RNA干扰分化抑制因子1基因表达对结肠癌细胞增殖的影响被引量：1

摘要目的观察RNA干扰分化抑制因子1(Id1)基因后对结肠癌细胞增殖的影响。方法将结肠癌HT-29细胞分为3组,空白组仅加培养血清,空载体组转染无义序列,抑表达组转染Id1抑表达质粒。采用RT-PCR和Western blot法检测HT-29细胞低氧诱导因子1α(HIF-1α)、血管内皮生长因子(VEGF)mRNA及其蛋白表达。各组细胞培养8 d,采集细胞培养上清液,并用于培养人脐静脉内皮细胞(HUVECs),MTT法检测HT-29细胞及HUVECs的增殖情况。结果与空白组、空载体组比较,抑表达组VEGF、HIF-1αmRNA及蛋白表达表达均下降(P均<0.05),HT-29细胞及HUMECs分别于培养第2、3天开始细胞增殖活性下降(P均<0.05)。结论 Id1通过对HIF-1α、VEGF的负性调控抑制结肠癌HT-29细胞体外增殖及HUVECs血管生成能力。
