摘要
目的探究长链非编码RNA PVT1对卵巢癌细胞自噬的影响及其调控机制。方法 Real-timePCR检测PVT1在卵巢癌组织样本中表达情况及PVT1在卵巢癌细胞株中的表达;免疫荧光实验和Western blot检测过表达PVT1对卵巢癌细胞自噬水平的影响;Western blot用于研究过表达PVT1对自噬相关蛋白ATG7表达的影响;利用共转染实验验证PVT1通过上调ATG7从而影响自噬水平。结果 Real-time PCR结果表明卵巢癌组织中PVT1的表达水平高于正常组织,各卵巢癌细胞系(ES2、OVCAR3、A2780)中PVT1表达高于正常卵巢细胞系(HOSEpiC)。Western blot和免疫荧光实验结果表明,与对照组相比,过表达PVT1能增加自噬标志蛋白LC3II/LC3I比例,能够显著促进卵巢癌细胞的自噬并通过上调ATG7的表达促进细胞自噬的发生。结论 lncRNA PVT1能够上调ATG7表达,促进卵巢癌细胞自噬的发生,有望成为卵巢癌新的治疗靶点。
Objective To investigate the effect and mechanism of lncRNA PVT1 modulating autophagy in ovarian cancer. Methods Real-time PCR was used to detect the expression of PVT1 in ovarian cancer samples and different ovarian cancer cell lines. Immunofluorescence assay and Western blot were performed to detect the effect of PVT1 overexpression on autophagy in ovarian cancer. Western blot was used to investigate the expression of ATG7 and LC3 in ovarian cancer. Co-transfection assay was performed to demonstrate that PVT1 could promote autophagy via increasing ATG7. Results Compared with the normal ovarian tissues, the expression of PVT1 was significantly enhanced in ovarian cancer tissues. Compared with the normal ovarian cell lines(HOSEpiC), the expression level of PVT1 was significantly increased in ovarian cancer cell lines(ES2、OVCAR3、A2780). Compared with the control group, overexpressing PVT1 could increase the ratio of LC3 II to LC3 I and promote autophagy through increasing ATG7 in ovarian cancer. Conclusion PVT1 could promote autophagy through increasing ATG7 expression, which is expected to be a new therapeutic target for ovarian cancer.
