黄芪糖蛋白提取方法的比较及抗氧化性研究 被引量:8

摘要 目的:建立一种黄芪糖蛋白提取方法,并测定其抗氧化活性。方法:采用4种提取方法(Tris-Hcl缓冲液,盐析法,碱提酸沉法,醇提法,结合sevage法除去多糖和游离蛋白)提取糖蛋白,测定其蛋白质和糖的含量,采用BioSpectrum凝胶成像系统对SDS-PAGE胶图进行分析,优选出黄芪糖蛋白最佳提取方法,通过测定其还原能力、清除超氧阴离子、羟自由基及DPPH自由基的能力,检测其抗氧化活性。结果:Tris-HCl缓冲液提取法最佳,蛋白质和糖的含量分别为6.8mg/mL和21.88 mmol/L,蛋白条带多,且条带清晰。黄芪糖蛋白具有一定的抗氧化活性,其中清除超氧阴离子的清除率为90%,接近维生素C的清除率。结论:Tris-Hcl缓冲液提取法更适用于黄芪糖蛋白的提取,其具有较好的抗氧化活性,为开发黄芪糖蛋白的新活性成分提供实验依据。 Objective:To establish a method for the extraction of astragalus glycoprotein and determine its antioxidant activity. Method:The polysaccharides were extracted by four extraction methods(Tris-HCl buffer,salting out,alkali extraction,alcohol extraction,sevage removal of polysaccharides and free protein). The contents of protein and sugar of glycoprotein were determined. SDS-PAGE was analyzed by BioSpectrum gel imaging system. The optimum extraction method of astragalus glycoprotein was selected. The antioxidant activity of Astragalus glycoprotein was evaluated by its reducing power,scavenging effect of superoxide radical,polysaccharides on hydroxyl radical and DPPH free radical. Results:The method of Tris-HCl buffer extraction was the best,the contents of protein and sugar were 6.8 mg/mL and 21.88 mmol/L respectively,and the bands of protein were clear. Astragaloside glycoprotein had a certain antioxidant activity. The clearance rate of superoxide radical was 90%. The effect was close to that of vitamin C. Conclusion:The method of Tris-HCl buffer extraction was more suitable for the extraction of astragalus glycoprotein. Astragalosin has antioxidant activity,which provides experimental basis for the development of new active components of astragalus protein.
