大孔吸附树脂分离纯化蒲公英中总黄酮的工艺研究 被引量:6

摘要 目的优化大孔吸附树脂分离纯化蒲公英中总黄酮的工艺条件及参数。方法以总黄酮含量为考察指标,采用静态吸附-解吸试验筛选最适树脂型号,通过动态吸附-解吸条件全面优化分离纯化工艺参数。结果 AB-8型树脂对蒲公英中总黄酮的吸附与解吸性能较好,最佳吸附条件为上柱液浓度以总黄酮含量计为9.2 g/L,pH为6,吸附流速2 BV/h,在树脂柱的径高比为1∶6,上样液体积为9 BV的条件下,吸附效果最佳;最佳解吸条件为4.5 BV 75%乙醇洗脱,洗脱流速为2 BV/h。结论 AB-8型大孔吸附树脂可用于蒲公英中总黄酮的精制纯化。 Objective To optimize the technological condition and the parameters of the separation and the purification of total flavone in taraxacum by macroporous adsorption resin. Methods With the total flavone amount as the investigation index,the static adsorption -desorption test was adopted to screen the optimal resin type and the technological parameters of the separation and the purification were comprehensively optimized by the dynamic adsorption and desorption conditions. Results Type AB - 8 resin was screened out due to its better adsorption and desorption properties. The optimal adsorption conditions were as follows:the upper column solution concentra-tion was 9. 2 g / L with the total flavone amount as the metering,pH = 6,the adsorption flow rate was 2 BV / h,under the conditions of the diameter height ratio of 1 : 6 in the resin column and the upper sample solution volume of 9BV,the adsorption effects were opti-mal;the optimal desorption condition was 4. 5 BV 75% ethanol elution with a elution flow rate of 22 BV / h. Conclusion Type AB - 8 macroporous adsorption resin can be used for the refining and the purification of total flavone in taraxacum.
