Study on Virus-Free Culture in Salvia Miltiorrhiza Bge.

摘要 为了有效降低丹参的病毒病危害,采取热处理和茎尖顶端分生组织培养相结合的方法,通过设计L4(23)正交实验,进行脱病毒技术的研究,并通过一系列生长数据的对比,如增殖系数、株高、根长、叶片面积等,探讨丹参脱病毒技术的相关处理条件和方法。并针对实验中出现的试管苗玻璃化现象进行遏制研究。结果表明:45℃条件下处理10 min,切取0.4 mm的茎尖顶端分生组织进行培养,脱病毒效果较好,脱病毒苗的生长优势明显,根系发达,存活率高,预示有较好的田间性状;在繁殖培养基MS+BA 0.2 mg/L+IAA 0.1 mg/L中加入0.1%活性炭,可有效遏制丹参玻璃化现象。 In order to effectively reduce the virus disease of Salvia miltiorrhiza Bge,the method of virus-free culture of meristem combining heat treatment was adopted. The orthogonal experiment was designed by comparing the growth rate,plant height,length of roots and blade area to establish the method and technique of eliminating the virus in Salvia miltiorrhiza Bge as well as eliminating the vitrification phenomenon of test-tube plantlet in the experiment. The results indicated that treatment 3 was the better one to eliminate the virus when the shoot tip was pretreated in the 45 ℃ water bath for 10 min and cut into 0. 4 mm size of stem apex. Under such circumstances,the test-tube plantlets grew faster with strong roots and developed blade and higher survival rate. These data mean that these plantlets would have better field characters in the late growth stage in the fields. Adding 0. 1% activated carbon into the reproductive culture medium( MS + BA 0. 2 mg / L + IAA 0. 1 mg / L) could effectively eliminate the vitrification phenomenon.
