巴西绿蜂胶原胶、乙醇提取液及蜂胶渣中挥发性成分的分析与比较

摘要 [目的]采用GC-MS法分别对巴西绿蜂胶原胶、乙醇提取液、蜂胶渣石油醚微波辅助萃取法所得挥发性成分进行分析比较,为蜂胶的综合利用提供参考。[方法]用GC-MS联用仪对巴西绿蜂胶原胶、乙醇提取液、蜂胶渣石油醚微波辅助萃取法所得挥发性物质进行分析鉴定。色谱柱:HP-5MS毛细管柱(30m×I.D.0.25mm×Film0.25μm);柱温:初始80℃,保持5min,以5℃.min-1速度升温至185℃,保持5min,Post:270℃,2min,分流进样,分流比20∶1;进样口温度:250℃。质谱条件:EI电离源,电子能量70eV,接口温度:280℃,离子源温度:230℃,扫描范围:30~500amu。[结果]GC-MS分析鉴定出蜂胶原胶挥发性成分中主要活性物质9种,主要是芳香酸及萜类衍生物;蜂胶乙醇提取液及蜂胶渣挥发性成分中主要活性物质分别为13种和5种,主要为有机酸酯类。[结论]蜂胶原胶、蜂胶乙醇提取液及蜂胶渣中的挥发性成分在含量及化学组成上存在差异,但活性组分间存在转化关系。 [Objective] To investigate the diversity of content and chemical composition of volatile components from raw material,ethanol extract(PEES)and dregs after ethanol extracting(DPEES)of Brazilian green propolis. [Methods] Microwave-assistant extraction technology was used to obtain volatile components from propolis and the chemical components were separated and identified by gas chromatography - mass spectrometry(GC-MS). The relative content of each component was determined by area normalization method. Chromatographic conditions were: capillary column HP-5MS(30m×0.25mm×0.25μm); column temperature was 80℃ which was held for 5 min,followed by an increase in temperature to 185℃ at 5℃.min -1 and this was held isothermally for 5 min; split injection,split ratio 20:1; injector temperature 250℃.Mass spectrometer conditions were: ionization mode,EI; electron energy,70 eV; interface temperature,280 ℃; ion source temperature,230 ℃; mass scan range,30~500 amu. [Results]9 active ingredients were identi-ed from Brazilian propolis volatile components and the the major components were aromatic compound and terpenoids,13 and 5 active ingredients were identi-ed from PEES and DPEES volatile components,the major components were organic acid esters. [Conclusion]The volatile constituents of Brazilian propolis,PEES and DPEES were different in content and chemical composition,while some active ingredients can transformed between each other.
