SPAG11基因在雄性牦牛生殖器官中表达的检测 被引量:4

Detection of expression of SPAG11 gene in the reproductive organs of male Bos grunniens

摘要 为了检测精子相关抗原11(sperm-associated antigen 11,SPAG11)基因在雄性牦牛生殖器官中的表达谱并分析其组织表达特性,从牦牛附睾头总RNA中RT-PCR扩增SPAG11基因(SPAG11C、D、E、U、V和W),半定量RT-PCR分析SPAG11mRNA在牦牛生殖器官中的表达水平。采用合成的SPAG11E多肽制备牦牛SPAG11E多克隆抗体,免疫组织化学检测SPAG11E蛋白在睾丸和附睾中的表达及定位情况。结果显示,SPAG11C、U、V和W基因在睾丸和附睾相对低表达,而SPAG11D和E基因相对高表达;SPAG11C和U基因在输精管相对低表达,而SPAG11E相对高表达;除SPAG11D在卵巢相对低表达外,SPAG11基因在其他雌性生殖器官均不表达。免疫组织化学结果显示,SPAG11E蛋白在睾丸的精子细胞和附睾内壁精子头部表达。结果表明,SPAG11基因的表达具有组织特异性,提示它在牦牛睾丸和附睾中可能发挥重要的免疫功能和生殖功能。 The aim of this study was to investigate expression profile of sperm-associated antigen 11(SPAG11)gene in the reproductive organs of male yak(Bos grunniens)and analyze the characteristics of tissues expression.SPAG11genes(SPAG11C,D,E,U,V and W)sequences were amplified by RT-PCR from caput epididymidis of yak.Semi-quantitative RT-PCR was used to gain expression profiles of yak SPAG11gene in male reproductive organs,and immunohistochemical staining was used to detect SPAG11E protein in the testis and epididymis.The results showed that SPAG11C,U,V and W were lower abundance in the testis and epididymis,but SPAG11Dand SPAG11Ewere readily observed in the testis and epididymis.SPAG11Cand SPAG11U were marginally detected in the spermaduct but SPAG11E was abundant. The expression of SPAG11Dwas relatively low in the ovary,however SPAG11mRNA was not detected in other tissues.The results of immunohistochemical staining indicated that SPAG11Eprotein was expressed in the testicular sperm cells and the inner wall of the epididymis sperm head.It turned out that SPAG11gene expression with tissue specificity,and it was likely that SPAG11had a specific role in the male reproductive tract of yak.
