摘要
目的探讨高效液相色谱法(HPLC)测定延胡索中有效成分延胡索乙素、延胡索丙素、小檗碱含量,分析传统炮制方式(醋制)对延胡索成分稳定性的影响。方法采用HPLC法,色谱柱Agilent TC-C18(250 mm×4.6 mm,5.0μm),流动相:乙腈:0.1%醋酸水溶液(三乙胺溶液调节p H为6.4)梯度洗脱,检测波长:282 nm,流速:1.0 ml/min。以延胡索乙素含量为指标,采用均匀设计法,以温度、醋量为因素,对延胡索的传统炮制进行研究。结果 1延胡索丙素在0.05~0.50μg/ml、延胡索乙素在0.20~2.00μg/ml、小檗碱在0.02~0.20μg/ml浓度范围内线性关系良好,平均加样回收率分别为96.54%、98.76%、99.48%;2延胡索生品经药典规定步骤处理后,生物碱含量降低一半左右,不同炮制过程对原药材中生物碱含量影响较少;3在炮制温度130℃,用醋量20%~50%条件下,醋制延胡索炮制品水煎液延胡索乙素的含量最高,稳定性较好。结论 HPLC测定延胡索中三种成分的含量,方法操作简单,重复性好,为延胡索质量标准的制定提供了科学依据,同时不同炮制品对延胡索中生物碱的含量影响较小,传统的炮制(醋制)有可能提高延胡索中延胡索乙素的稳定性。
Objective To investigate high Performance Liquid Chromatography( HPLC) in the detection of the content of Tetrahydropalmatine,protopine,berberine,and analyze the effect of the traditional processing method( vinegar) for the stability of corydalis constituents. Methods HPLC was taken,column Agilent TC-C18( 250 mm × 4. 6 mm,5. 0 μm),the mobile phase was composed of acetonitrile and 0. 1% aqueous acetic acid( use triethylaime solutions to adjust p H to 6. 4) with gradient elution and the detection wavelength is 282 nm with the flow rate of 1. 0 ml / min. The content of tetrahydropalmatine in decoction was as the index,uniform design method was taken. The temperature and the amount of vinegar were as the factors to study the corydalis traditional processing. Results 1 The linear relationship of propionate in 0. 05 ~ 0. 50μg /ml,tetrahydropalmatine at 0. 20 ~ 2. 00 μg /ml,berberine at 0. 02 ~ 0. 20 μg /ml concentration range was good,the average recovery rates were 96. 54%,98. 76% and 99. 48%. 2 The corydalis crude products processed by pharmacopoeia,the alkaloid content reduced by about half. The effect of different processing procedure for the alkaloids of radix was less. 3 In the conditions of processing temperature of 130 ℃,with vinegar amount 20 to 50%,the content of tetrahydropalmatine in vinegar Rhizoma corydalis processed products decoction was highest,and the stability was good. Conclusion The HPLC in the determination of three components of Rhizoma corydalis,the method is simple,reproducible,and provide a scientific basis for determination of the quality standards of Rhizoma corydalis. The effect of different processed products in the detection of alkaloids of radix in Rhizoma corydalis is small,and the traditional processing( with vinegar Ltd.) is possible to improve the stability of Tetrahydropalmatine.
