力;使用Western blot检测肺腺癌细胞株A549中MMP-2、MMP-9、VEGF表达水平。结果与空白对照比较,1μmol/L醌茜素培养液组的肺腺癌细胞株A549细胞生长受到一定程度的抑制,但是抑制并不显著(P>0.05);3μmol/L醌茜素培养液组的A549细胞生长受到抑制且较为显著(P<0.05);10、30μmol/L醌茜素培养液组的细胞生长抑制十分显著(P<0.01)。与空白对照比较,1、3μmol/L的醌茜素培养液组A549细胞侵袭能力受到了抑制,且较为显著(P<0.05);10、30μmol/L醌茜素培养液组A549细胞侵袭能力受到了十分显著抑制(P<0.01)。与空白对照相比较,1μmol/L醌茜素培养液组的A549细胞迁移能力受到抑制不显著(P>0.05),3μmol/L组的受到抑制较显著(P<0.05),10、30μmol/L组则受到抑制十分显著(P<0.01)。与空白对照相比较,1μmol/L组MMP-9、VEGF蛋白表达水平受到抑制(P<0.05),MMP-2表达变化不显著(P>0.05),3、10、30μmol/L组,MMP-2、MMP-9、VEGF蛋白表达均受到显著抑制(P<0.01)。结论醌茜素可以通过下调MMP-2、MMP-9、VEGF蛋白表达水平,达到抑制肺腺癌细胞株A549迁移和侵袭能力的效果。
Objective To investigate the effect of quinalizarin on migration and invasion of lung adenocarcinoma cell line A549.Methods Human lung cancer A549 cells were cultured in vitro and pretreated with quinalizarin culture solution at concentrations of 0,1,3,10 and 30μmol/L respectively.The blank control group was treated with quinalizarin culture solution at a concentration of 0μmol/L.The growth inhibition of lung adenocarcinoma A549 cells was detected by MTT assay.The invasion ability of lung adenocarcinoma A549 cells was detected by Transwell,and the quantity of lung cancer A549 cells was observed by staining.The migration ability of lung adenocarcinoma A549 cells was detected by scratch assay.The expression levels of MMP-2,MMP-9 and VEGF in lung adenocarcinoma cell line A549 were detected by Western blot.Results Compared with blank control group,the growth of lung adenocarcinoma cell line A549 in 1μmol/L quinalizarin culture solution group was inhibited to some extent,but the inhibition was not significant(P>0.05).The growth of A549 cells in 3μmol/L quinalizarin culture solution group was inhibited significantly(P<0.05).The cell growth inhibition in 10 and 30μmol/L quinalizarin culture solution groups was completely significant(P<0.01).Compared with blank control group,the invasion ability of A549 cells in 1 and 3μmol/L quinalizarin culture solution groups was inhibited significantly(P<0.05).The invasion ability of A549 cells in 10 and 30μmol/L quinalizarin culture solution groups was completely significantly inhibited(P<0.01).Compared with blank control group,the migration ability of A549 cells in 1μmol/L quinalizarin culture solution group was not significantly inhibited(P>0.05),and the inhibition was significant in3μmol/L group(P<0.05),and the inhibition was completely significant in 10 and 30μmol/L groups(P<0.01).Compared with blank control group,the protein expression levels of MMP-9 and VEGF in 1μmol/L group were inhibited(P<0.05),and the expression level of MMP-2 was not significantly changed(P>0.05),and the protein expression levels of MMP-2,MMP-9 and VEGF were significantly inhibited in3,10 and 30μmol/L groups(P<0.01).Conclusion Quinalizarin can inhibit the migration and invasion of lung adenocarcinoma cell line A549 by down-regulating the protein expression levels of MMP-2,MMP-9 and VEGF.
