摘要
为探讨异甘草素对人膀胱癌T24细胞糖酵解水平的影响及其作用机制。本研究采用SRB法测定异甘草素对人膀胱癌T24细胞增殖的影响;试剂盒测定T24细胞中ATP含量变化;试剂盒检测细胞培养液中葡萄糖摄取和乳酸含量影响;用比色法测定己糖激酶和丙酮酸激酶活性,酶标仪法测定乳酸脱氢酶的活性。结果显示:异甘草素对T24细胞有显著增殖抑制作用,且呈剂量依赖性(P<0.01),经异甘草素处理的T24细胞出现明显的形态改变;异甘草素可浓度依赖性的抑制T24细胞ATP的生成(P<0.05或P<0.01);异甘草素可减少T24细胞对葡萄糖的摄取和乳酸的释放(P<0.05或P<0.01);与对照组相比,经过异甘草素处理的T24细胞的HK、PK和LDH活性均明显降低(P<0.05或P<0.01)。由此可知,异甘草素可抑制T24细胞糖酵解水平,其机制可能与减少葡萄糖摄取,下调糖酵解的关键性酶的活性有关。
To evaluate the effects of isoliquiritigenin on levels of ATP and lactic acid of T24 cells and its involved mechanism.The cell viability was assayed by SRB method. The content of ATP of T24 cells was detected by ATP assay kit, and the level of glucose uptake and lactic acid in T24 cells were measured by glucose assay kit and lactic acid assay kit respectively. The activities of hexokinase(HK), pyruvate kinase(PK) and actic dehydrogenase(LDH) were detected by colorimetry and enzyme- linked immunosorbent assay(ELISA). Isoliquiritigenin inhibited the proliferation of T24 cells in a concentration- dependent manner(P<0.01)with obvious morphological changes. Isoliquiritigenin inhibited the ATP production(P<0.05 or P<0.01) and decreased the glucose uptake(P<0.05 or P<0.01) and the release of lactic acid(P<0.05 or P<0.01). The activities of LDH,HK,PK were significantly down- regulated(P<0.05 or P<0.01). These data demonstrates that isoliquiritigenin can block the glycolysis in T24 cells may through reduction of the glycolytic enzymes' activities.
