摘要
目的 :研究单核细胞趋化蛋白 1(monocytechemoattractantprotein 1,MCP 1)在糖尿病大鼠肾脏的表达。方法 :Wistar大鼠随机分 3组 :正常对照组 (NC组 ) ,糖尿病未干预组 (DM组 ) ,糖尿病大鼠胰岛素干预组 (DI组 )。以链脲佐菌素 (streptozotocin ,STZ)制备糖尿病大鼠模型 ,大鼠饲养 2 ,4周分别取出肾脏检测MCP 1mRNA的表达。收集2 4h尿测定尿白蛋白排泄率 (urinaryalbuminexcretionrate ,UAER)及肌酐 ,并心脏采血检测血肌酐、尿素氮。mRNA的表达采用RT PCR ,以GAPDH作内对照。结果 :2 ,4周肾组织中MCP 1mRNA的表达DM组高于NC组 (P <0 .0 1) ,DI组低于DM组 (P <0 .0 5 )。 2周时DM组、DI组UAER较NC组有升高趋势 ,但无统计学意义 (P >0 .0 5 ) ;4周时DM组、DI组UAER较NC组显著增加 (P <0 .0 1) ,DI组UAER较DM组显著降低 (P <0 .0 1) ,其变化趋势与MCP 1mRNA的表达基本相似。结论 :在糖尿病大鼠肾组织MCP 1mRNA表达明显增加 ;经胰岛素治疗后肾组织MCP 1mRNA的表达降低 。
Objective: To investigate the Expression of Monocyte Chemoattractant Protein 1 mRNA in the kidney of diabetic rats. Methods: The rats were randomly divided into three groups: normal control group(NC group);diabetic model group,without any therapy(DM group);treated insulin(DI group). Diabetic models were made by the intraperitoneal injection of streptozotocin. At the end of 2th week and 4th week,kidneys were taken out from all the rats to measure the expression of MCP 1 mRNA by RT PCR with the GAPDH as the internal control. At the same time,blood was drawn from the heart to measure the level of creatinine,Urea nitrogen,and 24 hour urine was collected to measure the level of creatinine and albumin(urine albumin excretion rate, UAER). Results: At the end of 2th and 4th week, the expression of MCP 1 mRNA in group DM was significantly higher than that in group NC(P<0.01),and the expression of MCP 1 mRNA in group DI was significantly lower than that in group DM(P<0.05). MCP 1 mRNA expression had positive correlation with UAER. At the end of 2th week, UAE in group DM and group DI had tendency to increase, but showed no significant difference between group DM and group NC, or group DI and group NC. At the end of 4th week,UAE in group DM and group DI was significantly higher than that in group NC(P<0.01).And UAE in group DI was significantly lower than that in group DM(P<0.01). Conclusion: The expression of MCP 1 mRNA is increased significantly in kidneys of diabetic rats and has positive correlation with renal lesion. Insulin can decrease MCP 1 mRNA expression in kidneys of diabetic rats, and improve kidney function.
