摘要 TGF-β信号通路控制着细胞的生长速度、分化方向、细胞外基质形成、胚胎发育及肿瘤发生等各个方面,但对于TGF-β信号通路的调控研究一直进展缓慢。1999年,3个研究小组几乎同时发现,Ski/SnoN蛋白通过募集转录共抑制子N-CoR蛋白而对TGF-β通路起抑制作用;之后又发现,TGF-β及Smad蛋白对Ski/SnoN通过介导泛素化连接而降解,具有负反馈调节作用。这一重大发现对于阐明TGF-β信号通路的功能提供了理论基础。 TGF-β signal pathway plays an important role in the cell growth, differentiation, formation of extracellular matrix, embryo development and carcinogenesis, etc. However, the regulation of TGF-β pathway is not totally understood. In 1999, three independent research groups found that Ski/SnoN protein could inhibit the TGF-β mediated transcription by recruiting N-CoR, a transcription co-repressor. Later studies suggested that TGF-β and SMADs degraded the Ski/SnoN protein by mediating ubiquitin linkage, showing negative feedback regulation. The important findings in Ski/SnoN laid the theoretical foundation for demonstrating the function of TGF-β signal pathway.
