摘要
目的 通过测定糖尿病患者血小板一氧化氮合酶 (NOS)活性 ,探讨糖尿病患者血小板激活的机制。方法 晨取 16例 2型糖尿病患者外周静脉血 ,利用凝胶色谱柱法收集血小板 ,同位素二步色谱法测定血小板NOS活性。 8例健康成人作为对照。结果 糖尿病患者血小板NOS活性明显低于正常人 (P <0 0 1) ,组胺刺激后仍低于正常人 (P <0 0 5 )。
Objective To probe the mechanism of platelet activation by measuring the activity of platelet nitric oxide synthase(NOS) in patients with diabetes mellitus.Methods Peripheral venous blood was collected from 16 cases of diabetes.The platelets were isolated with gel-filtration chromatography.The activity of NOS was measured as formation of 3?H?L-citrulline from 3?H?L-arginine.By contrast,eight normal subjects were included in.Results The activity of NOS decreased in diabetic patients compared with normal subjects (P<0.01),it was still lower after histamine stimulation (P<0.05).Conclusion The decrease of the activity of platelet NOS in diabetic patients facilitates platelet activation.
