摘要
目的 建立超临界流体萃取 (SFE)和高效液相色谱 (HPLC)的联用技术测定总合草苔虫中抗癌活性成分的方法。方法 正交设计法考察了影响草苔虫内酯 4 (bryostatin 4 )超临界流体萃取效率的主次效应及最佳萃取条件 ,并以HPLC定量检测bryostatin 4。结果 SFE萃取的最佳条件为 :压力 4 0MPa ,温度 4 5℃ ,改性剂量 1mL ,萃取体积 15mL。结论 SFE HPLC可用于检测总合草苔虫中的草苔虫内酯成分 ,该方法快速。
OBJECTIVE: To determine the bryostatin 4 in Bugula neritina on the basis of SFE and HPLC. METHODS: Orthogonal design and analysis of variance were used to optimize the four variables (temperature, pressure, volume of modifier and extraction volume) respectively. The bryostatin 4 in SFE extracts was determined by HPLC. RESULTS: The optimized conditions were as follows: pressure 40 MPa, temperature 45°C, 1 mL chloroform as modifier and extraction volume of 15 mL. The recovery of bryostatin 4 was 95.4% with RSD 0.64%. CONCLUSION: Our work provided a new way to detect the antinoeplastic components in Bugula neritina. SFE is rapid, convenient and less solvent consuming compared with solvent extraction.
