摘要
马乳酒样乳杆菌ZW3是从西藏开菲尔粒中分离得到的一株具有多种益生功能的乳酸菌。文章主要研究了Lactobacillus ke?ranofaciens ZW3(ZW3)对小鼠行为学和体内生物标记物的作用效果。利用行为学模型强迫游泳实验来评估ZW3抗抑郁样行为的能力;利用ELISA方法检测小鼠下丘脑和血清中的生物标记物来观察ZW3对小鼠体内化学物质的作用效果。研究结果显示,补充益生菌ZW3 4周后可以显著降低小鼠在强迫游泳中的不动时间(P<0.05);长期口服益生菌ZW3,显著增加小鼠下丘脑中5-羟色胺的水平,提高了44.43%,降低下丘脑中5-羟吲哚乙酸与5-羟色胺的比值和血清中犬尿氨酸与色氨酸的比值,纠正色氨酸代谢途径;血清中皮质酮含量降低了39.88%,恢复HPA轴的过度激活。结果表明,ZW3能明显改善小鼠的抑郁样行为。为马乳酒样乳杆菌ZW3应用在功能食品的研究与开发中提供理论依据。
Lactobacillus kefiranofaciens ZW3 is a strain with multiple probiotic functions isolated from Tibet Kefir grains.In this paper,Lactobacillus kefiranofaciens ZW3 was screened by the behavioral despair model,which had the potential to alleviate depression-like behavior.The behavioral model forced swimming experiment was used to evaluate the capacity of ZW3 antidepressant behavior.To observe the effect of ZW3 on chemical substances in mice,the biomarkers in the hypothalamus and serum were detected by ELISA.The results showed that after four weeks of ZW3 supplementation,the immobile time of mice in forced swimming test was significantly reduced(P<0.05).Administration of probiotic ZW3 significantly increased the level of serotonin in the hypothalamus of stressed mice,by 44.43%,decreased the ratio of 5-hydroxyindoleacetic acid to 5-hydroxytryptamine in the hypothalamus and the ratio of kynurenine to tryptophan in the serum,and corrected the metabolic pathway of tryptophan.Serum corticosterone levels were reduced by 39.88%,restoring overactivation of the HPA axis.The results showed that ZW3 could significantly improve depression-like behavior in mice.It provides a theoretical basis for the research and development of Lactobacillus kefiranofaciens ZW3 in functional food.
