摘要
目的:观察清热活血方对β_2-糖蛋白(β_2-GPI)诱导的抗心磷脂抗体(aCL)产生的影响,探讨清热活血方对抗磷脂抗体综合征(APS)的治疗作用及可能的作用机制。方法:4周龄的30只雌性昆明种小鼠被随机分为3组:A组(用药组)、B组(模型组)、C组(正常对照组);A组用清热活血方5g/(kg·d)灌胃;14d后,A、B两组多部位皮下注射β_2-GPI;检查血清aCL、外周血T细胞亚群和IL-2生物活性。结果:A组各周血清aCL OD值均较B组有不同程度降低(P<0.01)。与B组相比,A组可明显降低T_H/T_s比值(P<0.01),可使不同稀释度的IL-2活性明显降低(P<0.01)。经双变量直线分析,T_H/T_S比值和IL-2生物活性呈正相关(P<0.01)。结论:清热活血方可抑制β_2-GPI诱导的aCL的产生,其作用机理可能是抑制T_H/T_S比值和IL-2生物活性的升高。
Objective: To explore the therapeutical effect and the mechanism of Chinese herbs on APS by observing the effect of Qingre Huoxue Fang (QRHXF) on aCL induced by β2-GPI- Methods: 30 female mice of 4 weeks were randomly divided into 3 groups, 10 in each group. Group A was administered with QRHXF in dosage of 5g/(kg·d); after 14 days, group A and B was injected with 150 μg of purified human β2-GPI in CFA subcutaneously. The titre of acL were detected by ELISA; Subsets of T cell were grouped by SABC; and the activity of IL-2 was measured by MTT chomatometry. Results: Compared with group B, the optical density of aCL OD in group A was decreased (P<0.01); Compared with group B, TH/TS ratio was reduced obviously in group A (P<0.01); Compared with group B, the activity of IL-2 in group A decreased significantly (P<0.01): There is positive correlation betweenTH/TS and IL-2 activity in different dilutions (P<0.01). Conclusions: The mechanism of suppressive effect of QRHXF on aCL induced by β2-GPl may be realized by resuming the elevated TH/TS and IL-2. The state that QRHXF has no effect on normal mice indicates that QRHXF has immune-regulation function.
