摘要
目的探讨长链非编码RNA NEAT1(LncRNA NEAT1)在胃癌组织中的表达水平与胃癌患者临床病理参数和预后的关系。方法收集2015年3月至2017年2月上海交通大学附属第九人民医院手术切除的胃癌组织标本为胃癌组(n=125),以癌旁组织标本为正常组(n=125)。采用实时荧光定量PCR(qRT-PCR)检测LncRNA NEAT1在胃癌及癌旁组织中的表达水平,根据检测结果将胃癌患者分为LncRNA NEAT1高表达组(n=86)与低表达组(n=39),观察其与患者临床病理参数的关系。采用Kaplan-Meier分析LncRNA NEAT1表达与胃癌患者预后的关系,COX回归模型分析胃癌患者预后的影响因素。结果与正常组比较,胃癌组LncRNA NEAT1表达水平显著升高(P<0.05);LncRNA NEAT1表达与淋巴结转移、临床分期、分化程度及浸润深度显著相关(P<0.05),而与患者年龄、肿瘤位置及肿瘤直径无关(P>0.05);Kaplan-Meier分析显示LncRNA NEAT1高表达组患者无进展生存期、总体生存时间均显著低于低表达组(P<0.05);COX分析显示LncRNA NEAT1表达水平为胃癌患者预后的独立危险因素。结论 LncRNA NEAT1在胃癌组织中呈高表达并与患者临床病理参数密切相关,同时可作为预测胃癌患者预后的重要指标。
Objective To investigate the expression of long non-coding RNA NEAT1(LncRNA NEAT1)in gastric cancer tissues and its relationships with clinicopathological parameters and prognosis.Methods From March 2015 to February2017,gastric cancer specimens surgical resected in our hospital were collected and as the gastric cancer group(125 cases)and para cancerous tissue specimens were taken as normal group(125 cases).The expression level of LncRNA NEAT1 in gastric cancer tissues and para cancerous tissues was detected by real-time fluorescence quantitative PCR(qRT-PCR).According to the results,gastric cancer patients were divided into LncRNA NEAT1 high expression group(86 cases)and low expression group(39 cases);its relationship with clinical pathological parameters was observed.Kaplan-Meier was used to analyze the relationship between LncRNA NEAT1 expression and prognosis in patients with gastric cancer.The prognostic factors of gastric cancer were analyzed by univariate and multivariate COX regression models.Results Compared with the normal group,the expression level of LncRNA NEAT1 in gastric cancer group was significantly higher(P<0.05);LncRNA NEAT1 expression was significantly correlated with lymph node metastasis,clinical stage,degree of differentiation and depth of invasion(P<0.05),but not with age,tumor location and tumor diameter(P>0.05).KaplanMeier analysis showed that PFS and OS in the LncRNA NEAT1 high expression group were significantly lower than those in the low expression group(P<0.05);COX analysis showed that the expression level of LncRNA NEAT1 was an independent risk factor for the prognosis of gastric cancer.Conclusion LncRNA NEAT1 was highly expressed in gastric cancer tissues and was closely related to clinicopathological parameters;it could also be used as an important prognostic indicator in patients with gastric cancer.
